    Notes: Abstract Limonin can be effectively degraded by Rhodococcus fascians cells. These bacteria can be entrapped in κ-carrageenan, and used in a continuous stirred tank reactor to degrade limonin in a continuous process. The effects of temperature, limonin concentration, dilution rate, and aeration on the reactor behaviour have been tested, and the results correlated with changes in limonin conversion, substrate degradation rate, and free and immobilized biomass. Results showed that the immobilized cells were able to debitter limonin-containing media and the immobilized biomass was quite stable throughout the operational conditions tested. A population of free biomass was present in the reactor, the quantity of which was dependent on dilution rate. The immobilized bacteria increased its limonin-degrading capability when the substrate concentration was increased. The aeration was not strictly necessary for limonin degradation. Additionally, the immobilized cells were active and stable for more than 2 months of continuous operation, and were able to recover their limonin-degrading capability when used intermittently. Finally, none of the main components of a juice was noticeably altered during limonin degradation, so the reactor response was good enough to consider its application.

    Notes: Abstract The use of a biological procedure for l-carnitine production as an alternative to chemical methods must be accompanied by an efficient and highly productive reaction system. Continuous l-carnitine production from crotonobetaine was studied in a cell-recycle reactor with Escherichia coli O44 K74 as biocatalyst. This bioreactor, running under the optimum medium composition (25 mM fumarate, 5 g/l peptone), was able to reach a high cell density (26 g dry weight/l) and therefore to obtain high productivity values (6.2 g l-carnitine l−1 h−1). This process showed its feasibility for industrial l-carnitine production. In addition, resting cells maintained in continuous operation, with crotonobetaine as the only medium component, kept their biocatalytic capacity for 4 days, but the biotransformation capacity decreased progressively when this particular method of cultivation was used.

    Notes: Summary β-Glucosidase from sweet almond was immobilized onto a nylon support and used to hydrolyze picrocrocin, the glycoside precursor of the saffron essential volatile oil, safranal. The nylon support was derivatized as hydrazide and the enzyme attached through Schiff base to bonds. The coupling efficiency was 46.8%, the immobilization yield 29.5%, and the derivative showed 24.2 and 4.0 U/mg activity for p-nitrophenylglucoside and picrocrocin, respectively, as substrates. Kinetic parameters of the immobilized derivative were determined, with picrocrocin as substrate, showing KM=7.2 mM and Vmax=4,0 U/mg. Glucose behaved as a competitive inhibitor (Ki=95.0 mM). The immobilized derivative was thermally stable up to 45°C; from that temperature onwards thermoinactivation occured. The operational deactivation showed a biphasic pattern, t1/2 being 4.2 days for the first four days of continuous operation, and 20.1 days from that point on. The immobilized enzyme lost a 50% of its initial activity after 30.7 days of storage at 4°C.

    Notes: Summary The ability of Rhodococcus fascians cells to degrade limonin and limonin species (limonoate, limonoate-D-ring lactone and limonoate-A-ring lactone) was checked against pH. These studies showed a marked effect of pH on cell growth mainly due to substrate availability (limonin species). Evolution of limonin and its species within the medium were followed at different pH values. The best substrate for Rhodococcus fascians at pH 7.0 was limonoate whereas at pH 4.0 to 5.5 it appeared to be limonin. Results suggest that the citrus juice debittering process start only once the natural precursor of limonin (limonoate A ring lactone) has been transformed into limonin, the equilibrium displacement being governed by the citrus juice pH.

    Description / Table of Contents: Resume On a entrepris une recherche expérimentale afin de résoudre le problème de la minéralisation des fibres végétales dans les mortiers de ciment Portland. L’étude se divise en deux parties. Elle traite d’abord des questions suivantes: obturation des pores, résistances mécaniques et réduction alcaline; ensuite, de l’imprégnation des fibres avec une courte description des mélanges utilisés et du vieillissement des mortiers, avec ou sans adjuvants, renforcés de fibres de sisal imprégnées ou non dans différentes conditions. Les produits utilisés dans cette solution sont principalement des composés dérivés du bois, insolubles dans l’eau et non extractibles par l’eau ou la vapeur d’eau. Les résultats de cette recherche ouvrent des perspectives.
    Notes: Abstract Research has been carried out with the aim of solving the problem of vegetable fibre mineralization in Portland cement mortars. It consists of two parts. The first part deals with pore sealing, mechanical strength and alkaline reduction; the second with fibre impregnation including a short description of the compounds used and the ageing of mortars, with or without admixtures, which are reinforced with sisal fibres impregnated or unimpregnated under different conditions. Products used in this solution are mainly compounds derived from timber, insoluble in water and unextractable by water or water vapour. The results of the research look promising.
