    Notes: Abstract Limonin can be effectively degraded by Rhodococcus fascians cells. These bacteria can be entrapped in κ-carrageenan, and used in a continuous stirred tank reactor to degrade limonin in a continuous process. The effects of temperature, limonin concentration, dilution rate, and aeration on the reactor behaviour have been tested, and the results correlated with changes in limonin conversion, substrate degradation rate, and free and immobilized biomass. Results showed that the immobilized cells were able to debitter limonin-containing media and the immobilized biomass was quite stable throughout the operational conditions tested. A population of free biomass was present in the reactor, the quantity of which was dependent on dilution rate. The immobilized bacteria increased its limonin-degrading capability when the substrate concentration was increased. The aeration was not strictly necessary for limonin degradation. Additionally, the immobilized cells were active and stable for more than 2 months of continuous operation, and were able to recover their limonin-degrading capability when used intermittently. Finally, none of the main components of a juice was noticeably altered during limonin degradation, so the reactor response was good enough to consider its application.

    Notes: Summary This work presents the optimization of the chemical steps involved in nylon modification with dimethyl sulphate, polyethyleneimine, glutaraldehyde and 2-diethyl aminoethylamine to obtain a weak basic anion exchange support. Activated nylon laminated membranes were utilized for aminoacylase immobilization, allowing an ionically adsorbed enzyme derivative with high activity (0.16 U/mg E·cm2) and low removed activity (〈1%). Optimum immobilization conditions and kinetic parameters were also determined. This immobilized enzyme can be used in laminated enzyme membrane reactors.

    Notes: Abstract Grape (V. vinifera L. cv Gamay Fréaux) cells were entrapped in calcium alginate beads. The effect of bead diameter and gel concentration on the viability of the cells was checked. The reduction of 2,3,5-triphenyltetrazolium chloride as well as O2 consumption were used as viability test, and their results compared. The existence of diffusional limitations for O2 introduces an unaccuracy as high as 25% for the O2 consumption method. The reduction assay is more simple, precise and reliable.

    Notes: Summary β-Glucosidase from sweet almond was immobilized onto a nylon support and used to hydrolyze picrocrocin, the glycoside precursor of the saffron essential volatile oil, safranal. The nylon support was derivatized as hydrazide and the enzyme attached through Schiff base to bonds. The coupling efficiency was 46.8%, the immobilization yield 29.5%, and the derivative showed 24.2 and 4.0 U/mg activity for p-nitrophenylglucoside and picrocrocin, respectively, as substrates. Kinetic parameters of the immobilized derivative were determined, with picrocrocin as substrate, showing KM=7.2 mM and Vmax=4,0 U/mg. Glucose behaved as a competitive inhibitor (Ki=95.0 mM). The immobilized derivative was thermally stable up to 45°C; from that temperature onwards thermoinactivation occured. The operational deactivation showed a biphasic pattern, t1/2 being 4.2 days for the first four days of continuous operation, and 20.1 days from that point on. The immobilized enzyme lost a 50% of its initial activity after 30.7 days of storage at 4°C.

    Notes: Summary Mucor miehei lipase has been adsorbed on Celite and covalently bound to nylon. The obtained derivatives have been studied regarding their ability for synthetize several flavouring esters in biphasic aqueous/organic media. The influence of the immobilization procedure on the synthetic activity of the derivatives was considered. Solvent hydrophobicity and water content in the biphasic system influenced both enzyme stability and equilibrium displacements. In this way, solvents with log P〉3.5 and less than 1% water were optimal. It was important to consider pH effects on enzyme microenvironment when using acidic substrates. Optimum temperature and reuse of the catalyst were also checked.

    Notes: Abstract Microsomes from grapewine (V. vinifera L. cv. Camay Fréaux) cell homogenates were immobilized in calcium alginate beads. Cinnamic acid-4-hydroxylase activities for both freely suspended and immobilized microsomes were compared, resulting in a decrease of 30–40% of the initial activity as a consequence of immobilization. This decrease was probably due to both protein denaturation and microsomal mass losses during the gelation process. Nevertheless, immobilized microsomes showed an enhanced thermostability. Only 35% of the initial activity was lost when immobilized microsomes were incubated at 45°C for 10 minutes, whereas a 68% activity loss was observed when dealing with free microsomes.

    Notes: A commercial pectolytic enzyme preparation has been immobilized onto a nylon-polyethyleneimine copolymer, in order ot study the contribution of the pectin lyase activity inthe overall pectindegrading activitg. Optimal conditions for the immobilization process of the pectin lyase activity, such as chemical modification of the support, coupling pH and protein concentration, were determined.Kinetic parameters and temperature behaviour of both the soluble and immobilzied pectin lyase activitgy of the derivative were also determined. OPerational stability of the pectin lyase and overall viscosity reducring activities resulting in a halflife times of 3.8 and 8.5 days, respectively, for both pectolytic activities, when the immobilized derivatives were tested in a cross-flow reactor, using a highly esterified pedtin as substrate.
