    Notes: Tetraploid clones of potato with a superior efficiency in producing androgenetic plants (4 × EAPP-clones) have been obtained by culturing in vitro anthers of 2 × EAPP-clones. The latter were isolated by three cycles of recurrent selection from diploid breeding material (UHRIG 1985 a). In this paper we report on the capacity of 4 × EAPP-clones, when crossed to in vitro unresponsive 4 × genotypes, to transmit to their F1 their androgenetic potential, Five 4 ×× 4 × F1 crosses were considered, which produced on average 35 embryoids per flower – a value higher than that of nonresponsive 4 × genotypes (no embryoids obtained), but-also significantly better than the value found for 4× EAPP-clones (9.8 embryoids per flower). The hybrid families behaved differently from each other, with one producing up to 63 embryoids per flower. A range of per plant values was, moreover, found, revealing the existence of a large variability among sister plants belonging to the same F1 cross. The presented data indicate a rather simple inheritance of dominant genetic factors acting in favor ot androgenesis. They also suggest that the utilized 4× EAPP-clones were possibly heterozygous for such genes.Data are also reported on the ploidy level of anther plants obtained from 4× and 2× EAPP-clones. In this respect 2× EAPP-clones show the interesting capacity of generating, via anther culture, a consistent fraction of tetraploid plants (13,7 %).Based on the findings reported in this paper we propose, for tetraploid S. tuherosum L., a cyclic breeding procedure making use of anther culture and where ploidy level alternates, within a cycle, between 2× and 4×.

    Notes: The objectives of this study were (1) to ascertain the genetic differences for cold-tolerance traits in a series of diallel crosses among 12 maize populations grown in field experiments, and (2) to compare, in controlled environment rooms, the modification of various metabolic parameters of two sets of four F1 variety crosses which, according to the field experiments, differed largely in seedling early vigour (cold-tolerant [Ct] vs cold-sensitive [Cs] Fl sets). Under field conditions, cold-tolerance adaptability, as monitored by shoot dry matter accumulation, appeared genetically controlled; moreover, considerable differences existed among genotypes. Field data suggested that shoot dry weight was a good indicator of plant adaptability to periods of unfavorable cool conditions. The growth-chamber experiments showed that shoot dry weights of the two sets of hybrids (Ct vs Cs), were more uniform at the highest temperature regimes than under suboptimal temperatures. Moreover, the laboratory study indicated that the interval of time of attain a specific stage of development provided a meaningful and useful criterion for differentiating the capacity of maize plants to grow at low temperatures. Although a clear association between a particular metabolite and the capacity of plants to grow at low temperatures was not observed, it was of interest to note that the total N concentration in the shoot was significantly higher in the Ct plants compared to the Cs ones.

    Notes: Amplified fragment length polymorphism (AFLP) and microsatellite (simple sequence repeat, SSR) techniques were used to map the _RGSpeking gene, which is resistant to most isolates of Cercospora sojina in the soya bean cultivar ‘Peking’. The mapping was conducted using a defined F2 population derived from the cross of ‘Peking’(resistant) בLee’(susceptible). Of 64 EcoRI and MseI primer combinations, 30 produced polymorphisms between the two parents. The F2 population, consisting of 116 individuals, was screened with the 30 AFLP primer pairs and three mapped SSR markers to detect markers possibly linked to RcsPeking. One AFLP marker amplified by primer pair E-AAC/M-CTA and one SSR marker Satt244 were identified to be linked to ResPeking. The gene was located within a 2.1-cM interval between markers AACCTA178 and Satt244, 1.1 cM from Satt244 and 1.0 cM from AACCTA178. Since the SSR markers Satt244 and Satt431 have been mapped to molecular linkage group (LG) J of soya bean, the ResPeking resistance gene was putatively located on the LG J. This will provide soya bean breeders an opportunity to use these markers for marker-assisted selection for frogeye leaf spot resistance in soya bean.

    Notes: Several genetic maps for sugar beet (Beta vulgaris L.), from different German research groups, have been published and it is now possible to consider combining them with the aid of the common markers. The computer program JOINMAP (versions 1.3 and 2.0) was used for pair-wise combination of three populations. Several problems arose: the genetic background of the populations, different population structures (F2 versus F1× F1), different number of polymorphic loci for common probes in the populations to be combined, different estimates of the recombination rates between the same markers and differences between the JoinMap versions. The maps from two F2 populations could be integrated into a single map, but it was more appropriate to construct separate maps for the F2 populations and the F1× F1 population using common markers as reference points only.

    Notes: A description is given of the major modification induced in the pattern of photosynthates accumulation by the delayed senescence behaviour of the maize inbred line Lo87602. The major effects noted are: (1) high water and chlorophyll contents in leaves at physiological maturity, (2) high sucrose content in the stalk during grain filling, (3) busks and cobs with more water, sucrose and proteins, (4) high protein content in the grain. The high level of sucrose in the stalk at physiological maturity protects the plant from stalk rotting organisms.The delayed senescence phenotype depends on a major dominant gene, the same genetic factor which favours the accumulation of a high level of sucrose in the stalk.

    Notes: The germplasm of the potato (S. tuberosum ssp. tuberosum) has been modified since the beginning of this century by breeders who introgressed important agronomic traits, for example disease resistance genes, from several wild and cultivated Solanum species of the Americas. In this paper we show that the Rflp analysis of potato-breeding material can be used to detect chromosomal regions descended from more-distantly related Solanum species. The Rflp patterns of individuals of ten Solanum species, ten breeding lines of S. tuberosum ssp. tuberosum and the cultivated variety ‘Bintje’ were analyzed. Rflp data for each of eight single loci of known genomic position were used for the computation of locus-specific phenograms by distance matrix methods. Several of the potato-breeding lines deviated clearly from the clustered species S. tuberosum, S. stenotomum and S. canasense for one or more of the loci considered. These deviations indicated the presence of “exotic” germplasm at a particular locus. The possibility of detecting such germplasm has implications for mapping the agronomic traits for which the wild species were introgressed and which may still be linked to “foreign” chromosome fragments.

    Notes: Although desiccation tolerant, the resurrection plant Craterostigma plantagineum is sensitive to relatively low levels of sodium chloride. Exposure to sodium chloride, but not dehydration, led to accumulation of sodium ions in leaves and roots and caused irreversible wilting. The effects of salt and dehydration on transcript accumulation patterns were studied by using selected cDNA clones that were related to water stress. Most of the clones represented genes that were up-regulated in response to both treatments. Among the transcripts specifically up-regulated by dehydration were RNAs encoding transcripts with homology to aquaporins. Expression analysis revealed dehydration-specific profiles of late embryogenesis abundant (LEA) genes, which differed from the patterns observed for the same genes under sodium chloride stress. The interconversion of octulose and sucrose, which is characteristic for the desiccation/rehydration cycle in C. plantagineum leaves, was not activated by sodium chloride. The present results suggest that dehydration-specific responses involve the synchronized expression of specific genes and the presence of a determined concentration of sucrose. These dehydration responses were not detected in response to sodium chloride treatment.
