Zusammenfassung An isolierten Basalmembranen aus der Nierenrinde von Ratten mit einer Aminonucleosidnephrose, wurden die Verteilungskoeffizienten von Harnstoff-C14, Methylglucose-C14, Inulin-C14, Polyvinylpyrrolidon (PVP)-J131, Serumalbumin-J131 und von γ-Globulin-J131 gemessen und folgendes gefunden: 1. Der Verteilungskoeffizient nimmt mit steigendem Molekulargewicht ab. 2. Als Grenzwert für die Penetration in die nephrotische Basalmembran wurde ein Molekulargewicht von 190.000 gefunden. Das entspricht einem maximalen Porenäquivalentradius von 65 Å. 3. Als Ursache der unterschiedlichen Verteilungskoeffizienten der einzelenen Substanzen wurde — wie bei der normalen Basalmembran — eine Normalverteilung der Porenäquivalentradien nachgewiesen; bei einem mittleren Porenäquivalentradius von 36±16 Å. Die Ergebnisse zeigen, daß die Porenäquivalentradien der Basalmembran bei Aminonucleosidnephrose gegenüber der normalen Basalmembran deutlich vergrößert sind. Es darf als sicher angenommen werden, daß die Proteinurie eine Folge dieser Porenerweiterung ist.

Summary The participation coefficients of Urea-C14, Methylglucose-C14, Inulin-C14, Polyvinylpyrrolidon-J131, Serum Albumin-J131 and γ-Globulin-J131 were evaluated on isolated basement membranes from the cortex of the rat kidney with aminonucleoside nephrosis. The following observations were made: 1. The participation coefficient decreases as the molecular weight of the test substance increases. 2. A molecular weight of 190.000 is the borderline molecular weight for the penetration of a molecule into the nephrotic basement membrane. This indicates a maximal pore equivalent radius of 65 Å. 3. The different participation coefficients for different substances can be explained on the basis of a “normal distribution” of the pore equivalent radii. The mean pore equivalent radius of the nephrotic basement membrane is 36±16 Å. Our results indicate that the pore equivalent of the nephrotic basement membranes are greater than those of the normal basement membranes. It is suggested that the proteinuria of the nephrotic syndrome results from this increase in diameter of the pores of the nephrotic basement membranes.
