    Notes: Abstract: The probabilities of distributions of amino acids and amino acid pairs in normal human haemoglobin α-chain and its seven variants causing α-thalassaemia were studied to elicit the randomness of their positioning. The results show that 47.37% of distributions of amino acids conform to a simple random distribution and the distributions of the remaining amino acids are very near to a simple random distribution. However, variants of these patterns lead to nine distribution probabilities to increase and thus to occur more often, one distribution probability slightly decreases but its rank with respect to its distribution probability is unchanged and four distribution probabilities to decrease and thus occur less often among 14 distribution probabilities there being the least random in occurence. The decreased distribution probabilities in variants are exclusively related to leucine.

    Notes: There are seven known vitamin K-dependent proteins in blood. These proteins require calcium ion for expressing their full biological activities. Calcium ion also induces conformational changes in this class of proteins. Taking advantage of the ligand induced conformational changes, a number of unique approaches of affinity chromatography have been developed. These methodologies have been very useful tools for both the purification and for understanding the structure-function relationships of this class of proteins. One method is the use of metal ion dependent immunoaffinity chromatography. The antigen can be dissociated from the antibodies with either the removal or addition of calcium ion under physiological conditions. The other method is pseudoaffinity chromatography. This method uses conventional ion-exchange or hydrophobic resin and manipulates the mobilities of the proteins on these resins by the presence or absence of calcium ions. Researchers working with other calcium binding proteins or other proteins that are known to undergo ligand induced conformational changes may benefit from the experience of these unique conformation-specific affinity chromatography approaches.

    Notes: Epitaxial crystallization behavior of HDPE/iPP double layers under quenching and annealing conditions has been studied by means of transmission electron microscopy (TEM). The results obtained from bright field TEM observations indicate that in the as-quenched state the HDPE that is in direct contact with the surface of the oriented iPP substrate recrystallizes in the form of oriented crystallites dispersed on the iPP substrate. The electron diffraction results show that besides the two normally observed epitaxial orientations between HDPE and iPP, there is also a special orientation with [001]HDPE ‖ [001]iPP. The HDPE which is in contact with the clean surface of a glass slide crystallizes in small lamellae with random orientation. In the boundary region, the epitaxially crystallized HDPE small lamellae stop right on the boundary of the oriented iPP film. If the quenched samples are annealed at 128°C (below Tm of HDPE) for 2 h, the small HDPE crystals grow to thick lamellae in both areas. But only the epitaxial orientation of HDPE with [001]HDPE ‖ [101]iPP has been observed.
