Notizen: Thin films of giant magnetostrictive materials were formed by dc magnetron sputtering and ion beam sputtering processes. A systematic investigation was made to examine the effect of these deposition processes on the magnetic properties and magnetostriction of TbFe2 and (Tb, Dy)Fe2 thin films. The magnetostriction of the films formed by dc sputtering was in the range from 200 to 400 ppm at a magnetic field H=15 kOe, whereas the films formed by the ion beam sputtering exhibited a slightly higher magnetostriction. For both processes, the increase in Ar partial pressure in the deposition processes strongly changed magnetic anisotropy from perpendicular to in plane, and increased magnetostriction and magnetostrictive response at low magnetic fields. However, the mixture of Xe gas into Ar gas in the dc sputtering lowered magnetization and gave almost no effect on magnetic anisotropy. © 2000 American Institute of Physics.

Notizen: MgO thin films were prepared by ion plating systems and by an ion beam sputtering system with a plasma filament type ion source. Triode-type and rf ion plating processes were employed. MgO films were prepared with various oxygen partial pressures ranging from 10−3 to 10−2 Pa. In the triode-type ion plating process, the MgO(200) was the preferred growth orientation of film. In the rf ion plating process, MgO(111) and (220) reflection peaks appeared in the x-ray diffraction patterns of films. The films were transparent, and those values of transmittance of ultraviolet region were increased as increasing oxygen partial pressure. In the ion beam sputtering process, the film showed amorphous structure and low transmittance. © 2000 American Institute of Physics.

Notizen: Background  The naevoid basal cell carcinoma syndrome (NBCCS) is an autosomal dominant disorder characterized by tumorigenesis such as multiple basal cell carcinomas, odontogenic keratocysts and developmental abnormalities such as calcified dural folds and rib-anomalies. Recently, it has been shown that ultraviolet (UV) B exposure produced more BCCs in ptch knockout mice than wild mice. Objectives  To Investigate the role of UV in development of BCCs in NBCCS, cellular sensitivity to killing by UVB and removal of UVB-induced oxidative DNA damage were examined using fibroblasts derived from patients with NBCCS under physiologically relevant doses of UVB exposure.Patients and methods  Three patients with NBCCS, a 59-year-old male patient, an 18-year-old boy and a 13-year-old boy were examined by photobiological analysis. Cellular sensitivity to killing by UVB and UVC and removal of oxidative DNA damage caused by UVB were tested using fibroblasts derived from these patients. We measured cellular 8-hydroxydeoxyguanosine (8-OHdG) after UVB exposure up to 24 h after UVB exposure using high-performance liquid chromatography. Results  All three cell strains derived from the patients with NBCCS were hypersensitive to killing by UVB (D10: 50–70% of normal) but not by UVC. After UVB exposure, the production of 8-OHdG increased dose dependently up to 3200 J m−2 in both NBCCS cells and normal cells. In normal cells, 8-OHdG after UVB exposure returned to its basal level during 24 h, whereas in NBCCS cells the amount of 8-OHdG after 800 J m−2 of UVB exposure did not return to its basal level even after 24 h. The result indicates the removal of 8-OHdG could be impaired in NBCCS cells. Ability in removal of thymine dimers of NBCCS cells was similar to that of normal cells. Conclusions  Hypersensitivity to UVB can be one of the diagnostic tools of NBCCS for those whose clinical features have not yet completed. Hypersensitivity to cell killing and the impairment of removal of 8-OHdG after UVB exposure may play some role in developing BCCs and other tumours in NBCCS.

Notizen: 1. Renal effects of nisoldipine, a potent calcium channel blocker, were examined in anaesthetized dogs.2. Intrarenal arterial infusion of nisoldipine (10, 50 ng/kg per min) did not influence mean systemic blood pressure and renal haemodynamics, but did cause dose-related increases in urine flow and urinary excretion of electrolytes. During nisoldipine infusion at the higher dose, the levels of fractional excretion of sodium, potassium and chloride were significantly elevated.3. In the lithium clearance study, fractional lithium excretion, an index of fractional sodium excretion in the proximal tubules, was unchanged even with the higher dose of nisoldipine, whereas fractional sodium excretion in the distal tubules markedly increased.4. Nisoldipine elicited a significant increment in osmolar clearance without altering the reabsorption of free water.5. Thus, intrarenal administration of nisoldipine at a non-hypotensive dose to anaesthetized dogs produced a significant diuretic effect without alteration in renal haemodynamics. Results in lithium clearance and free water clearance studies suggest that nisoldipine exerts a diuretic action by inhibiting sodium transport at the distal nephron segments beyond the proximal tubules.

Notizen: 1. We examined the effects of torasemide (0.3 and 1 mg/kg i.v.) on renal haemodynamics and function employing renal clearance and stop-flow techniques in anaesthetized dogs and compared these with furosemide (1 and 3 mg/kg i.v.).2. Torasemide and furosemide did not influence renal haemodynamics, in the renal clearance study, but caused a dose-related and significant increase in urine flow and urinary excretion of sodium and potassium. Torasemide and furosemide increased fractional excretion of sodium in the distal tubules with a relatively small increase in the fractional excretion of lithium (index of sodium excretion at the proximal tubules, FELi). The diuretic profile of torasemide was of long duration, compared with that of furosemide.3. Torasemide and furosemide inhibited sodium reabsorption at the distal portion of the tubules in the stop-flow study.4. It is suggested from these results, that the main diuretic site of action of torasemide is the ascending limb of the loop of Henlé.

Notizen: 1. Effects of calcium-interacting agents on the calcium-induced stimulation of renin release from kidney cortical slices pretreated with calcium-free medium were examined.2. The exposure of calcium to the slices pretreated with calcium-free medium enhanced the release of renin, followed by a decreased response in the release. The amount of lactate dehydrogenase released from the slices did not correlate with that of renin.3. High potassium depolarization significantly potentiated the decreased response of renin release, with no influence on the stimulation of the release by exposure to calcium.4. The decrease in renin release was attenuated by calcium-interacting agents, such as nifedipine, TMB-8 and W-7, but these agents were without effect on the stimulation of the release by exposure to calcium.5. Thus, the calcium-calmodulin system apparently is not involved in the increased response of renin release following exposure to calcium.
