Notizen: Summary To better understand the role of autoimmunity in the pathogenesis of dermatitis herpetiformis, linear IgA bullous dermatosis or other skin disorders, the antigenic specificity of the immune reactants bound in vivo in the skin must be identified. In order to do so, one must first be able to elute these immune reactants from the skin. We describe here a simple method of eluting not only specifically bound IgG, but also IgA and other immunoglobulins and complement components from skin biopsy material. The method involves cutaneous washing of the entrapped serum proteins in PBS pH 7 and pH 5 buffers followed by specific immunoglobulin elutions at pH 3 and 2. The IgA deposits which could not be removed by this treatment were eluted by a combination of low pH (0.5 M citrate pH 2) and a chaotropic agent (2 M NaCl). The relative concentration of IgA in eluates when quantitated by fluoroimmunoassay were three-to five-fold higher in dermatitis herpetiformis skin biopsy specimens, than in eluates of bullous pemphigoid or normal skin biopsy specimens.

Notizen: Summary This paper describes the results from a continuing research project started in 1984, which is investigating improved methods for predicting the underground climate in hot mines. The Homestake Gold Mine in South Dakota was the site for testing methods, based on radial heat flow theory, to measurein situ values of the thermal conductivity and diffusivity of rock. The methods used proved to be quite within the capability of mine staff to carry out for themselves. The results were acceptable and verified thatin situ thermal rock properties can differ considerably from laboratory determined values, in this case by a factor of approximately two. A mine climate simulation program, CLIMSIM, was applied to case studies from the Homestake mine using the measured thermal rock parameters. The results showed good correlation between the measured and simulated values.

Notizen: Positron annihilation lifetime spectroscopy has been used to study the isothermal relaxation response of compression molded bisphenol-A polycarbonate at temperatures of 263, 273, and 303 K. The temperature dependence of both the lifetime and intensity of the ortho-Positronium (o-Ps) pickoff component is discussed in terms of ductile-to-brittle transition behavior and free volume theory. An additive exponential model and the Williams-Watt model were used to analyze the relaxation as a function of temperature and provided results consistent with the anticipated molecular mobility of polycarbonate at sub-Tg temperatures.

Notizen: We present the first results from a new apparatus for high spectral and angular resolution studies of sputtered neutral species. The sputtering experiments employ laser-induced fluorescence and a compact fiber-optic-based detection system to yield state-specific angular distributions for the sputtered atom flux. The angular resolution is of the order of 3° and a 260 MHz residual Doppler shift is estimated for the observed atomic transitions. The angular distribution for ground-state zirconium atoms shows clear deviations from the cosine dependence expected from linear cascade theory when sputtered by 1.5 keV Ar+ ions incident at 60° from the surface normal. The peaking of the distribution in the forward direction suggests that a significant fraction of the atoms is ejected by a direct mechanism.

Notizen: Electron-stimulated desorption of neutrals from methanol-dosed 6063 aluminum alloy has been monitored by laser ionization at 193 nm wavelength. Primary electron energies were in the range 1-3 keV. Time-of-flight spectra for the neutral desorbate were characterized by a low value of the peak velocity (250 m s-1) and a non-Boltzmann form of the distribution. Neutral desorption was ascribed to the parent methoxy (CH3O) species, based on mass spectrometry of the laser-ionization fragmentation pattern as a function of intensity. Ionic desorption was dominated by H+, which was observed to have kinetic energy peaked at ∼3 eV. Approximate estimates of neutral and ion yields were made, based on ion-optical modeling of apparatus transmission characteristics.
