    Notes: Abstract To investigate the direct effects of methamphetamine (MAP) on cardiac lesions seen in MAP abusers, isolated adult rat ventricular cardiomyocytes (ARCs) were exposed to MAP (0.05–1.0 mM) in medium 199 containing 10% fetal calf serum. Isolated ARCs attached to laminin-coated substrata and began to spread into polygonal shapes with pseudopodia at day 6 in normal culture. However, the cell attachment and spreading were inhibited by exposure to MAP (0.5 and 1.0 mM) for the first 7 days in culture. On the other hand, exposure to MAP (0.05 and 0.1 mM) for 7 days after a 6-day period of normal culture, led to a larger cross surface area of cells with more abundant actin bundles compared to control cells (p 〈 0.05). This development of spreading area resembled that of norepinephrine-treated ARCs. In addition, immunoreactive atrial natriuretic peptide (ANP) granules developed and accumulated around the nuclear region of ARCs exposed to MAP and the number of ANP positive cells tended to increase in a dose-dependent manner. These results suggest that chronic exposure to a high concentration of MAP may directly inhibit development of ARCs in culture and that a continuous exposure to a low concentration of MAP may facilitate the development of cellular hypertrophy. Therefore, hypertrophied cardiomyocytes in MAP abusers may be provoked by multifactorial incidents of direct and indirect actions of MAP.

    Notes: Summary HPLC determination of pheophytinatonicke(II) (Pheo-Ni) prepared by the replacement of magnesium(II) in chlorophyll with nicke(II) is described. The good separation of PheoNi was obtained by using chemically bonded C18 as the stationary phase and acetone-methanol (50∶50, vol/vol) as the mobile phase. Conventional spectrophotometric method was also used for the determination of PheoNi. For the synthetic samples prepared by mixing (pheophytinato a) nicke(II) [(Pheo-a) Ni] and (pheophytinato b) nicke(II) [(Pheo-b) Ni], analytical values obtained by the spectrophotometric method were very high compared to those obtained by HPLC. In the proposed HPLC method, (Pheo-a) Ni and (Pheo-b). Ni could be determined in the concentration range of 0.028–30μg/ml and 0.038–30μg/ml with relative standard deviations (n=10) of 3.1% and 0.8%, respectively.

    Description / Table of Contents: Zusammenfassung Es wurde eine neue Methode zur Identifizierung von fetalem Hämoglobin (Hb F) in Blutspuren mit der umgekehrten Phase-Hochleistungsflüssigkeitschromatographie durchgeführt. Die Unterscheidung zwischen fetalen Blutspuren und denen von Erwachsenen erfolgte aufgrund der Existenz der Spitzen der γ-Ketten von Hb F. In den Chromatogrammen der untersuchten Blutspuren von Erwachsenen traten fast keine γ-Ketten auf. Diese Methode ermöglichte ferner die Bestimmung des Hb F-Wertes mit dem Flächenverhältnis der Globin-Ketten auf den Chromatogrammen. Hb F-Werte der sechs Fälle von Nabelschnurblutspuren auf dem Filterpapier lagen zwischen 81.1% und 91.3%. Die vorliegende Untersuchung zeigt, daß sich der Hb F-Wert für mindestens 12 Wochen nicht ändert. In der Praxis der Rechtsmedizin ist diese Methode im Hinblick auf die Einfachheit, hohe Empfindlichkeit, Schnelligkeit und Zuverlässigkeit sehr wertvoll.
    Notes: Summary A new method for the identification of fetal hemoglobin (Hb F) in blood stains by reverse-phase high performance liquid chromatography is described. Differentiation between fetal and adult blood stains is based on the existence of γ-chain peaks which are characteristic of Hb F. Very few γ chains appeared on chromatograms of all the adult blood stains examined. The level of Hb F could be determined by measuring the total of chromatogram γ-globin chain areas, and expressing it as a percentage of total Hb. Levels in six cord blood stains on filter paper ranged from 81.1% to 91.3% and remained constant for at least 12 weeks. This method is of great value for its simplicity, sensitivity and speed, and most importantly for its reliability in the field of forensic medicine.
