    Notes: The solar-wind driven magnetosphere–ionosphere system is a classic example of a complex dynamical system (CDS). The defining properties of a CDS are (1) sensitivity to initial conditions; (2) multiple space-time scales; (3) bifurcation sequences with hysteresis in transitions between attractors; and (4) noncompositionality. Noncompositionality means that the behavior of the system as a whole is different from the dynamics of its subcomponents taken with passive or no couplings. In particular the dynamics of the geomagnetic tail plasma depends on its coupling to the dissipative ionospheric plasma and on the nature of the solar-wind driving electric field over a suitably long (many hours) previous time interval. These complex dynamical system features are shown here in detail using the known WINDMI model for the solar-wind driven magnetosphere–ionosphere (MI) system. Numerous features in the bifurcation sequence are identified with known substorm and storm characteristics. © 1999 American Institute of Physics.

    Notes: Two prototype Langmuir probes (based on a JET design) have been installed near the outer strike point on the divertor floor in the DIII-D tokamak. Results from these graphite probes were used to design a full array of Langmuir probes for the DIII-D divertor and an additional set of probes for the bias ring of the Advanced Divertor program. The design allows for the projected area of the probes to be relatively insensitive to the magnetic field angle in the divertor region, while projecting the intercepted heat flux over a much larger area. The prototype probes have survived the full separatrix plasma flux during both L- and H-mode operation. I/V characteristics during high power neutral beam operation exhibit electron temperatures approaching 50 eV and large negative floating potentials near the outer separatrix strike point. Edge localized modes cause an increase in the ion flux as large as a factor of 10 above the H-mode level. As a result, a new set of faster, higher voltage electronics will be used for the full array. The electronics will be capable of floating either with the divertor floor or with the bias ring.

    Notes: Abstract— Young rat cerebral-cortex slices were incubated with 32Pi in the absence and presence of ACh plus eserine, norepinephrine, dopamine or serotonin for 1 h. their cellular and subcellular fractions were isolated, and the specific radioactivities of the various phospholipids determined.In the neuronal- and astroglial-enriched fractions ACh plus eserine increased the 32P-labelling of phosphatidyl inositol (PhI) phosphatidic acid (PhA) and phosphatidylcholine (PhC) by increments which ranged from 108 per cent for PhI to 30 per cent for PhC and in the presence of norepinephrine or dopamine these increments ranged from 180 per cent for PhI to 29 per cent for PhC.In the subcellular fractions ACh plus eserine exerted maximal stimulatory effect on the labelling of the synaptosomal phospholipids, which was 88 per cent for PhI and 79 per cent for PhA, followed by those of microsomes, mitochondria and nuclei.ACh plus eserine exerted no effect on [l4C]glucose incorporation, but inhibited the incorporation of [14C]glycerol into phospholipids by amounts which ranged from 30 per cent for PhI to 3 per cent for PhE.Although the rate of incorporation of 32Pi into phospholipids of 0.2 mm slices was higher than that of the 0.5 mm slices the stimulatory effect of ACh plus eserine on the 32Pi incorporation into the lipids of the latter was higher.When neuronal- and astroglial enriched fractions were first isolated from the cerebra then incubated with 32Pi or [14C]choline, labelling of phospholipids in the neuronal fraction was higher than that of the astroglial fraction; however, ACh plus eserine had no effect on the incorporation of 32Pi into the lipids of either fraction.ACh plus eserine stimulated the activity of phosphatidic acid phosphatase in the various subcellular fractions by increments which ranged from 13 per cent in nuclei to 37 per cent in microsomes.It was concluded that the nonspecific localization of the neurotransmitter effect could be due to the widespread distribution of the enzymes which appear to be responsive to cholinergic and adrenergic neurotransmitters.

    Notes: Abstract— Synthesis of phosphatidylcholine, phosphatidylinositol and palmityl carnitine in synaptosomes isolated from rat brain was investigated and compared with the synthesis of these compounds in microsomes and mitochondria. Electron microscopic and marker enzyme studies showed the contaminants in the synaptosomal preparation to consist of a few microsomes and almost no free mitochondria. In synaptosomes, addition of 1,2-diglyceride exerted no effect on the incorporation of [14C]choline into phosphatidylcholine or on the incorporation of [3H]myo-inositol into phosphatidylinositol, but it stimulated the incorporation of CDP[1,2-14C]choline into phosphatidylcholine by more than 50 per cent. The incorporation of the latter in intact synaptosomes, lysed synaptosomes and purified mitochondria was 15-6, 27 and 9-9 per cent, respectively, of that in the microsomes. The incorporation of [3H]myo-inositol into the phosphatidylinositol of synaptosomes and purified mitochondria was 15-8 and 11-1 per cent, respectively, of that in the microsomes. Maximal incorporation of [3H]myo-inositol occurred at pH 7–5 in a medium containing Mg2+ and CTP; it was linear with time and protein concentration and was inhibited by 1 mM Ca2 + but unaffected by the presence of ATP. This incorporation of myo-inositol appeared to occur through the reversal of the CDP-diglyceride: inositol transferase reaction. The demonstration of carnitine palmityl transferase in synaptosomes indicated that, as in mitochondrial and erythrocyte membranes, fatty acids can be transported across the synaptosomal membrane. In contrast to mitochondria where maximal incorporation of [14C]carnitine into palmityl carnitine was observed after 20 min of incubation, the incorporation in synaptosomes increased as a function of time up to 60 min of incubation.We conclude that synaptosomes can carry on de novo synthesis of lipids, although at a limited rate. From the present data we cannot state with certainty how much of this synthesis is attributable to membranes originating from the endoplasmic reticulum.

    Notes: Abstract— Paired iris smooth muscles from rabbits were prelabelled either in vitro by incubation for 30 min at 37°C in an iso-osmotic salt medium containing glucose, inositol, cytidine and 32Pi, or in vivo by administration of the isotope intracamerally into each eye 1 h before death. One of the pair was then incubated at 37°C for 10 min in an unlabelled medium containing 10 mm of 2-deoxyglucose and the other was incubated in the presence of norepinephrine (NE) or other adrenergic agents. Triphosphoinositide (TPI) was found to contain more 32P than any other phospholipid (almost 39% of total lipid radioactivity) in both the in vitro and in vivo experiments. NE (50 μm) increased the loss of 32P from TPI (the TPI effect') by 28–30% in the 32P-labelled muscle. The TPI effect was accompanied by a significant increase in 32P labelling of phosphatidic acid (PA) and phosphatidylinositol (PI) but not phosphatidylcoholine. In this tissue the TPI effect was found to be mediated through α-adrenergic receptors. At 14 days after surgical sympathetic denervation, incorporation of 32P into phospholipids of the denervated muscle increased by an average of 6% over that of the normal muscle. The increase in TPI, PI and PA was 7%, 4% and 9% of that of the control respectively. There was little change in phospholipid content of the denervated muscle. The increase in sensitivity to NE (12.5 μm) caused by denervation produced about 18% increase in the TPI effect and a 25% increase in the 32P labelling of PA, but not PI. In view of our previous findings on the requirement of the TPI effect for Ca2+, this observation could suggest that an increase in Ca2+ influx, following the interaction between the neurotransmitter and its receptor could stimulate TPI-phosphodiesterase, thus leading to increased PA via increased diglyceride. This denervation-induced supersensitivity to NE appears to be postsynaptic in nature. 32Pi was injected intracamerally into each eye 1 h before electrical stimulation of one of the sympathetic trunks. After stimulation for 30 min there was a significant loss of 32P from TPI and a significant increase in the labelling of PI and PA of the stimulated muscle. It is concluded that TPI and its enzymes could play an important role in neurotransmission at the neuromuscular junction of smooth muscle.

    Notes: English style crumpets were packaged in the atmosphere of CO2 and N2 (3:2) and stored at 20°, 22°, 24° and 30°C. In all cases, package volume and headspace CO2 concentration decreased during the first week of storage. At 30°C, the volume started to increase after 12 days due to production of CO2 and other metabolites by microorganisms. At 24°C, the volume did not start to increase until after 25 days, while at 22° and 20°C it remained stable or gradually decreasing. Aerobic plate counts and metabolites were higher in the product stored at 30°C for 19 days than those stored at 20°C for 1 month. Product pH after 1 month at 20°C was lower (5.9) than that at 30°C for 19 days (6.5) due, essentially, to the absorption of CO2.
