    Notes: Summary 1. Adenosine analogues inhibit calcium-dependent K+-evoked release of [3H]norepinephrine from guinea pig cerebral cortical and hippocampal vesicular preparations. Inhibition requires high concentrations (100µM) of the adenosine analogues and is abolished in the presence of high concentrations (2 mM) of calcium ions. The inhibitory effect of 2-chloroadenosine is blocked by theophylline. The structure activity profile (N 6-d-phenylisopropyladenosine ≥N 6-l-phenylisopropyladenosine ≥ 2-chloroadenosine 〉N 6-cyclohexyladenosine, adenosine 5′-cyclopropylcar-boxamide) is not that expected of either A1 (high-affinity) or A2 (low-affinity) adenosine receptors. 2. Calcium-dependent K+-evoked release of [3H]dopamine from guinea pig striatal vesicular preparations is inhibited by apomorphine. However, only 2-chloroadenoine causes an inhibition of K+-evoked release of [3H]dopamine. Other adenosine analogues such asd- andl-phenylisopropyladenosine and adenosine 5′-cyclopropylcar-boxamide cause a facilitation of K+-evoked release. The facilitation is abolished or reduced in the presence of high concentrations (2 mM) of calcium ions. The sites of action of adenosine analogues do not appear to have structural requirements identical to those expected of A1 (high-affinity) or A2 (low-affinity) adenosine receptors. 3. The results indicate that adenosine analogues can have either inhibitory or facilitory effects on K+-evoked release of catecholamines from central synaptic terminals.

The calcium-dependent K+-evoked release of [3H]norepinephrine from guinea pig cerebral cortical vesicular preparations is inhibited by norepinephrine, clonidine, and epinephrine. Isoproterenol has no effect and phentolamine prevents the inhibition by norepinephrine. The results indicate that anα-adrenergic receptor mediates an inhibitory input to the calcium-dependent release process. The inhibition by norepinephrine is prevented by high concentrations (3.0 mM) of calcium ions. 2. A cyclic AMP phosphodiesterase inhibitor, ZK 62771, slightly elevates [3H]cyclic AMP levels in the guinea pig cerebral cortical preparation and potentiates the marked elevation of [3H]cyclic AMP elicited by the adenylate cyclase activator, forskolin. 3. Neither ZK 62771 nor forskolin alone has significant effects on K+-evoked release of [3H]norepinephrine from the cerebral cortical vesicular preparation; however, a combination of ZK 62771 and forskolin inhibits K+-evoked release by as much as 60%. The inhibition is reversed by high concentrations (2.0 mM) of calcium ions. The results suggest that a marked accumulation of cyclic AMP elicited via both activation of adenylate cyclase and inhibition of phosphodiesterase can be inhibitory to neurotransmitter release from central synaptic terminals.

    Notes: Summary 1. The calcium antagonists D-600 (1–10µM) and diltiazem (10–25µM) inhibit K+-evoked release of [3H]norepinephrine from guinea pig cerebral cortical vesicular preparations. The inhibition of release is partially reversed by increasing concentrations of calcium to 2 mM. Diltiazem at 100µM has no effect on K+-evoked release of [3H]norepinephrine at 0.15 mM calcium but does inhibit release at 2.0 mM calcium. 2. The calcium antagonist nifedipine and dantrolene, an agent purported to antagonize release of calcium from intracellular storage sites, have no effect on K+-evoked release of [3H]norepinephrine. 3. The calcium antagonists D-600 (1µM) and diltiazem (10µM) inhibit K+-evoked release of [3H]dopamine from guinea pig striatal vesicular preparations. Higher concentrations of drug, namely, 10µM for D-600 and 100µM for diltiazem, cause a potentiation rather than an inhibition of K+-evoked release. The potentiation is reduced in magnitude upon raising the extracellular calcium to 2.0 mM. Indeed, 10µM D-600 then inhibits K+-evoked release of [3H]dopamine. 4. The results indicate that putative calcium antagonists can have both inhibitory and facilitory effects on calcium-dependent K+-evoked release of catecholamines from central synaptic endings. Furthermore, certain peripheral calcium antagonists such as nifedipine and dantrolene may prove ineffective in central systems.

    Notes: Mixtures of both synthetic and naturally occurring (human plasma) cholesteryl esters have been examined by capillary gas chromatography/mass spectrometry (GC/MS). A magnetic sector mass spectrometer was used and a variety of ionization modes were assessed with a view to obtaining structural information on intact cholesteryl esters. By employing ammonia as reagent gas, with negative ion scanning, spectra were produced from which the nature of steryl and fatty acyl moieties could be readily deduced. Analyses were performed at an ion source temperature of 300°C in order to maintain the integrity of the gas chromatographic profile. The technique described is of general use for the GC/MS analysis of steryl esters, particularly in conjunction with magnetic sector instruments.

    Notes: An azurin, a small respiratory copper protein from the bacterium Pseudomonas fluorescens biotype G, has been studied by mass spectrometry to determine sequence information. The study of homologously related proteins by mass spectiometry is particularly attractive, since the correct nature of major parts of the deduced sequences can be confirmed by comparison with the sequences of the protein from related organisms. An oxidized tryptohan residue has been identified amongst the products from a cyanogen bromide digest of this wild type azurin. In the same digest, a product is also found to arise from cleavage of the peptide chain at the C-terminal side of the same tryptophan residue. These results are rationalized.

    Notes: High-performance liquid chromatography/mass spectrometry (HPLC/MS) with thermospray ionization has been applied to the analysis of free ecdysteroids (polyhydroxylated steroids that serve as arthropod moulting hormones). Of a range of analytical conditions that were tested, optimum results were obtained by the use of re versed-phase HPLC using a methanol + water mixture containing 0.1 M ammonium acetate. The positive-ion spectra of ecdysteroids showed pseudo-molecular ions, [M + H]+, and fragment ions corresponding to the sequential losses of hydroxyl groups. The [M + H - H2O]+ and [M + H - H2O]+ ions were the most abundant for all the compounds tested. Ecdysteroids bearing a C-20 hydroxyl group (e.g. 20-hydroxyecdysone) showed ions characteristic of cleavage of the side-chain from the steroid nucleus. This mode of fragmentation was much less obvious in the case of ecdysteroids lacking C-20 hydroxylation (e.g. ecdysone). Full-scan mass spectra were readily obtained from 1 μg of free ecdysteroids, while a limit of detection of 5 ng (signal-to-noise ratio 3) was attained in the selected-ion monitoring mode. The application of HPLC/MS to the study of ecdysteroids in biological materials is demonstrated by the analysis of extracts of newly laid eggs of the desert locust (Schistocerca gregaria) and pupae of the cotton leafworm (Spodoptera litoralis).

    Notes: Nucleic acid extracts from 1400-year-old radish seeds recovered from excavations at Qasr Ibrim, Upper Egypt, have been analysed by gas chromatography/mass spectrometry and gas chromatography/mass spectrometry/mass spectrometry. tert-Butyldimethyisilyl derivatives of the purine and pyrimidine bases were prepared, after treatment of the nucleic acid extracts with concentrated formic acid. Under electron ionization these derivatives yield prominent [M-57]+ ions that were found to be of value for use in analyses employing selected-ion monitoring and product-ion studies. These two techniques were used in the sensitive and selective detection of nucieotide bases in crude extracts of ancient seeds. The results obtained were supported by complementary analyses of hydrolysates of ancient nucleic acids and authentic bases by means of retention time and spectral comparisons. This work demonstrates for the first time that mass spectrometry can be used in the direct chemical examination of nucleotide bases in ancient materials. This analytical approach is currently being used to address questions regarding the possible chemical (diagenetic) changes occurring in the nucleic acids of ancient biological material.

    Notes: The electronic effect of substituting CH with N in poly(pyrazolyl)-borato ligands and their transition-metal complexes is shown to be a decrease in energy of the filled metal and ligand orbitals. This conclusion is based on the cyclovoltammograms and photoelectron spectra of bis(hydrotris(azolyl)borato)-iron(II) and -cobalt(II) complexes (azolyl = pyrazolyl and 1,2,4-triazolyl) and on MO calculations. 57Fe Mössbauer spectra of bis(hydrotris(1,2,4-triazolyl)-borato)iron(II) show that there is a fine-tuning of the HOMO-LUMO gap by a shift in transition temperature for the spin equilibrium. 15N NMR spectroscopy supports the assignment of a higher negative charge to the exodentate N-4 nitrogen than to the endodentate N-2 or N-3 positions in the poly(azolyl)borato anions, where azolyl is 1,2,4-triazolyl or tetrazolyl. The electron withdrawing effect of the additional nitrogen atoms and the in-corporation of water of crystallization by O-H ⃛ N bonding both assist in the formation of (azolyl)C-H ⃛ O bonds.

    Notes: Metastable ions arising from pseudomolecular [M — H]- ions produced by fast-atom bombardment negative-ion mass spectrometry of a range of free ecdysteroids, ecdysteroid conjugates and polar metabolites were investigated by means of linked scanning at constant B/E. Free ecdysteroids displayed daughter-ion spectra which allow 20-hydroxyecdysteroids and ecdysteroids lacking C-20 hydroxylation to be readily distinguished. The ejection of acetic acid from acetylated ecdysteroids was also readily detectable. Characteristic metastable-ion decomposition of ecdysteroid acids was not observed, presumably as a result of charge localization. High-mass daughter ions were also lacking in the case of phosphate conjugates.
