    Notes: Summary A Pseudomonas sp. which was isolated from phenol-containing soil was immobilized in alginate and polyacrylamide-hydrazide (PAAH) and cultivated in a special airlift fermenter. The immobilized Pseudomonas sp. was able to degrade phenol at initial concentrations up to 2 g/l in less than 2 days, although the free cells did not grow at this concentration. The immobilization materials act as a protective cover against phenol, PAAH being more effective than alginate. The degradation activity as well as the outgrowth of bacteria can be manipulated by the concentration of the immobilization material, the temperature and the nitrogen content in the medium. The cells grew predominantly in microcolonies in the outer area of the beads when nitrogen was available as 1.0g NH4NO3/l and 0.5g (NH4)2SO4/l.

    Notes: Summary Mycelia of Claviceps purpurea CBS 164.59 were immobilized in 2%, 4%, and 8% calcium alginate. Alkaloid production by free cells declined after 60 days, while immobilized cells retained their activity for 200 days. The cumulative alkaloid production for all fermentation cycles using 8% calcium alginate immobilized mycelia was 25 times higher than that from free cells. The best yields of the ergopeptide ergometrine were reached with 4% gel immobilized mycelia, while higher gel concentrations caused a shift in the alkaloid biosynthesis towards high clavine alkaloid production. Beginning with the third cycle of reincubation the immobilized mycelia showed a marked tendency to fragmentize into vacuolated arthrosporoid-like structures and produced violet-black pigments so that the beads recalled sclerotial structures of parasitically living Claviceps.

    Notes: Summary The spores of Aspergillus niger were entrapped in calcium-alginate beads and precultivated in growth media with various amounts of nitrogen. During the following citric acid production in shaking cultures an optimum of acid formation and yield was observed after the precultivation with 100–200 mg/l NH4NO3. The productivity of the immobilized Aspergillus was found to be 1.5 times higher than in the case of free pellets. The outgrowth of free mycelia into the medium could be provided by increasing the ratio particle-volume: medium volume, using a 1-l air-lift fermenter, by which means the productivity was increased twice as much as obtained in shaking culture.

    Notes: Summary Following growth on n-alkanes, undecanoic acid in high concentrations completely inhibits the acylation of fatty acids formed during the terminal oxidation so that the intracellular fatty acid pattern is composed exclusively of components from the de novo synthesis. An inhibitory effect of undecanoic acid stems presumably from the effect it has on the long-chain acyl-coenzyme A synthetase I, whereas the corresponding long-chain acyl-coenzyme A synthetase II, which is bound to specific cell organelles remains untouched by this inhibition. The strongly reduced growth, even following glucose oxidation, probably comes from the effect of C11-acid on specific intramitochondrial situated enzymes.

    Notes: Summary The morphological development of citric acid producing Aspergillus niger immobilized in Ca-alginate and K-carrageenan was studied. The fungus normally developed a dense mycelium layer below and on the gel bead surfaces so that substrate and oxygen in this area had direct contact with mycelia. By this way mycelia are not only immobilized by entrapment but also in a “pellet-like” matter. Limitation of the nitrogen source induces a more interior mycelium growth, and outgrowing of free cells was minimized. In sucrose media no effect on the particle stability was observed whereas the application of potassium acetate as substrate caused the complete dissolving of the matrices.
