    Notes: Isoenzyme variation of 198 isolates of Phytophthora infestans collected from many locations in Japan during 1987–90, and of four pre-1987 isolates, was examined using starch gel electrophoresis. A previously unreported allele at malic isoenzyme locus/ME (90) was observed. An association between mating types and isoenzyme genotypes at three isoenzyme loci, glucosephosphate isomerase (GPI-1), peptidase (PEP-1) and ME, was found. At PEP-1, the A2 isolates from Japan had a previously unreported genotype (96/96). Normal segregation at the malic isoenzyme locus occurred in a cross of Japanese and Mexican parents. The results provide evidence of a change in the population genetic structure of P. infestans in Japan.

    Notes: Abstract Benign biliary stricture developing after hepatectomy is uncommon and its management remains controversial. We experienced a case of biliary stricture following hepatectomy for traumatic hepatic rupture, which was successfully treated with percutaneous transhepatic dilatation. Percutaneous transhepatic dilatation may be considered as one of the options in the treatment of anatomically complicated cases.

    Notes: Summary The renal effects of manidipine hydrochloride were investigated in ten hospitalised patients with mild-to-moderate essential hypertension. After a one-week placebo period, manidipine was given for 1 week in a dose rising from 5 mg to 10 mg or 20 mg daily to normalise the mean blood pressure measured after 2 h. Blood pressure had decreased from 171/101 to 147/86 mm Hg at the end of manidipine treatment. The pulse rate was unaltered. Renal vascular resistance decreased from 1.90 to 1.33 dyn · s · cm−5/1.48 m2 × 104, and renal blood flow and glomerular filtration rate increased from 522 to 662 ml · min−1 · 1.48 m−2 and from 81 to 93 ml · min−1 · 1.48 m−2, respectively, in spite of a fall in renal perfusion pressure. Manidipine reduced the filtration fraction from 0.260 to 0.243, suggesting a preferential reduction in efferent arteriolar resistance. The fractional excretion of sodium and potassium did not change. Manidipine did not produce any significant alteration in plasma renin activity or in the plasma aldosterone concentration. The results indicate that manidipine has favourable renal effects and a concomitant hypotensive action in patients with mild-to-moderate essential hypertension.

    Notes: Summary We have developed a simple HPLC assay to measure the activity of S-mephenytoin 4-hydroxylase in human liver microsomes, and have assessed its practical applicability by determining the kinetic parameters of the enzyme in 10 different human liver samples. The recovery of 4-hydroxymephenytoin and phenobarbital (the internal standard) after the precipitation of microsomal protein was almost complete, and the coefficients of variation for the intra-and interassay measurement of S-mephenytoin 4-hydroxylase activity were 〈6.4 and 8.0%, respectively. Eadie-Hofstee plots for the formation of 4-hydroxymephenytoin gave a straight line for all of the 10 samples studied. There was large interindividual variability in the kinetic parameters estimated: 4.6- (36 to 166 μM), 11.8- (0.9 to 10.6 nmole/mg protein/h) and 30.1-times (0.10 to 3.01 μl/mg protein/min) for Km, Vmax and Vmax/Km, respectively. The mean (±SD) Km, Vmax and Vmax/Km were 72.4±40.4 μM, 4.23±2.88 nmole/mg protein/h and 1.33±1.02 μl/mg protein/min, respectively. Thus, the assay was sufficiently accurate and reproducible to permit estimation of the kinetic parameters of S-mephenytoin 4-hydroxylase in human liver microsomes, and it appears to be applicable to an in vitro study of the possible involvement of S-mephenytoin-type oxidation polymorphism in drug metabolism.

    Notes: Summary The nucleotide sequences of the 5′-portion of the HCV genome, consisting of 2672 nucleotides, were determined by analysis of clones derived from 5 different patients with liver cirrhosis and hepatocellular carcinoma. Comparison of 5 genomes and 8 isolates already reported demonstrated the significant genome diversity in the structural regions. Genetic divergence was outstanding in the envelope regions (E1 and NS1/E2, 〉59.7% identity) in contrast to the sequence conservation of the 5′ non-coding (〉93.8% identity) or core regions (〉82.0% identity). Although hypervariable regions were found in the NS1/E2 region as reported, hydropathicity was maintained for all clones, reflecting the biological functions of these regions.

    Notes: Abstract Three- and ten-ply SiC fibre-reinforced Ti-6Al-4V composites were joined using a laser beam. With a 300 μm thick Ti-6Al-4V filler metal, fully penetrated welds without apparent fibre damage, could be obtained in welding directions both parallel and transverse to the fibre direction by controlling the welding heat input. Excess heat input resulted in the decomposition of SiC and subsequent TiC formation, and also caused degradation of joint strength. The welding of the three-ply composite in which full penetration was achieved at lower laser power, exhibited higher flexibility in heat input than that of the ten-ply composite. Heat treatment at 1173 K after welding improved the joint strength because of the homogenization of the weld metal and decomposition of TiC. The strengths of the transverse weld joints after the heat treatment were approximately 650 and 550 MPa for the three- and ten-ply composites, respectively. With the welding direction parallel to the fibre direction, the strengths both parallel and transverse to the weld joint were equivalent to those of the base plate.
