    Notes: A total of 259 accessions of wild Triticum species originating from different countries, along with 91 triticale (6×)× bread wheat true-breeding derivatives, two bread wheat, and three triticale cultivars were screened for resistance to the Russian wheat aphid, a serious insect pest of the wheat crop. Twenty-four entries with low damage ratings on the basis of amount of leaf rolling and leaf chlorosis were retested along with resistant and susceptible controls. On the basis of leaf roll damage ratings, eight entries including four Triticum monococcum var. boeoticum (T. boeoticum), one T. monococcum var. monococcum (T. monococcum), two T. timopheevii var. araraticum (T. araraticum), and one triticale cultivar were significantly superior to ‘Karl’ (susceptible control) wheat. Among these, four accessions — three T. boeoticum and one T. araraticum— were significantly superior to all other entries and were equal to the resistant control (PI 372129) in resistance rating based on leaf rolling and leaf chlorosis (except T. boeoticum TA 202). The leaf chlorosis damage rating of all accessions were significantly lower than that of the susceptible check.

    Notes: The bottleneck restricting introgression of useful genes directly from diploid into hexaploid wheats is the low number of BC1F1 seeds obtained. In crosses between hexaploid wheat (Triticum aestivum L.; AABBDD) and Aegilops squarrosa L. (DD) or T. urartu Thum. (AA), this bottleneck may be overcome simply by pollinating a sufficient number of F1 spikes. However, hybrids between hexaploid wheat cultivars (T. aestivum) and T. monococcum L. (AA) generally are highly female-sterile, often having no pistils. One T. monococcum accession, PI 355520, when crossed with T. aestivum, produced hybrids with female fertility in the same range as that of T. aestivum/A. squarrosa or T. aestivum/T. urartu hybrids, ca. 0.5 to 1.0 backcross seed per spike. We found that female fertility was controlled by two duplicate genes in PI 355520, and that this accession can be used as a bridging parent to introgress genes from other T. monococcum accessions into hexaploid wheat. Pairing of homologous chromosomes was less frequent and weaker in such crosses than in T. aestivum/A. squarrosa crosses, but homoeologous bivalents occurred at a rate of almost 0.5 II per cell. Restitution division was detected in crosses involving all three diploid species and was confirmed cytologically in crosses with PI 355520. Chromosome numbers of BC1F1 plants ranged from 35 to 67; plants with 49 or more chromosomes occurred at frequencies of 0.09 to 0.21 among progeny of A. squarrosa and T. urartu and 0.29 in progeny of T. aestivum/T. monococcum crosses involving PI 355520. These results are consistent with those of previous studies, demonstrating the potential of direct Hexaploid/diploid crosses for rapidly introgressing useful genes into Hexaploid wheat with minimum disturbance of the background genotype.

    Notes: The wild tetraploid wheat species Tr$$ (Zhuk) Zhuk Var. araratieum is a source of pest resistance genes for T$$ aesti$$ L. Our objectives were to describe the breeding behaviour of T.arartuititm when backcrossed to common wheat and transfer resistance to leaf rust (caused by Pu$$) and powdery mildew (caused by Blumeria $$wheat. Crosses were made between five wheat genotypes and $$ accessions. Fertifity and chromosome numbers of BC$$; plants were determined. Resistance to leaf rust was transferred toBC2 -derived families from 10 different T’ararati$$an accessions. Leaf rust resistance genes in nine T. araratieum accessions can be assigned to at least four loci. Leaf rust resistance transferred from three accessions was inherited in the hexaploid derivatives as a single. $$ gene in each case. Resistance to powdery mildew was also detected in the T. araratie$$ backcross derivatives. Fertile hexaploid derivatives expressing T’araratieum-derived resistance genes can be recovered after two backcrosses to wheat cultivars.

    Notes: Direct introgression of genes from diploid Aegilops squarrosa L into hexaploid wheat (Tnticum aestivumL.) is an efficient way of developing stable, hexaploid lines with unique, useful genes. 39 such lines derived front direct crosses to determine the effects of Ac. squarrosa germplasm were field-tested on productivity. Whereas some lines were agronomically inferior to the recurrent parent, others periormed at an equall or better level. In addition to the larget traits (Hessian-fly and leaf-rust resistance), other traits, including soilborne-mosaic-virus resistance and protein concentration, were improved.

    Notes: Abstract Radiation-induced wheat-rye chromosome translocation lines resistant to Hessian fly, Mayetiola destructor (say), were analyzed by in situ hybridization using total genomic and highly repetitive rye DNA probes pSc119 and pSc74. In situ hybridization analysis revealed the exact locations of the translocation breakpoints and allowed the estimation of the sizes of the transferred rye segments. T6BS·6BL-6RL and T4BS· 4BL-6RL are terminal translocations with either most of the complete long arm of rye chromosome 6R or only the distal 57% of the 6RL arm attached to the long arms of wheat chromosomes 6B and 4B, respectively. The breakpoint in T6BS·6BL-6RL is located at a fraction length (FL) of 0.11 in the long arm of T6BS 6BL-6RL and at FL 0.46 in the long arm of T4BS·4BL-6RL. Ti4AS·4AL-6RL-4AL is an intercalary translocation with the breakpoint located at FL 0.06 in the long arm of wheat chromosome 4A. The inserted 6RL segment, with the Hessian fly resistance gene, has a size of 0.7 μm, and is the smallest and, so far, the first radiation-induced intercalary translocation identified in wheat.

    Notes: Abstract The chromatin in interphase nuclei is much less condensed than are metaphase chromosomes, making the resolving power of fluorescence in situ hybridization (FISH) two orders of magnitude higher in interphase nuclei than on metaphase chromosomes. In mammalian species it has been demonstrated that within a certain range the interphase distance between two FISH sites can be used to estimate the linear DNA distance between the two probes. The intephase mapping strategy has never been applied in plant species, mainly because of the low sensitivity of the FISH technique on plant chromosomes. Using a CCD (charge-coupled device) camera system, we demonstrate that DNA probes in the 4 to 8 kb range can be detected on both metaphase and interphase chromosomes in maize. DNA probes pA1-Lc and pSh2.5·SstISalI, which contain the maize locia1 andsh2, respectively, and are separated by 140 kb, completely overlapped on metaphase chromosomes. However, when the two probes were mapped in interphase nuclei, the FISH signals were well separated from each other in 86% of the FISH sites analyzed. The average interphase distance between the two probes was 0.50 µm. This result suggests that the resolving power of interphase FISH mapping in plant species can be as little as 100 kb. We also mapped the interphase locations of another pair of probes, ksu3/4 and ksu16, which span theRp1 complex controlling rust resistance of maize. Probes ksu3/4 and ksu16 were mapped genetically approximately 4 cM apart and their FISH signals were also overlapped on metaphase chromosomes. These two probes were separated by an average of 2.32 µm in interphase nuclei. The possibility of estimating the linear DNA distance between ksu3/4 and ksu16 is discussed.
