    Notes: Abstract— Seven-day-old rats were injected intraperitoneally with l-phenylalanine (1 g/kg) and the time course of brain polyribosome disaggregation and changes in brain levels of phenylalanine, tryptophan and tyrosine were determined. Disaggregation of brain polyribosomes preceded the increase in levels of phenylalanine in brain, and followed the same time course as depletion of tryptophan from brain.The effects of several metabolites of phenylalanine (which are formed in phenylketonuria) on protein synthesis in vitro was determined for brain and liver systems. None of the compounds tested was inhibitory at concentrations below 10 mM and in all cases hepatic protein synthesis was more sensitive to inhibition than was the corresponding system from brain.Ribosomal dimers, formed in brain after injection of phenylalanine, were incapable of supporting high levels of protein synthesis in vitro, a finding that suggested that the inhibition of protein synthesis in vitro in cell-free systems of brain tissue after injection of phenylalanine into young rats was mediated by disaggregation of brain polyribosomes associated with tryptophan deficiency in brain.

    Notes: Background: Whether normal peripheral blood mononuclear cells (PBMCs) transferred to severe combined immunodeficient (SCID) mice produce specific IgE remains unclear. Methods: Mice received injections of Dermatophagoides farinae antigen (Df)-stimulated PBMCs from healthy persons (IgE RAST score of 0). Results: High titers of Df-specific IgE were detected. The Df-specific IgE activity produced was comparable to or higher than that produced by cells from patients with asthma although the time to maximal production was longer. IgE derived from PMBCs of healthy persons or patients with asthma induced histamine release from cultured human basophils that had been stimulated with Df antigen or an anti-IgE antibody. Treatment of Df-stimulated PBMCs with a high dose, but not a low dose, of interleukin-4 stimulated production of Df-specific IgE by PMBCs from healthy persons or patients with asthma. In contrast, intravenous injection of IFN-γ into reconstituted SCID mice decreased Df-specific IgE production by PBMCs from patients with asthma. In PMBCs from healthy persons, IgE class-switching may occur later and block the effects of treatment with IFN-γ. Conclusions: PBMCs from healthy persons and persons with asthma have clones reactive to allergen and produce functional IgE specific for relevant antigens in mite-sensitive bronchial asthma.

    Notes: We examined the effect of milrinone, a phosphodiesterase III inhibitor, on neuromuscular block induced by vecuronium. Thirty adult patients were randomly assigned to one of two equal groups: the milrinone group and the control group. Subjects in the milrinone group received an intravenous loading dose of milrinone 5 μg.kg−1.min−1 for 10 min, followed by an infusion at a rate of 0.5 μg.kg−1.min−1. Subjects in the control group received normal saline at a rate of 0.1 ml.kg−1.h−1. Thirty minutes after the beginning of the infusion of milrinone, anaesthesia was induced with intravenous thiopental 4 mg.kg−1 and fentanyl 2 μg.kg−1, and was maintained with isoflurane in oxygen and nitrous oxide. Neuromuscular blockade was monitored electromyographically at the adductor pollicis muscle. The times from the administration of vecuronium 0.1 mg.kg−1 to the onset of neuromuscular block and the return of the first, second, third, and fourth response of the train-of-four were compared between the two groups. Times to the recovery of the ratio of the first twitch to the control twitch to 25%, 50% and 75%, and times to the recovery of train-of-four ratio to 25%, 50% and 75% were also compared between the two groups. The onset of neuromuscular block in the milrinone group was significantly slower than in the control group. The times to the returns of the four twitches of the train-of-four, times to recovery of the ratio of the first twitch to the control twitch to 25% and 50%, and the times to the recovery of the train-of-four ratio to 25% and 50% were significantly shorter in the milrinone group than in the control group. We conclude that milrinone delays the onset of neuromuscular blockade but hastens its recovery in anaesthetised patients receiving vecuronium.

    Notes: Abstract: The β-decay of 146La was studied using the on-line isotope separator KUR-ISOL. Gamma-gamma angular correlation measurements were performed with a 4-Ge detectors system. Spin assignments of three levels were made: 3+ for the 1576.5 keV level, 4+ for the 1627.1 keV level and 5+ for the 1810.2 keV level. The mixing ratios (E2/M1) were deduced to be δ183.2= 0.25 ± 0.08, δ638.9= 0.33 ± 0.05, δ959.0= 1.19+0.16 −0.14, δ1015.9= 5.4+3.1 −1.5 and δ1318.1= 6.5+1.7 −1.1. These were compared to the calculated values obtained in three cases involving different Majorana force parameter values. The band structure of 146Ce is discussed based on the results of calculation using the IBM-2 theory.

    Notes: Abstract The lattice and bulk thermal expansions perpendicular to the layer plane of siliconated pyrolytic carbon, PC(Si), produced by pyrolyzing a mixture of propane gas and silicon tetrachloride vapour at the deposition temperatures of 1440 to 2025° C, have been measured over the temperature ranges 20 to 550° C and 20 to 960° C, respectively. The expansion behaviours of PC(Si) are related to the density and the degree of preferred orientation of crystallites, as is the case for pyrolytic carbon without silicon “PC”. At a deposition temperature of about 1700° C, the bulk thermal expansion coefficient of PC(Si) is about three times as large as that of PC.

    Notes: Abstract The TRAX gene encodes a Translin-associated 33-kDa protein partner, TRAX. The TRAX protein has extensive amino acid homology with Translin, and contains bipartite nuclear targeting sequences, suggesting a possible role in the selective nuclear transport of Translin lacking any nuclear targeting motifs. In the present study, genomic clones of the human TRAX gene were isolated to determine the complete genomic organization. The genomic structure of the human TRAX gene was similar to that of the human Translin gene, consisting of six exons and five introns, encompassing approximately 27 kb in genomic DNA. Northern blot analysis revealed a predominant transcript of approximately 2.7 kb, and its distribution in various tissues was like that of Translin. Chromosomal mapping by fluorescence in situ hybridization (FISH) analysis allowed localization of the TRAX gene to human chromosome 1q41.
