Zusammenfassung Es wird eine neue Möglichkeit ultrahistochemischer Untersuchungen mit Hilfe des Raster-Elektronenmikroskops mitgeteilt. Die Kombination eines Raster-Elektronenmikroskops mit einem Halbleiterdetektor und einem Vielkanalanalysator ermöglicht die Herstellung von Oberflächenbildern biologischer Präparate in Vergrößerungen von 20:1 bis 20000:1 und gleichzeitig auch eine gezielte Röntgenmikroanalyse der in diesen und noch kleineren Arealen (bis zu 1 µm Durchmesser) enthaltenen Elemente (von Na bis U). In ersten Untersuchungen an menschlichen Thrombocyten wird gezeigt, daß darüber hinaus durch histochemische Reaktionen bestimmte Elemente zur Ablagerung gebracht und semiquantitativ nachgewiesen werden können. Mit Hilfe chromaffiner und argentaffiner Reaktionen wurden mit dieser Methode halbquantitative Bestimmungen des Gehalts der Thrombocyten an Adrenalin, Noradrenalin, Dopamin und Serotonin durchgeführt. Es bestehen gute Gründe dafür, daß mit diesem neuen Prinzip halbquantitative und wahrscheinlich auch quantitative Bestimmungen des Gehalts oberflächlicher Gewebeschichten an einer ganzen Reihe von Stoffen möglich werden. Voraussetzung ist nur, daß diese Substanzen mit Hilfe spezifischer Reaktionen in Abhängigkeit von ihrer Menge ein im Gewebe nicht enthaltenes Element dort zur Ablagerung bringen.

Summary A new technique of ultra-histochemical analysis using the scanning electron microscope is discussed. The combination of a scanning electron microscope with a semi-conductor detector and a multichannel analyzer allowed us to produce surface views of biological specimens in magnifications from 20:1 down to 20000:1. Simultaneously, accurate X-ray microanalysis of the elements contained within these, and even smaller, areas (down to 1 µm diameter) from Na to U was possible. First trials with human platelets showed that—besides these facilities—, certain elements can be deposited through histochemical reactions and measured semi-quantitatively. This method, in combination with chromaffin and argentaffin reactions, permitted us to semi-quantitatively measure the concentrations of adrenalin, noradrenalin, dopamine, and serotonin in human platelets. There are strong reasons indicating that this new principle is well suited for the semi-quantitative, and probably, quantitative assessment of the concentrations of a whole series of substances in surface layers of tissues. The only prerequisite is that, by means of a specific reaction, an element of these substances normally not yet contained in the tissue to be studied be deposited therein, in a proportion corresponding to the concentration of the substance.
