    Notizen: Crystals have been grown of a mannose-specific lectin from bluebell (Scilla campanulata) bulbs in a form suitable for X-ray diffraction studies. The crystals, which diffract to high resolution, grew in hanging drops by vapour diffusion, equilibrating with a solution of 70% saturated ammonium sulfate at pH 4.7–4.8 at 293 K, in the absence of any mannose saccharides. Crystals are orthorhombic, P21212, with unit-cell dimensions a = 70.78, b = 93.69, c = 46.92 Å. The functional lectin molecule is organized as a tetramer of four identical 14 kDa subunits, with only two subunits in the asymmetric unit. Data to 1.86 Å resolution have been recorded and the structure determined by the molecular replacement method.

    Notizen: A mannose-specific lectin from Calystegia sepium has been crystallized by the hanging-drop vapour-diffusion method using ammonium sulfate as precipitant. The needle-shaped crystals are orthorhombic, space group C2221 with cell dimensions a = 55.2(1), b = 55.9 (1), c = 196.1 (1) Å. Fresh crystals diffract to 1.9 Å resolution on a synchrotron radiation source. The asymmetric unit contains a dimer of two identical 16 kDa subunits with a packing density of 2.36 Å3 Da−1. Intensity data have been observed beyond 2.0 Å, but reasonable statistics restricted the usable range to 2.0 Å.

    Notizen: Abstract Insect feeding trials were carried out to determine the effects of a range of mannose-specific lectins on third instar nymphs of the rice brown planthopper (BPH),Nilaparvata lugens. Stål. Dose response curves show thatGalanthus nivalis agglutinin (GNA) has the strongest toxic effect of the lectins tested, and is effective at concentrations considerably lower than those previously reported.Narcissus pseudonarcissus agglutinin (NPA) andAllium sativum agglutinin (ASA) exhibit a significant antimetabolic effect towards the insect but were less effective (on a molar basis) than GNA.LC50 values for GNA, NPA and ASA are approximately 4 μM, 11 μM and 〉40μM respectively. These mannose-specific lectins are serologically identical, but differ in the number of subunits per protein molecule; ASA is a dimer, NPA is a trimer and GNA is a tetramer. The results obtained support the hypothesis, that the effectiveness of the mannose-binding lectins as antimetabolites is determined by the number of subunits per molecule. Two N-acetylglucosamine binding lectins, the dimericOryza sativa agglutinin (OSA) and the monomericUrtica dioica agglutinin (UDA), were also tested but at a concentration of 0.1% w/v exhibited no significant antimetabolic effect towards BPH, although the related lectin wheatgerm agglutinin (WGA) has previously been demonstrated to be toxic towards the insect.

    Notizen: Abstract The spectrum of lectin binding sites as it emerges during embryonic development of Drosophila was analysed by means of fluorescein-labelled lectins. As development and morphogenesis proceed, the reaction pattern becomes more and more complex. Mannose/glucose-, mannose-, N-acetylglucosamine- and poly-N-ace-tylglucosamine-specific lectins bind ubiquitously. Nuclear envelopes only have binding sites for wheat germ agglutinin. N-acetylgalactosamine-binding lectins are specific for ectodermal derivatives. Gaβ-3-N-acetylgalac-tosamine-binding lectins are highly selective markers for neural structures, haemocytes and Garland cells. It is also shown that Drosophila laminin is differentially glycosylated. The possible implications of differential and germ layer-specific glycosylation are discussed.

    Notizen: Abstract Rice (Oryza sativa, L.) lectin is specifically synthesized during seed formation. It accumulates very rapidly between 8 and 16 days post anthesis and reaches its final content 20 days post anthesis. The rate of synthesis of this protein dramatically increases during the developmental phase coinciding with the rapid lectin accumulation. When the lectin accumulation stops, its rate of synthesis declines rapidly and eventually stops in fully mature embryos. It appears, therefore, that rice lectin is a maturation-specific protein.

    Notizen: Summary Each of the three genomes in hexaploid wheat controls the expression of a specific lectin in the embryo. The chromosomes which control their synthesis were determined using nullisomic-tetrasomic and inter-varietal chromosome substitution lines of ‘Chinese Spring’. All three wheat lectins were shown to be controlled by the homoeologous group 1 chromosomes. Using ditelosomic lines of ‘Chinese Spring’ the lectin genes could be localized on the long arms of chromosomes 1A and 1D. Inter-specific addition and substitution lines of Aegilops umbellulata chromosomes to ‘Chinese Spring’ indicated that chromosome 1U, which is homoeologous to the group 1 chromosomes of wheat, controls lectin synthesis.

    Notizen: Abstract The range of sap-sucking insect pests to which GNA, (the mannose specific lectin from snowdrops (Galanthus nivalis) has been shown to be insecticidal in artificial diets has been extended to include the peach potato aphid (Myzus persicae). A gene construct for constitutive expression of GNA from the CaMV35S gene promoter has been introduced into tobacco plants. A transgenic tobacco line which expresses high levels of GNA has been shown to have enhanced resistance toM. persicae in leaf disc and whole plant bioassays,demonstrating the potential for extending transgenic plant technology to the control of sap-sucking insect pests.
