    Notes: Abstract We describe a new and highly effective optical frequency discriminator and laser stabilization system based on signals reflected from a stable Fabry-Perot reference interferometer. High sensitivity for detection of resonance information is achieved by optical heterodyne detection with sidebands produced by rf phase modulation. Physical, optical, and electronic aspects of this discriminator/laser frequency stabilization system are considered in detail. We show that a high-speed domain exists in which the system responds to the phase (rather than frequency) change of the laser; thus with suitable design the servo loop bandwidth is not limited by the cavity response time. We report diagnostic experiments in which a dye laser and gas laser were independently locked to one stable cavity. Because of the precautions employed, the observed sub-100 Hz beat line width shows that the lasers were this stable. Applications of this system of laser stabilization include precision laser spectroscopy and interferometric gravity-wave detectors.

    Notes: Abstract We describe a study, performed using heterodyne techniques, of the frequency fluctuations of two completely independent ring dye lasers locked to independent reference cavities. Single laser linewidths of less than 750 Hz were achieved, the principal limitation being residual vibrations from the noisy laboratory environment. With future design and environmental improvements, ultranarrow linewidths are expected thus providing a useful tool for a great variety of high precision experiments.

    Notes: Abstract The functional role of GABAergic inhibition in shaping the frequency tuning of 96 neurons in the torus semicircularis of the leopard frog, Rana pipiens, was studied using microiontophoresis of the GABAA receptor antagonist, bicuculline methiodide. Bicuculline application abolished, or reduced in size, the inhibitory tuning curves of 72 neurons. In each case, there was a concommitant broadening of the excitatory tuning curve such that frequency-intensity combinations that were inhibitory under control conditions, became excitatory during disinhibition with bicuculline methiodide. These effects were observed irrespective of the excitatory tuning curve configuration prior to bicuculline methiodide application. Results indicate an important role for GABA-mediated inhibition in shaping the frequency selectivity of neurons in the torus semicircularis of the leopard frog. Bicuculline application also affected several other response properties of neurons in the leopard frog torus. Disinhibition with bicuculline methiodide increased both the spontaneous firing rate (18 cells) and stimulus-evoked discharge rate (81 cells) of torus neurons, decreased the minimum excitatory threshold for 18 cells, and altered the temporal discharge pattern of 47 neurons. Additional roles for GABAergic inhibition in monaural signal analysis are discussed.

    Notes: Abstract In response to phosphorus limitation, the fungusNeurospora crassa synthesizes a number of enzymes that function to bring more phosphate into the cell. The NUC-2 protein appears to sense the availability of phosphate and transmits the signal down-stream to the regulatory pathway. Thenuc-2 + gene has been cloned by its ability to restore growth of anuc-2 mutant under restrictive conditions of high pH and low phosphate concentration. We mapped the cloned gene to the right arm of linkage group II, consistent with the chromosomal position of thenuc-2 mutation as determined by classical genetic mapping. Thenuc-2 + open reading frame is interrupted by five introns and codes for a protein of 1066 amino acid residues. Its predicted amino acid sequence has high similarity to that of its homolog inSaccharomyces cerevisiae, PHO81. Both proteins contain six ankyrin repeats, which have been implicated in the cyclin-dependent kinase inhibitory activity of PHO81. The phenotypes of anuc-2 mutant generated by repeat-induced point mutation and of a strain harboring a UV-inducednuc-2 allele are indistinguishable. Both are unable to grow under the restrictive conditions, a phenotype which is to some degree temperature dependent. Thenuc-2 + gene is transcriptionally regulated. A 15-fold increase in the level of thenuc-2 + transcript occurs in response to a decrease in exogenous phosphate concentration.
