    Description / Table of Contents: Summary Background: The dynamic radionuclide nephrography (Tc99m-MAG3) is an important method in the evaluation of obstruction of the upper urinary tract. The aim of the present study was to assess whether radionuclid nephrography under standardized conditions with adequate hydration and Furosemid induced diuresis can reliably detect a significant obstruction at the pelvi-ureteric junction. We therefore tested whether minimal tracer washout (10–25%) in the presence of normal renal function could justity a non-operative managment. Methods: 55 children with sonographically diagnosed hydronephrosis (dilation of the renal pelvis of more than 12 mm and calical dilation) had dynamic radionuclide nephrography after intravenous hydratisation (30–40 ml/kg bw). Results: 24 children showed a washout above 50%, 13 of more than 25%. Three patients had only a minimal washout (10–25%), which improved impressively as demonstrated in a control nephrography 2 months later in all of them. 15 children without any significant washout underwent corrective surgery: Six months after operation 13 of them showed a washout above 50%, the two remorning or more than 30%. Conclusion: Standardized radionuclide nephrography allows an important functional assessment of the urodynamic relevance of the pelvi-ureteric junction obstruction. With minimal tracer washout (10–25%) in the presence of normal renal function (〉40% divided renal function) primary conservative management seems justified. A washout below 50% no longer can be accepted as on indication of urodynamicly important obstruction necessitating operative corrective surgery.
    Notes: Zusammenfassung Fragestellung: Die dynamische Radionuklidnephrographie (99mTc-MAG3) stellt die wichtigste Untersuchungsmethode zur Klärung einer oberen Harnwegobstruktion dar. Untersucht wurde, ob die Methode, unter standardisierten Bedingungen (i.v.-Hydratisierung, Furosemidgabe und Sedierung) angewandt, eine strenge Operationsindikation erlaubt und sich als Verlaufsbeobachtung eignet. Dabei wurde insbesondere geprüft, ob bei minimalem Abfall der Traceraktivität (10–25%) und guter Nierenfunktion ein konservatives Vorgehen möglich ist. Methode: Bei 55 Kindern mit ausgeprägter Hydronephrose (〉12 mm Dilatation des Nierenbeckens und der Kelche in der Sonographie) wurde eine Radionuklidnephrographie veranlaßt. Die Untersuchung erfolgte nach i.v. 4stündiger Hydratisierung mit 30–40 ml/kg KG. Ergebnisse: Bei 24 Kindern war ein Abfluß von mehr als 50% und bei 13 von mehr als 25% der Aktivität nachweisbar. Bei 3 Patienten war nur eine geringe Ausscheidung (10–25%) zu dokumentieren, die sich bei einer Kontrollradionuklidnephrographie nach 8 Wochen bei allen Patienten eindrucksvoll besserte. Von 15 Kindern mit fehlendem Tracerabfall wurden 6 Monate nach erfolgreicher Korrektur der Ureterabgangsstenose bei 13 Kindern mehr als 50% der Traceraktivität ausgeschieden, bei den beiden anderen mehr als 30%. Schlußfolgerungen: Die standardisierte Radionuklidnephrographie ermöglicht eine wichtige funktionelle Aussage über die urodynamische Relevanz der Harntransportstörung. Ein geringer Auswascheffekt (10–25%) erlaubt bei Säuglingen mit normaler Nierenfunktion primär ein konservatives Vorgehen, und ein Tracerabfall 〈50% darf nicht länger als Maß für eine urodynamisch bedeutsame Obstruktion mit der Konsequenz einer operativen Korrektur angesehen werden.

    Notes: Abstract  Subsidence due to longwall underground coal mining changes the hydraulic properties, heads, yields, and in some cases the groundwater chemistry of overlying bedrock aquifers. A 7-year study of a sandstone aquifer overlying an active longwall mine in Illinois has supported a comprehensive model of these impacts. Subsidence caused increases in permeability and storativity over the longwall panel. These changes initially caused a major decline in water levels in the sandstone, but the aquifer recovered slightly within a few months and fully within several years after mining. The enhanced hydraulic properties combined with potentiometric recovery resulted in a zone of greater well yield. However, at sites with very poor transmissivity and inadequate recharge pathways, recovery may not occur. Also, at the study site, the physical enhancement was accompanied by a deterioration in groundwater quality from slightly brackish, sodium bicarbonate water to more brackish water with increased sulfate levels.

    Notes: Abstract  The historic processing of precious metal ores mined from the Comstock Lode of west-central Nevada resulted in the release of substantial, but unquantified amounts of mercury-contaminated mill tailings to the Carson River basin. Geomorphic and stratigraphic studies indicate that the introduction of these waste materials led to a period of valley-floor aggradation that was accompanied by lateral channel instability. The combined result of these geomorphic responses was the storage of large volumes of mercury-enriched sediment within a complexly structured alluvial sequence located along the Carson River valley. Much of the contaminated sediment is associated with filled paleochannels produced by the cutoff and abandonment of meander loops, and their subsequent infilling with contaminated particles. Geochemically, these deposits are characterized by variations in mercury levels that exceed three orders of magnitude. Continued lateral instability, coupled with an episode of channel-bed incision, followed the decline of Comstock mining, and has reexposed contaminated debris within the banks of the river. Erosion of bank sediments reintroduces mercury-enriched particles to the modern channel bed. It is suggested on the basis of geochemical and sedimentological data that during the bank erosion process, much of the mercury associated with fine (〈63 μ) valley-fill deposits are carried downstream without being incorporated to any appreciable extent within the channel-bed sediments. In contrast, mercury associated with larger and denser particles, particularly mercury-gold-silver amalgam grains, are accumulated in the channel-bed sediments as the river traverses polluted reaches of the Carson River valley. Concentration patterns developed along the modern channel indicate that the valley fill is the primary source of mercury to the river today. Thus, these data imply that efforts to reduce the influx of mercury to the aquatic environment should examine methods for reducing bank erosion rates.

    Notes: Abstract  An RFLP linkage map was constructed for cultivated sunflower Helianthus annuus L., based on 271 loci detected by 232 cDNA probes. Ninety-three F2 plants of a cross between inbred lines RHA 271 and HA 234 were used as the mapping population. These genetic markers plus a fertility restoration gene, Rf 1, defined 20 linkage groups, covering 1164 cM of the sunflower genome. Of the 71 loci 202 had codominant genotypic segregation, with the rest showing dominant segregation. Thirty-two of the 232 probes gave multiple locus segregation. There were 39 clusters of tightly linked markers with 0 cM distance among loci. This map has an average marker-to-marker distance of 4.6 cM, with 11 markerless regions exceeding 20 cM.

    Notes: Abstract  A 823-bp Sau3AI fragment (pSau3A10) was subcloned from a sorghum bacterial artificial chromosome (BAC) clone, 13I16, that contains DNA sequences specific to the centromeres of grass species. Sequence analysis showed that pSau3A10 consists of six copies of an approximately 137-bp monomer. The six monomers were organized into three dimers. The monomers within the dimers shared 62–72% homology and the dimers were 79–82% homologous with each other. Fluorescence in situ hybridization (FISH) analysis indicated that the Sau3A10 family is present only in the centromeres of sorghum chromosomes. Sequencing, Southern hybridization, and Fiber-FISH analyses indicated that the Sau3A10 family is tandemly arranged and is present in uninterrupted stretches of up to at least 81 kb of DNA. Slot-blot analysis estimated that the Sau3A10 family constitutes 1.6–1.9% of the sorghum genome. The long stretches of Sau3A10 sequences were interrupted by other centromeric DNA elements. Southern analysis indicated that the Sau3A10 sequence is one of the most abundant DNA families located in sorghum centromeres and is conserved only in closely related sorghum species. Methylation experiments indicated that the cytosine of the CG sites in sorghum centromeric regions is generally methylated. The structure and organization of the Sau3A10 family shared similarities with centromeric DNA repeats in other eukaryotic species. It is suggested that the Sau3A10 family is probably an important part of sorghum centromeres.
