    Notes: Keratinous tissues play two major roles in the adaptation of vertebrates to their environment: a strong mechanical support and a chemical barrier. In order to determine whether these properties may originate from different zones in the tissues, microdiffraction experiments on the micrometre scale have been carried out on feather shaft, horse and human hair, and porcupine quill samples. The existence of several structural layers has been revealed in all the tissues, some corresponding to highly ordered α- or β-type keratin and the others to more or less amorphous keratin. The existence of lipid granules has also been evidenced, mainly in the outer layers. This study shows one of the possibilities which are now offered by third-generation synchrotron sources for the structural microanalysis of biological tissues.

    Notes: The microfocus X-ray beamline at the European Synchrotron Radiation Facility has been used to investigate the variation in molecular orientation and crystallinity in the wall of a container fabricated from poly(ethylene terephthalate). Two-dimensional wide-angle X-ray scattering patterns were recorded and displayed in real time as the specimen was tracked across the incident X-ray beam enabling the measurement of textural changes to be made with a spatial resolution of 2̃ μm.

    Notes: With the opening of the first real `third-generation' synchrotron source in Grenoble, in fall 1994, X-ray sources of unprecedented brilliances and qualities became available to the scientific community. Different X-ray analytical techniques could now be applied on a level that was unimaginable only a decade ago. Here are some preliminary results from an experiment where different analytical techniques have been applied on a micrometer level carried out at the most powerful synchrotron microbeam currently available in the world, the microfocus beamline (BL1) at ESRF. This beamline can now provide micrometer-sized X-ray beams with a flux density up to 1010 photons μm−2 at an energy of 13 keV and with a bandwidth of 10–4. In this experiment, X-ray diffraction and X-ray fluorescence have been combined in order to obtain a precise and comprehensive micro-analytical description of micrometer-sized fly ash particles. These types of particles are heavily inhomogeneous with a very irregular shape that makes them inaccessible to conventional micro-analysis. The experiment was performed in a scanning mode and two-dimensional images of different analytical information were reconstructed from the data recorded during the scan. The major features and limitations of this micro-analytical technique will be outlined and different examples on how the analytical information can be used for generating two-dimensional images of the sample will be demonstrated and discussed.

    Notes: The microfocus X-ray beamline at the European Synchrotron Radiation Facility has been used to investigate the variation in molecular orientation and crystallinity in spherulites of the organic polymer poly-3-hydroxybutyrate (PHB). This is the first report of the correlation of optical and X-ray measurements on spherulitic polymer films where X-ray diffraction patterns have been recorded and displayed continuously in real time while the specimen was tracked in steps of 10 μm across an incident X-ray beam with a diameter as small as 10 μm.

    Notes: Summary A transformation system that allows regeneration of transgenic pea plants from calli selected for antibiotic resistance was developed. Explants from axenic shoot cultures and seedling epicotyls were cocultivated with nononcogenic Agrobacterium tumefaciens strains, and transformed callus could be selected on callus-inducing media containing either 15 mg/l hygromycin or 75 mg/l kanamycin. After several passages on regeneration medium, shoot organogenesis could be reproducibly induced on hygromycin-resistant calli, but not on the calli selected for kanamycin resistance. Regenerated shoots could subsequently be rooted and transferred into the greenhouse. In addition, the effects of different callus-inducing and growth media on organogenesis were investigated. The transformation of the calli and regenerated plants was confirmed by DNA analysis.

    Notes: Summary An analysis of the progeny of primary transgenic pea plants in terms of transmission of the transferred DNA, fertility and morphology is presented. A transformation system developed for pea that allows the regeneration of fertile transgenic pea plants from calli selected for antibiotic resistance was used. Expiants from axenic shoot cultures were co-cultivated with a nononcogenic Agrobacterium tumefaciens strain carrying a gene encoding hygromycin phosphotransferase as selectable marker, and transformed callus could be selected on callus-inducing media containing 15 mg/l hygromycin. After several passages on regeneration medium, shoot organogenesis could be reproducibly induced on the hygromycin resistant calli, and the regenerated shoots could subsequently be rooted and transferred to the greenhouse, where they proceeded to flower and set seed. The transmission of the introduced gene into the progeny of the regenerated transgenic plants was studied over two generations, and stable transmission was shown to take place. The transgenic nature of the calli and regenerated plants and their progeny was confirmed by DNA and RNA analysis. The DNA and ploidy levels of the progeny plants and primary regenerants were studied by chromosome analysis, and the offspring of the primary transformants were evaluated morphologically.

    Notes: Abstract Total levels of IgA, IgM, and IgG were measured in unstimulated whole saliva and serum from 63 individuals with selective IgA deficiency. Values were compared with the incidence of upper respiratory tract infections, antibiotic treatments (necessitated by upper respiratory tract infections), and HLA antigens. A statistically significant increase in salivary IgM and IgG levels was noted in individuals with selective IgA deficiency compared to healthy normal individuals. Healthy individuals with selective IgA deficiency did not have increased concentrations of salivary IgM compared to infectious-prone patients. Nor was there any correlation found between proneness to infections and HLA antigens or between salivary IgM or IgG levels and HLA antigens in this patient material.

    Notes: By using the total reflecting properties of x-rays inside glass capillary tubes, intense fine or microbeams of x-rays can be obtained. In combination with conventional diffraction x-ray tubes, microbeams with a diameter of a few micrometres and with an intensity sufficient for trace element analysis can be achieved. The basic physics behind the capillary technique is reviewed and an energy-dispersive x-ray fluorescence microbeam spectrometer for trace element analysis is described. Examples of applications of the microbeam spectrometer are given and the future perspectives of the capillary technique are discussed.
