    Notes: Specific antibodies isolated by immunoabsorption on four main insolubilized allergens from Dermatophagoides pteronyssinus (DPT) had the following isotypic distribution: in 16 atopic patients, 52% IgG, 40% IgM, 8% IgA, 0.1% IgE and, in 12 non-atopic individuals, 48% IgG, 46% IgM, 6% IgA, 0.03% IgE. The ratios between geometric means of antibody values in each class (atopic vs non-atopies) were 2.4 for IgG, 2.0 for IgM, 2.8 for IgA and 66.7 for IgE. The amount of anti-DPT antibodies in IgG subclasses (did not follow the usual distribution of total IgG subclasses, i.e., IgG1 〉 lgG2 〉 IgG3 〉 IgG4. In atopies the order was IgG2 〉 IgG1 〉 IgG4 〉 lgG3 and in non-atopies, IgG4 〉 IgGl - IgG2 〉 IgG3 although 6/12 of the latter had no detectable (〈 0.5 μg/ml plasma) IgG4.

    Notes: Abstract The structure of bis(acetylacetonato)copper(II) has been redetermined from single crystal X-ray diffraction data. The unit cell parameters area=11.331(9),b=4.697(3),c=10.290(9) Å, andβ=91.84(7)dg. A finalR=0.056 for 727 observed reflections was obtained. The O-Cu-O chelate (bite) angle is 93.2(2)°. The two Cu-O bond lengths are 1.914(4) and 1.912(4) Å. The complex has a slight chair structure and a step angle of 7.05°.

    Notes: Summary The present study aimed at comparing the effects of nifedipine, a dihydropyridine “Ca2+ antagonist”, and of BAY K 8644, a dihydropyridine “Ca2+ agonist”, on the short term (5 min) 45Ca uptake and the cytosolic Ca2+ concentration of rat pancreatic islet cells incubated in the presence of physiological concentrations of glucose. Nanomolar concentrations of nifedipine increased the short term 45Ca uptake while micromolar concentrations decreased it. Cd2+, an inorganic Ca2+ channel blocker, reduced the stimulatory effect of nifedipine. Low concentrations of BAY K 8644 stimulated 45Ca uptake whereas high concentrations decreased it. In contrast, verapamil, a phenylalkylamine type calcium antagonist, only provoked a dose-dependent reduction in 45Ca uptake. Lastly, low concentrations of both nifedipine and BAY K 8644 raised the fluorescence intensity of fura 2 loaded islet cells. These findings indicate that nifedipine and BAY K 8644 may exhibit agonistic and antagonistic actions on B-cell voltage-dependent Ca2+ channels. This dualistic behaviour is markedly concentration-dependent and appears to be inherent to the 1,4-dihydropyridine compounds.

    Notes: Abstract The present study aimed at comparing the effects of low concentrations of BPDZ 44, a new pyridothiadiazine derivative, and diazoxide on86Rb outflow,45Ca outflow,45Ca uptake and insulin release from rat pancreatic islets. Both drugs caused similar modifications, but the effects of BPDZ 44 on the cationic and secretory events were much more marked than those of diazoxide. It is suggested that BPDZ 44 could be valuable tool for further studies of the KATP channels.

    Notes: Abstract l-Glutamine enhances insulin release evoked byl-leucine in isolated rat pancreatic islets. The enhancing action ofl-glutamine, which is a rapid but steadily increasing and not rapidly reversible phenomenon, is not attributable to any major change in either K+ or Ca2+ outflow from the islet cells. It coincides with an apparent increase in Ca2+ inflow rate and, hence, with Ca accumulation in the islets. The initial ionic response tol-leucine is not qualitatively altered by the presence ofl-glutamine. In their combined capacity to stimulate45Ca net uptake in the islets,l-glutamine can be replaced byl-asparagine but not byl-glutamate, whereasl-leucine can be replaced byl-norvaline orl-isoleucine, but not byl-valine, glycine orl-lysine. Such a specificity is identical to that characterizing the effect of these various amino acids upon insulin release. It is postulated that the release of insulin evoked by the combination ofl-leucine andl-glutamine involves essentially the same remodelling of ionic fluxes as that evoked by other nutrient secretagogues with, however, an unusual time course for the functional response tol-glutamine.

    Notes: Abstract A rise in extracellular glucose concentration from 8.3 to 16.7 mM stimulates both Ca2+ inflow and K+ exit in perfused rat pancreatic islets. These ionic changes are associated with an increase in bioelectrical spiking activity. From a quantitative analysis of45Ca and86Rb outflow from prelabelled islets, it is proposed that each electrical spike coincides, approximately, with the entry of 0.8 fmol of Ca2+ and exit of 3.6 fmol of K+ per mm2 of cell surface.

    Notes: Summary A rise in the extracellular concentration of glucose from an intermediate to a high value changes the burst pattern of electrical activity of the pancreatic B-cell into a continuous firing, and yet activates the B-cell Ca2+-sensitive K+ permeability. The hypothesis that glucose exerts such effects by inhibiting the Na+, K+-ATPase was investigated. Ouabain (1 mM) mimicked the effect of 16.7mm glucose in stimulating86Rb,45Ca outflow and insulin release from perifused rat pancreatic islets first exposed to 8.3mm glucose. The stimulation by ouabain of86Rb outflow was reduced in the absence of extracellular Ca2+ and almost completely abolished in the presence of quinine, and inhibitor of the Ca2+-sensitive K+ permeability. In the presence of ouabain, a rise in the glucose concentration from 8.3 to 16.7mm failed to stimulate86Rb outflow. However, the rise in the glucose concentration failed to inhibit86Rb influx in islet cells, while ouabain dramatically reduced86Rb influx whether in the presence of 8.3 or 16.7mm glucose. These findings do not suggest that inhibition of the B-cell Na+, K+-ATPase represents the mechanism by which glucose in high concentration stimulates86Rb outflow and induces continous electrical activity in the B-cell.

    Notes: Abstract Restriction fragment length polymorphism was assessed in wild and cultivated populations of Hevea brasiliensis using random probes from an Hevea nuclear library. One-hundred-and-sixty-four individuals were surveyed, and the results discussed in the light of previous work performed on isozyme variation. Both studies show that germplasm collections have led to an effective enrichment of the genetic resources available for Hevea breeding, and that cultivated clones have conserved a relatively high level of polymorphism, despite their narrow genetic base and their high level of inbreeding. An equivalent level of polymorphism is revealed by random nuclear probes and isozymes. However, the genetic structuring of the diversity appears more striking using RFLP markers. Wild accessions can be divided into three genetic groups according to their geographical origin. The present results are an essential guide to the incorporation of wild material in breeding schemes.
