Synthesis of 17-ThiopregnanesHydrogen sulfide reacts with the 16β, 17β-epoxyketones 4a, c-e to give the trans-mercaptohydrins 5a, c-e, which, when treated with acetone or acetophenone, yield the 1,3-oxathiolanes 7a, c-e, and 8d, respectively. In addition to 7d, there was also isolated the 1β, 17β-thioacetonide 6a, c-e. and 8d, respectively. In addition to 7d, there was also isolated the 16β, 17β-thioacetonide 10. By oxidation of 7d there was obtained the sulfone 9. Acetylation of 5d yielded the 17α-thioacetate 13 and a second product which is probably the 17β-epimer 14. Treatment of 5e with methane sulfonyl chloride at O°C resulted in the formation of the bissteroid 15 whereas at 60°C there was isolated the episulfide 16 in addition to 15. Thiocyanic acid was capable of opening the 16β, 17β-epoxide 4d to form the isomeric thiocyanates 18 and 19 as well as the thiocarbonate 20

Schwefelwasserstoff reagiert mit den 16β, 17β-Epoxyketonen 4a, c-e zu den trans-Mercaptohydrinen 5a, c-e, die mit Aceton bzw. Acetophenon die 1,3-Oxathiolane 7a, c-e bzw. 8b bilden. Als Nebenprodukt von 7d wurde das 16β, 17β-Thioacetonid 10 isoliert. Durch Oxidation von 7d erhält man das Sulfon 9. Die Acetylierung von 5d führt zum 17α-Thioacetat 13 und vermutlich zum 17β-Epimeren 14. Unter Einwirkung von Mesylchlorid auf 5e bei O°C entsteht das Bissteroid 15, während bei 60°C das Episulfid 16 neben 15 isoliert wird. Rhodanwasserstoffsäure öffnet das 16β, 17β-Epoxid 4d unter Bildung der isomeren Thiocyanate 18 und 19 sowie des Thiocarbonats 20.
