    Notes: We determine best-fitting Euler vectors, closure-fitting Euler vectors, and a new global model (NUVEL-1) describing the geologically current motion between 12 assumed-rigid plates by inverting plate motion data we have compiled, critically analysed, and tested for self-consistency. We treat Arabia, India and Australia, and North America and South America as distinct plates, but combine Nubia and Somalia into a single African plate because motion between them could not be reliably resolved. The 1122 data from 22 plate boundaries inverted to obtain NUVEL-1 consist of 277 spreading rates, 121 transform fault azimuths, and 724 earthquake slip vectors. We determined all rates over a uniform time interval of 3.0m.y., corresponding to the centre of the anomaly 2A sequence, by comparing synthetic magnetic anomalies with observed profiles. The model fits the data well. Unlike prior global plate motion models, which systematically misfit some spreading rates in the Indian Ocean by 8–12mm yr−1, the systematic misfits by NUVEL-1 nowhere exceed ∼3 mm yr−1. The model differs significantly from prior global plate motion models. For the 30 pairs of plates sharing a common boundary, 29 of 30 P071, and 25 of 30 RM2 Euler vectors lie outside the 99 per cent confidence limits of NUVEL-1. Differences are large in the Indian Ocean where NUVEL-1 plate motion data and plate geometry differ from those used in prior studies and in the Pacific Ocean where NUVEL-1 rates are systematically 5–20 mm yr−1 slower than those of prior models. The strikes of transform faults mapped with GLORIA and Seabeam along the Mid-Atlantic Ridge greatly improve the accuracy of estimates of the direction of plate motion. These data give Euler vectors differing significantly from those of prior studies, show that motion about the Azores triple junction is consistent with plate circuit closure, and better resolve motion between North America and South America. Motion of the Caribbean plate relative to North or South America is about 7 mm yr−1 slower than in prior global models. Trench slip vectors tend to be systematically misfit wherever convergence is oblique, and best-fitting poles determined only from trench slip vectors differ significantly from their corresponding closure-fitting Euler vectors. The direction of slip in trench earthquakes tends to be between the direction of plate motion and the normal to the trench strike. Part of this bias may be due to the neglect of lateral heterogeneities of seismic velocities caused by cold subducting slabs, but the larger part is likely caused by independent motion of fore-arc crust and lithosphere relative to the overriding plate.

    Notes: Stereotaxic surgery was performed on the trigeminal motor nuclei (TMNu) of 47 actively growing rats in order to test the effects of neuromuscular alterations on TMJ disc morphology. Lesioned animals received a small electrolytic lesion to their leftside TMNu, while shams had their TMNu stimulated without an actual lesion being produced. Both lesioned and sham-lesioned animals demonstrated significant (P〈0.05; P〈0.01) morphological alterations of their discs 28, 56, and 84 days postoperatively. Data indicated that (i) TMNu lesions do indeed affect disc morphology, and (ii) even subtle damage to trigeminal motoneurones can influence TMJ disc morphology. It is therefore suggested that at least some TMJ disease processes have a neuromuscular basis.

    Notes: Previous studies have shown that type I diabetes (IDDM) increases the risk of developing periodontitis by 2-3-fold. IDDM patients exhibit destruction of the pancreatic beta cells, most probably caused by an autoimmune reaction. Evidence is accumulating to support the role of the autoimmune response in periodontal pathogenesis. A cytokine, interleukin (IL)-10, has been reported to selectively promote the expansion of a B lymphocyte lineage (CD5/LY1/B1) which has the propensity for secreting high levels of autoantibody. Therefore, the purpose of this project was to evaluate IL-10 production, percentage of CDS B cells and the frequency of anti-collagen secreting cells in peripheral blood mononuclear cells of age, gender and race matched IDDM patients and controls. IL-10 production was evaluated by an ELISA using the supernatant of adherent peripheral blood cells cultured for 24 h in the presence of Porphyromonas gingivalis lipopolysaccharide (LPS). In 8 of 31 patients, IL-10 levels were significantly increased in IDDM compared to controls and a higher percentage of CD5 B cells was also observed by flow cytometry. In addition, these patients exhibited a higher frequency of anti-collagen secreting cells as elucidated by an ELISPOT. Moreover, treatment with a neutralizing anti-IL-10 antibody diminished the anti-collagen antibody response by 70%. These findings support the concept that a subset of IDDM patients possess an extremely robust IL-10 response following exposure to Gram-negative LPS, which could predispose them to the development of periodontitis through a heightened autoimmune mechanism.

    Notes: Abstract. The objective of this research was to determine the effectiveness of a biochemical assay which measures proteolytic enzyme activity in gingival crevicular fluid (GCF) and to relate this enzyme activity to clinical parameters traditionally utilized for periodontitis detection. A clinical trial was conducted on 8 periodontitis subjects with ≥4 sites exhibiting a loss of attachment of ≥5 mm and probing depths of ≥5 mm with bleeding on probing. On each subject, a plaque index was performed, followed by GCF sampling at those sites which exhibited a loss of attachment and probing depths. GCF was analyzed for activity against benzoyl-L-arginine-p-nitroanilide in the presence (BAPNA w/gly-gly) and the absence (BAPNA w/o gly-gly) of glycyl-glycine and against MeOSuc-Ala-Ala-Pro-Val-pNA and Suc-Ala-Ala-Pro-Phe-pNA for neutrophil serine proteinases activity (elastase and cathepstn G, respectively). Subsequently, a gingival index was performed, attachment levels and probing depths were recorded using a constant force probe with bleeding on probing being noted. A split-mouth design was employed and half mouths were randomly assigned to the following treatment groups: group A, half of the mouth received scaling/root planing and polishing: group B, half of the mouth received no treatment (control). Subjects were treated, then instructed on toothbrushing and interdental cleaning. After 4 weeks, subjects returned to receive a plaque index; GCF sampling, gingival index, attachment levels, probing depths and bleeding on probing as described above. Using a patred Student t-test, the findings suggest that BAPNA w/gly-gly was significantly less in treatment sites than in non-treated control sites (p=0.05). No such correlation was found for other activities, including neutrophil serine proteinases which were shown to occur in GCF in free, proteolytically active forms. In addition, significant treatment effects were detected for probing depths (p= 0.03) which reduced by 1.3 mm and attachment levels (p=0.02) which gained 0.7 mm. The reduction of P. gingivalis from treated periodontitis sites as detected by a significant decrease in BAPNA w/gly-gly may prove to be a valuable marker for periodontal disease activity.
