    Notizen: The influence of temperature on the embryonic development of three European cyprinid fishes, Abramis brama, Chalcalburnus chalcoides mento and Vimba vimba, was studied with respect to hatching efficiency and the functional relationship with the duration of development. The functional dependance of the duration of development on temperature is well described by a power function with a correcting constant for temperature. Comparison with published data for other cyprinids shows that the response of embryos to temperature, together with the various temperature controlled responses of adult fishes, is a valuable aid for the ecological characterization of the different species.

A method for determining free Ca2+-ions in the erythrocyte is described, using a commercially available ORION-Ca-electrode and calomel reference electrode assembly, where changes in free Ca2+-ion concentration upon addition of 0.01% digitonin could be measured.2. The average value found for fresh cells from 20 healthy donors at 37°C (pH = 7.4) was 0.20 ± 0.04 μmol/L referred to a haematocrit of 10%.3. Decrease of the simultaneously determined adenosinetriphosphate (ATP) concentration indicates that ATP is presumably needed to activate the Ca-ATPase.4. In vitro addition of the calcium antagonists pentoxifylline and nifidepine, respectively, induced a normalization of the intraerythrocytic Ca2+-ion concentration after previous increase with the ion carrier ionophore A23187.5. The advantages and possible clinical applications of this method are discussed.

    Notizen: Summary GAWK is a recently discovered peptide isolated from extracts of human pituitary gland and subsequently shown to be identical to sequence 420–493 of human chromogranin B. The distribution of this peptide was studied in human gut, pancreas, adrenal and pituitary glands using antisera to two portions of the 74 amino acid peptide (sequences 1–17 and 20–38). In addition, the co-existence of GAWK immunoreactivity with other peptides and chromogranin B was investigated using comparative immunocytochemistry. In the gut, GAWK was localised mainly to serotonin-containing cells of the mucosal epithelium, where electron microscopy showed it to be stored in typical electron-dense (250 nm diameter) granules, and to a moderate population of nerve fibres in the gut wall. Considerable quantities of GAWK-like immunoreactivity were measured in the gut, up to 36.3±18 pmol GAWK 1–17/g wet weight of tissue (mean±SEM) and 12.4±2.9 pmol GAWK 20–38/g. Chromatography of gut extracts revealed several GAWK-like immunoreactive peaks. GAWK-like immunoreactivity was also detected in endocrine cells of pancreas, pituitary gland and adrenal medulla, where the highest concentrations of GAWK-like immunoreactivity were measured (GAWK 1–17 2071.8±873.2 and GAWK 20–38 1292.7±542.7 pmol/g). Endocrine cells containing GAWK-like immunoreactivity were found also to be immunoreactive for chromogranin B. Our results define a discrete distribution of GAWK immunoreactivity in human endocrine cells and nerves and provide morphological support for the postulated precursor-product relationship between chromogranin B and GAWK. Details of the functions of this peptide are awaited.

    Notizen: Summary The co-localization of various antigens in rat chromaffin granules was investigated by the immunogold staining procedure. In ultrathin serial sections staining of chromaffin granules was obtained with antisera against chromogranin A, chromogranin B, secretogranin II and neuropeptide Y. These results indicated that these antigens are costored within chromaffin granules. To further corroborate this point a double immunogold staining procedure was used. This method unequivocally established that chromogranin A, chromogranin B, secretogranin II and neuropeptide Y are co-localized in the same chromaffin granules. These results are relevant for studies demonstrating changes in the level of these peptides in adrenal medulla. The co-localization makes it likely that such changes lead to a different relative composition of the secretory quanta of chromaffin granules.

    Notizen: Summary Antisera against chromogranin A, B and C were used to study the distribution of these acidic proteins in bovine endocrine and nervous tissues. The three chromogranins occur together in several endocrine organs (adrenal medulla, anterior pituitary, endocrine pancreas) and in sympathetic ganglion cells. In the posterior pituitary, only chromogranin C and in the intermediate lobe only A and C are found. The parathyroid gland contains only A, and enterochromaffin cells are immunoreactive for A and B. Cells of the thyroid gland and some cells of the anterior pituitary apparently do not contain any chromogranins. It is concluded that the three chromogranins are not always stored together and that they are not present in all endocrine cells. This distinct localization of the chromogranins indicates some special, although still undiscovered, function for these proteins.

    Notizen: Summary Antisera were raised against synthetic peptides derived from the primary amino acid sequence of human chromogranin B. These antisera recognized in one- and two-dimensional immunoblotting a component previously designated as chromogranin B. In human chromaffin granules, the major endogenous processing product of chromogranin B is formed by proteolytic cleavage of the protein near theC-terminus. Immunohistochemical localizations were obtained with antisera against human chromogranins A and B and against a synthetic peptide corresponding to the B sequence. In human tissues, chromogranin B is co-stored with chromogranin A in the adrenal medulla, the anterior pituitary, parafollicular cells of the thyroid, in some cells of the endocrine pancreas and in some enterochromaffin cells, whereas only chromogranin A is found in the parathyroid gland and enterochromaffin cells of the gastric corpus mucosa. In the nervous system, no immunostaining was observed for chromogranin A and only a weak one for chromogranin B in some cells of the spinal cord. However, the Purkinje cells of the cerebellum were strongly positive for chromogranin B.

    Notizen: Abstract Measurements of three high-spin FeIIS4 complexes at various temperatures and applied magnetic fields are reported and the results are compared with those of reduced rubredoxin. Simulations of the measured spectra by means of the fully developed stochastic theory of lineshape prove that intermediate relaxation behavior is present and is determined essentially by the phonon energy distribution and by one single parameter W0, the scaling factor, which contains the strength of spin-lattice coupling.

    Notizen: Abstract We have performed Mössbauer studies on several oxy-heme model compounds, and for two of them we also have derived the threedimensional structure from X-ray studies; I: [Fe(O2) (SC6HF4) TPpivP] [Na⊂18C6] and II: [Fe(O2) (SC6HF4)TPpivP] [K⊂222]. Compound I shows a pronounced temperature-dependence of ΔEQ, i. e. 2.17 mms−1 at 4.2 K and 1.55 mms−1 at 295 K, while ΔEQ of compound II is practically temperature-independent, i.e. ca. 2.05 mms−1. The X-ray structure analysis of these model compounds provides us with the fact that O2 occupies three different sites in I and only two sites in II. From the experimentally known occupancies of these sites at specific temperatures we were able to apply relaxation calculations for the electric field gradient tensors in I and II, and we found for both compounds that our calculated ΔEQ(T)-data are in agreement with our experimental results. These findings imply that the dynamic jump behavior of O2 among the different sites in oxy-heme model compounds is responsible for the temperature-dependence (like in HbO2) or temperature-independence of ΔEQ (like in cytochrome P450).

    Notizen: Abstract Transition metal ions are essential for the specific function of many biomolecules, like transport and activation of small molecules, electron transfer, etc. Their active sites contain (i) single paramagnetic or diamagnctic metal ions, or (ii) clusters of spin-coupled paramagnetic centers. Examples are for (i) hemoglobin or rubredoxin with one Fe, and for (ii) ferredoxins with [2Fe-2S] and [4Fe-4S] clusters, cytochrome c oxidasc containing Fe and Cu, or peroxidases with metal-radical pairs. Here, we present the results of our Mössbauer and EPR investigation of irondioxygen interaction in horse liver alcohol dehydrogenase (HLADH), of ironradical interaction in synthetic analogues of heme peroxidases and of iron-iron double exchange interaction in a synthetic ferric/ferrous mixed valence complex. We hope to demonstrate that the understanding of spin coupling can yield valuable insight in the relation between electronic structure and function of biological iron complexes.
