    Notes: Abstract: Escherichia coli was grown in carbon- and energy source-limited fed batch cultures to study the effect of osmotic stress and different feed rates on the growth kinetics. An unstructured model based on the linear equation for substrate consumption provided an adequate description of the bacterial growth during the first phase of biomass production (20 h), except for cultures exposed to osmotic stress by the addition of 0.5 M NaCl. The addition of salt to the culture media had a large effect on the energetics, that could not simply be described in terms of an increased maintenance requirement. In the later phase of growth, an extensive decline in viability for all cultures was observed. Coincidentally, the specific sugar uptake rate approached a lower limit. It is concluded that the total obtainable biomass in a fed batch culture is strongly affected by the magnitudes of the substrate feed rate and the ionic strength of the culture medium.

    Notes: Abstract: The production of the fusion protein staphylococcal protein A/E. coliβ-galactosidase in Escherichiacoli was studied in batch and fed batch cultivations. Batch cultivation of a recombinant E. coli strain yielded a final cell dry weight of 16.4 g 1-1 with a final intracellular product concentration of recombinant protein corresponding to approximately 38% of the cell dry weight. Fed batch cultivation made it possible to increase the final cell dry weight to 77.0 g 1-1. The intracellular product concentration (25%) was lower as compared to batch cultivation resulting in a total concentration of recombinant protein of 19.2 g 1-1.

    Notes: The protein releases, the particle size distribution and the viscosity of disrupted E. coli suspensions from Dyno Mill KDL, Manton Gaulin 15 M-8TA and Microfluidizer M-110 were determined. The effects of these parameters on separation of the cell debris from the protein solution by centrifugation and by filtration were also examined. All three disintegration methods investigated give approximately the same protein and enzyme releases but considerably different physical properties of the cell disintegrates which influences centrifugation and filtration. The separation degree of biomass during centrifugation is only slightly affected by increasing degree of disruption (increasing protein releases) in the bead mill, while an increase in the degree of disruption in the two high pressure homogenizers drastically reduces the centrifugal degree of separation. However, increasing degrees of disruption result in shorter filtration times during filtration for all three disintegration methods. The results show further that the cell concentration only has a minor influence on protein releases in the Microfluidizer high-pressure homogenizer, while an increase in the biomass content reduces the separability of the cell disintegrate both in filtration and in centrifugation.

    Notes: Summary The temperature induced intracellular production of the fused protein SpA-βgal (protein A/β-galactosidase) withE. coli was compared in batch and fed batch culture. By introducing fed batch cultivation a final cell dry weight of 77.0 g/l was achieved, as compared to 16.4 g/l in batch cultivation. The concentration of SpA-βgal in the fed batch cultivation was very high, 19.2 g/l. This corresponded to 25% of the cell dry weight.
