    Notes: Summary The oxidation of the C-1 atom of glucose through the pentose shunt of adipose tissue, under maximal stimulation by insulin, was enhanced by rat serum. The material responsible for the increase of the efficacy of insulin by serum was ultrafiltrable and partly dialysable. At submaximal concentrations of insulin, the serum-induced enhancement of responses to insulin resulted in an increase of the relative potency of insulin (potentiation). Sodium acetate modified the log dose-response curve of insulin in the same way as serum did. Furthermore, acetate was active at low concentrations, suggesting that serum acetate might contribute to the sensitization to insulin by serum. In the presence of glucose and insulin, acetate is known to activate the rate of synthesis of fatty acids, which, in turn, determines the activation of the pentose shunt. Other constituents of serum known to influence the metabolism of adipose tissue in the same way as acetate may be expected to contribute to the sensitization to insulin by serum.

    Notes: Summary The hyperglycemic response to two 2-ml injections of guinea pig anti-insulin serum (GPAIS), given at a 1.5 h interval, was measured after 18 h of fast in nephrectomized rats, intact animals and animals with anuria caused by ligating the ureters. In controls plasma glucose rose by + 196±4.0 mg/100 ml within 1.5 h after G-PAIS injection, then levelled off with the onset of glucosuria and began to decline at 4.5 h. Anuria due to ureteral obstruction did not affect the initial rise of plasma glucose (+ 197±11.9mg/100ml at 1.5h). In contrast, the initial rise of plasma glucose amounted to only + 136± 5.3 mg/100 ml at 1.5h in nephrectomized rats. Blood sugar remained lower in the latter animals than in those with ligated ureters for the rest of the experiment. The blunted glycemic response to GPAIS in anephric rats, thus, is due to the loss of kidney tissueper se. It is suggested that, after sudden insulin deprivation, the kidneys might release sizeable amounts of newly synthesized glucose into the circulation.

    Notes: Summary The “dilution effect” on serum insulin-like activity (serum ILA) as measured by the rat fat pad bioassay, i.e. an apparent increase of the potency of serum insulin-like material after dilution, was confirmed.—The “dilution effect” was found to affect exclusively the fraction of serum ILA not suppressible by anti-insulin serum (nonsuppressible ILA). The slope of the log-dose response curve of the heterogenous nonsuppressible fraction of serum ILA is flatter than the slope of the log-dose response curve of standard insulin. Neither the potency of the fraction of serum ILA suppressible by anti-insulin serum (suppressible ILA), nor that of crystalline insulin added to serum, were increased by dilution.

    Notes: Summary Renal cortical and plasma renin activities and the electron microscopic appearance of juxtaglomerular epitheloid cells were investigated in the unclamped renin-depleted kidney of rats with two-kidney-Goldblatt hypertension after removal of the clamped kidney. Within 24 h the renal renin content fell to still lower values, on the 2d day, it rose to one half its normal value, on the 7th day it reached its control value. Renin activity of plasma did not significantly differ from zero until the 3d day after removing the clamped kidney, reached 20% of normal on the 7th day and normal values between the 7th and the 21 st day. Sparse secretory granules appeared in the remaining renal cortex 12 h after removal of the clamped kidney. After 24 h secretory granules were formed by confluence of Golgi vesicles. After 2 days cisternae appeared in the rough-surfaced endoplasmic reticulum. After 4 days the electron-microscopic picture became normal. Renin appears to be elaborated in the rough-surfaced endoplasmic reticulum and concentrated into granules by the Golgi-apparatus. A fraction of the renal renin does not appear to be available for secretion.
