    Notes: A novel method for determining magnetic anisotropies from hysteresis loops is presented. While sweeping the loop, a magnetic field is applied perpendicularly to the sweep-field axis. This causes the magnetization to rotate reversibly in a wide field range and still reach saturation at finite fields. An example is given whereby surface and volume anisotropies are determined from magneto-optical Kerr effect loops in Co films grown on stepped Cu(001). © 1997 American Institute of Physics.

    Notes: Several genetic maps for sugar beet (Beta vulgaris L.), from different German research groups, have been published and it is now possible to consider combining them with the aid of the common markers. The computer program JOINMAP (versions 1.3 and 2.0) was used for pair-wise combination of three populations. Several problems arose: the genetic background of the populations, different population structures (F2 versus F1× F1), different number of polymorphic loci for common probes in the populations to be combined, different estimates of the recombination rates between the same markers and differences between the JoinMap versions. The maps from two F2 populations could be integrated into a single map, but it was more appropriate to construct separate maps for the F2 populations and the F1× F1 population using common markers as reference points only.

    Notes: Abstract  We investigated the effects of nicotine and its derivate nicotine di-d-tartrate on primary cultured human nasal epithelial cells. Both substances evoked an in-crease in the intracellular free calcium concentration. In the presence of extracellular Ca2+ the cytosolic Ca2+ ([Ca2+]i) increase was long lasting, whereas in the absence of external Ca2+ there was a transient increase of [Ca2+]i indicating that nicotine has an influence on Ca2+ conductances across the membranes and on intracellular Ca2+ stores. Both effects could be blocked by the nicotinic receptor antagonist methyllycaconitine (MLA). Apical or basolateral application of nicotine during transepithelial transport measurements with confluent monolayers of cultured human nasal cells resulted in a significant, reversible decrease of amiloride-sensitive sodium absorption with an apparent half-maximal blocker concentration of about 950 μM. To exclude the possibility that remnant neuronal components were responsible for the observed effects we used tetrodotoxin and verapamil to block putative neuronal channels and 4-(4-diethyla- mino)styryl-N-methylpyridinium iodide (4-di-2-Asp) to stain neuronal tissue. Both experimental approaches demonstrated that there were no neuronal-mediated effects. These results indicate the direct effects of nicotine on human nasal epithelium, giving the first evidence of the existence of nicotinic receptors in non-excitable cells.

    Notes: Abstract  Transepithelial short-circuit current (I SC), potential (V T) and resistance (R T) of confluent monolayers of human nasal epithelium cultured from patients with and without cystic fibrosis (CF) were measured. In our Ussing chamber experiments with monolayers derived from non-CF and CF patients neither I SC (non-CF: 14.1 ± 1.0 μA/cm2, n = 77; CF: 16.7 ± 1.5 μA/cm2, n = 42), nor R T (non-CF: 288 ± 15 Ω· cm2; CF: 325 ± 20 Ω· cm2) showed any significant differences, only V T showed moderate but significant different values (non-CF: –3.6 ± 0.4 mV; CF: –5.6 ± 0.7 mV, respectively). Total I SC in CF cells was nearly completely inhibited by amiloride (92 ± 9.6%), while in non-CF tissue amiloride-insensitive conductances mediated a considerable amount of the I SC (36.3 ± 6.1%), indicating a lower activity of amiloride-sensitive Na+ conductances in non-CF cells. In both tissues the amiloride-sensitive I SC could also be blocked by the amiloride analogues benzamil, phenamil and 5-(N-ethyl-N-isopropyl)2’,4’-amiloride (EIPA) with different affinities. However, amiloride had a significant lower affinity in CF tissue (half-maximal blocker concentration, K 1/2 = 586 ± 59 nM) compared with non-CF tissue (K 1/2 = 294 ± 22 nM). Astonishingly, phenamil, a blocker which irreversibly blocks all epithelial Na+ channels hitherto described, inhibited the Na+ conductances of human nasal epithelium in a completely reversible way, but nevertheless with high affinity (non-CF: K 1/2 = 12.5 ± 1.2 nM; CF: K 1/2 = 17.1 ± 1.1 nM). Even in high doses none of these blockers had any effect on intracellular Ca2+ concentration as measured with Fura-2. From these findings, we conclude that the epithelial Na+ conductances of human CF nasal epithelium show modified regulation or are functionally different from those of other tissues.

    Notes: Abstract. Removal of extracellular divalent cations activated a Cl− channel in the plasma membrane of Xenopus laevis oocytes. This so-called Ca2+-inactivated Cl− channel (CaIC) was present in every oocyte and was investigated using two-electrode whole-cell voltage clamp and single-channel patch-clamp techniques. Beside other Cl− channel inhibitors, anthracene-9-carboxylic acid (9-AC) and 3′azido-3′deoxythymidine (AZT), a nucleoside analogue commonly used as an antiviral drug, blocked at least partly the CalC-mediated currents. Using the Cl−-sensitive dye 6-methoxy-N-(sulfopropyl)quinolinium (SPQ) we could visualize the transport of Cl− from the oocyte cytoplasm to the surrounding medium after activation of the CaIC by Ca2+ removal. In the absence of external Cl− and Ca2+, the emission intensity of SPQ declined continuously, indicating a quenching of fluorescence by the efflux of Cl− in the millimolar range. In the presence of external Ca2+, no emission changes could be observed during the same time period. Chelating external Ca2+ in absence of Cl− immediately activated Ca2+-inactivated Cl− channels leading to subsequent emission decrease of SPQ. Investigations on the selectivity of the CaIC revealed only poor discrimination between different anions. With single-channel measurements, we found an anion selectivity sequence I− 〉 Br− 〉 Cl−≫ gluconate as it is also typical for maxi Cl− channels. Contrary to the majority of all other transport systems of the Xenopus oocyte, which show reduced activity due to membrane depolarization or endocytotic removal of the transport protein from the plasma membrane during oocyte maturation, the CaIC remained active in maturated oocytes. Single-channel measurements on maturated oocytes, also known as eggs, showed the presence of Ca2+-inactivated Cl− channels. However, this egg CaIC revealed an altered sensitivity to external Ca2+ concentrations. All these data confirm and extend our previous observations on the CaIC and give clear evidence that this channel is peculiar among all Cl− channels described up to now.

    Notes: Abstract. The cloned intestinal peptide transporter is capable of electrogenic H+-coupled cotransport of neutral di- and tripeptides and selected peptide mimetics. Since the mechanism by which PepT1 transports substrates that carry a net negative or positive charge at neutral pH is poorly understood, we determined in Xenopus oocytes expressing PepT1 the characteristics of transport of differently charged glycylpeptides. Transport function of PepT1 was assessed by flux studies employing a radiolabeled dipeptide and by the two-electrode voltage-clamp-technique. Our studies show, that the transporter is capable of translocating all substrates by an electrogenic process that follows Michaelis Menten kinetics. Whereas the apparent K0.5 value of a zwitterionic substrate is only moderately affected by alterations in pH or membrane potential, K0.5 values of charged substrates are strongly dependent on both, pH and membrane potential. Whereas the affinity of the anionic dipeptide increased dramatically by lowering the pH, a cationic substrate shows only a weak affinity for PepT1 at all pH values (5.5–8.0). The driving force for uptake is provided mainly by the inside negative transmembrane electrical potential. In addition, affinity for proton interaction with PepT1 was found to depend on membrane potential and proton binding subsequently affects the substrate affinity. Furthermore, our studies suggest, that uptake of the zwitterionic form of a charged substrate contributes to overall transport and that consequently the stoichiometry of the flux-coupling ratios for peptide: H+/H3O+ cotransport may vary depending on pH.
