Notes: Background The germline (GL) ɛ promoter is regulated by IL-4 and is essential for class switching to IgE. IL-4-induced gene expression is largely mediated through activation of latent transcription factor STAT6 (signal transducer and activator of transcription 6).Objective We investigated whether increased levels of IgE in allergic individuals may be associated with alteration in the level or activation of STAT6 and subsequent increase in GL ɛ promoter activity. Methods Electrophoretic mobility shift assay and Western blotting assays were used to investigate the level of expression and activation of STAT6 in Epstein–Barr virus (EBV)-transformed B cell lines from children with birch pollen allergy and their non-allergic siblings. The activity of the GL ɛ promoter was tested in a transient transfection assay. Results STAT6 was expressed at the same level in all B cell lines tested. In two out of five sibling pairs STAT6 was activated by IL-4 more efficiently in the allergic individuals but in the three other pairs the opposite was found. In transient transfections, no difference in IL-4-induced GL ɛ promoter function was detected, although basal promoter activity varied between allergic and healthy siblings in two out of five pairs. Conclusions We demonstrate for the first time that upon IL-4 signalling STAT6 transcription factor activation differs in B cells from different individuals. Although we did not find any association between STAT6 activation and allergy, we do not exclude a possibility that stronger activation of this transcription factor is associated with an expression of allergic phenotype.

Abstract: Zusammenfassung Am Tavistock Institute wurden Begriffe von Autoritätsbeziehungen und vom Aufrechterhalten und Überschreiten von Grenzen entwickelt.
Notes: Summary Concepts of authority relations and boundary maintenance and crossing, developed at the Tavistock Institute, which have been found useful for understanding group and organizational dynamics, are presented. This conceptual viewpoint is then utilized in a case study of a group relations seminar comprised of high school students in a compensatory education program. Since this seminar had an unanticipated impact on the larger organization, of which it was a part, this perspective is also invoked to analyze this phenomena. Specifically, an attempt is made to inter-relate a group member's dream, the leaders' roles, an episode involving the total institution, and various staff reactions. Finally, some organizational aspects of resistance to change are highlighted.

Description / Table of Contents: Summary Fortynine lung emboli in the neurosurgical department of the Hamburg-Eppendorfer University Hospital were investigated in relation to the weather. The weather conditions on the day of death and also during the 8 preceding days were analysed. For comparison the dependency on clinical factors was also investigated. No significant correlation attributable to the weather conditions was found. The significance of modern differential medico-meteorological methods for this kind of research was expounded.

Notes: Abstract The detection of NMR in oriented nuclei via the Mößbauer effect in the daughter state is described and its specific advantages and drawbacks compared to other methods. As an example, the NMR of57Co in Fe was observed in external fields of 0–8 kG. The zero field frequency yields a magnetic momentμ 57/μ n =4.708±0.012. The different value found from the slope of the resonance frequency vs. external field is attributed to a Knight shift of (+2.9±1.2)%.

Notes: D,L-Leucine-NCA and D,L-methionine-NCA were copolymerized in varying mole fractions by using primary base initiation. The methionine segments were oxidized to both sulfoxide and sulfone functions by using peroxide oxidation. The compositions of the resulting polymers were established with the aid of 60 MHz NMR. The transport properties of aqueous solutes through membranes formed from these polymers were studied by means of a diffusive dialysis measurement. The transport of NaCl, urea, creatinine, uric acid, and bacitracin were found to be strongly dependent on the degree of hydration of the polymers. The relative rates of transport of these solutes were affected by interaction with the polymers, when the degree of hydration was not excessive. The degree of hydration was a function of the extent of methionine oxidation.
