    Notes: Chitosan samples manufactured under different conditions were compared for effectiveness of coagulating an activated sludge suspension grown on vegetable canning wastes. Computer analysis of data from Buchner funnel filterability tests resulted in quadratic polynomial equations describing the response curves for volume of filtrate versus dosage, expressed as g/liter chitosan/100 g sludge suspended solids (SSS). The quotient of the filtrate volume and dosage at the inflection points of the equations obtained for 10 test samples and 1 commercial chitosan sample were compared to evaluate the response (effectiveness) per unit amount for each chitosan product. The product made by a standard procedure (deproteinated with 3% NaOH at 100°C for 1 hr, demineralized with 1N HCL at ambient temperature for 30 min, and deacetylated with 50% NaOH at 145-150°C under N2 for 5 or 15 min) gave the best performance as a coagulating agent for this activated sludge system. Other products, including the commercial preparation, required higher dosages to achieve the same effectiveness. Products deacetylated in the presence of sir rather than nitrogen decreased waste treatment effectiveness, which approximated the trends of reduced viscosity and molecular-weight distribution. The products containing minerals were less effective than products from which minerals had been removed prior to deacetylation, but they were more effective than the enzyme treated sample and the commercial product. In general, although chitosan products obtained after 15 min deacetylation were more effective than those receiving 5 min deacetylation, effectiveness did not correlate linearly with viscosity and molecular-weight distribution trends. However, chitosan products deacetylated for 15 min did show that the higher-molecular-weight products (0.65-1.1 × 106) were more effective coagulating agents for activated sludge than the manufactured product having the lowest molecular weight (0.47 × 106) and the commercial reference sample (0.56 × 106). Thus, higher values for molecular weight were predictive of greater effectiveness for coagulation of activated sludge suspensions.

    Notes: A generalized mechanistic model for the enzymatic hydrolysis of cellulose is developed and expressed mathematically. The model is based on Michaelis-Menten-type kinetics for concurrent random and endwise attack of the substrate involving end-product inhibitions and three types of enzymes: an endo-β;-1,4-glucanase, an exo-β-1,4-glucanase, and β-glucosidase. Basic parameters of the model which can explain synergistic and other effects observed experimentally are quantified and discussed. It is shown that cellulose degradation kinetics are expected to be strongly affected by the ratio of endo- to exocellulases in the reaction mixture as indicated by previous experimental data, and the substrate degree of polymerization, a factor not fully appreciated in previous studies, which appear to be overridingly important in many practical cases.

    Notes: Ten chitosan products, prepared as described in part I of this study, were evaluated in jar tests that measured their effectiveness for coagulation of suspended solids and removing turbidity from cheese whey. A polynominal regression analysis was found to be useful for determining the optimal effectiveness of each chistosan preparation, and was expressed as the percent reduction on turbidity per unit concentration of chitosan added. The effectiveness of the chitosan products was found to be inversely related to their molecular-weight values. This situation was different from the findings described in part II of this study, in which the filterability of activated sludge was tested. Enzymatic deproteination yielded chitosan products that performed better than those produced by alkali deproteination. Demineralized products were also more effective than those that had not been demineralized. The preparations deacetylated under a nitrogen atmosphere were more effective than those deacetylated in air, but this was shown to be true only for the first 5 min of deacetylation. When deacetylated for 15 min, no differences were noted. In this study, differences in performance between the various products were largely due to the differing dosages required to achieve the maximum reduction in turbidity of cheese whey, while the maximum responses achieved by the various products tested were about the same. A commercial product, which was less effective as a sludge coagulating agent in part II of this study, was more effective for cheese whey coagulation and turbidity removal than the majority of the experimental chitosan preparations tested.

    Notes: A temperature dependence study of the ultrasonic amplitudes, velocities, and relaxation times for a presumed conformational transition of noncomplexed aqueous 18-crown-6 (1,4,7,10,13,16-hexaoxacyclooctadecane) is discussed. At all temperatures a single relaxation was observed within a 15-255-MHz frequency range. The equilibrium constant for the presumed conformational transition \documentclass{article}\pagestyle{empty}\begin{document}$ {\rm CR}_1 \mathop \rightleftarrows\limits^{K_{12} } {\rm CR}_2 $\end{document} was determined to be K21 = (2 ± 2) × 10-2. The activation parameters are ΔH21≠ = 10.2 ± 1.0 kcal/mol, ΔS21≠ = 7.7 ± 0.2 cal/(mol·deg), ΔH12≠ = 7.4 ± 1.0 kcal/mol, and ΔS12≠ = 7.7 ± 0.2 cal/(mol·deg), while the thermodynamic enthalpy and entropy were found to be -2.6 ± 1.0 kcal/mol and 0 ± 0.2 cal/(mol·deg), respectively. The rate constants at 25.0°C for the presumed conformational transition are k21 = (1.0 ± 0.3) × 107 sec-1 and k12 = (6.2 ± 0.2) × 108 sec-1.

    Notes: The oxidation of malonic acid by ceric ions has been investigated in sulfuric acid solution under a variety of conditions. The initial rate at low ceric ion concentrations is first order in each of the two reactants and has an activation energy of 11.6 kcal/mol; the instantaneous rate constant increases somewhat with time during a single run. At higher concentrations of ceric ion, semilogarithmic plots are sigmoid with a reduced rate constant at long times. The rate decreases slightly with increasing sulfuric acid concentration. Rates of carbon dioxide evolution may be much less than rates of ceric ion reduction because of supersaturation effects. The observations can be explained if dissolved oxygen reacts with organic radicals to catalyze the rate of initial attack on malonic acid, but oxygen must also be consumed irreversibly during these reactions. Computations with plausible rate constants have simulated the experimental observations. These oxygen effects can rationalize peculiar almost discontinuous changes in rate when bromomalonic acid is oxidized by ceric ion. These effects may also explain the previously puzzling observation that some Belousov-Zhabotinsky solutions are oscillatory in bulk but become quiescent but excitable when spread in a thin film in contact with air.

    Notes: Emission processes from lower excited states S1 (fluorescence) and T1 (phosphorescence) have been studied in the gas and liquid phases when biacetyl is excited into the second singlet state S2. (In agreement with Kasha's rule no fluorescence is observed from the S2 state.)In the liquid phase, when biacetyl is excited into the singlet states S1 and S2, no difference is observed between these emission processes. This phenomenon certainly results from an efficient nonradiative transition between the second excited singlet state S2 and the first excited state S1 with practically no excess vibrational energy. The quantum yield of this transition is almost unity and does not depend on the nature of the solvent.In the gas phase no emission processes are observed when biacetyl is excited into the S2 state at low pressure (less than 10 mm Hg). High pressure of inert gas is necessary in order to observe these processes. As for excitation into the S1 state with vibrational energy, loss of vibrational energy through collisions occurs from the S2 state. The quantum yield of the S2 → S1 transition by excitation at 290 nm is estimated around 0.5-0.6 at 6 atm of inert gas (ethane, ethylene, or carbon dioxide).

    Notes: A 1000 liter fermentor has been used to produce a continuous feed of Escherichia coli containing a high level of β-galactosidase. We have investigated the individual unit operations for the isolation of the enzyme: cell disruption, nucleic acid removal, protein precipitation, and solid-liquid separation after each stage. Using the information obtained we have been able to operate a semicontinuous process which when fully continuous would yield 100 g protein/hr, comprising 23% β-galactosidase.
