Summary A method for quantitative determination of bile acids was developed. This method is based upon specific reactions to detect free and conjugated dihydroxycholanic acids which cannot be separated by means of thin-layer chromatography. Free and conjugated deoxycholic acid may be determined by reaction with salicylaldehyde-sulfuric acid-acetic acid, free and conjugated chendeoxycholic acid with ethylacetate-sulfuric acid-acetic anhydride, both in presence of the corresponding isomeres after thin-layer separation. To determine cholic acid and its conjugates the reaction with 65% sulfuric acid is used.

Zusammenfassung Es wird ein Verfahren zur quantitativen Bestimmung von Gallensäuren nach Dünnschichtchromatographie beschrieben, welches auf der Anwendung spezifischer Nachweisreaktionen für die dünnschichtchromatographisch nicht oder nur ungenügend zu trennenden Dihydroxycholansäuren und ihrer Conjugate beruht. Freie und konjugierte Desoxycholsäure läßt sich mit Salicylaldehyd-Schwefelsäure-Eisessig, freie und konjugierte Chenodesoxycholsäure mit Äthylacetat-Schwefelsäure-Essigsäure-anhydrid nach Dünnschichtchromatographie jeweils in Gegenwart des entsprechenden Isomeren bestimmen. Die Messung der Cholsäure und ihrer Conjugate erfolgt nach Reaktion mit 65% Schwefelsäure.
