Notes: Abstract The potent inhibition of the shikimate pathway enzyme 5-enolpyruvylshikimate-3-phosphate (EPSP) synthase by the broad-spectrum herbicide glyphosate (N-[phosphonomethyl]glycine) was confirmed for the enzymes extracted from various bacteria, a green alga and higher plants. However, 5 out of 6 species belonging to the genus Pseudomonas were found to have EPSP synthases with a 50- to 100-fold decreased sensitivity to the inhibitor. Correspondingly, growth of these 5 species was not inhibited by 5 mM glyphosate, and the organisms did not excrete shikimate-3-phosphate in the presence of the herbicide.

Notes: Summary We investigated the effects of phentolamine and another more selective α2-adrenoceptor antagonist, rauwolscine, on insulin release in vivo (in female Wistar-rats) and in vitro (in perfused rat pancreas and in isolated perifused mouse islets). Phentolamine was found to significantly increase glucose-induced insulin release. On the other hand, rauwolscine failed to do so, when applied in a concentration that effectively antagonized the inhibitory effect of clonidine. These results demonstrate that phentolamine is capable of directly stimulating insulin release. This effect is thus not mediated by α-adrenoceptors. For this reason phentolamine is not an appropriate tool to study possible inhibitory effects of the sympathetic nervous system on insulin release. An enhanced insulin response as may be observed in animals and in man in the presence of phentolamine does not furnish evidence for a tonic inhibitory control of the islet cells by the sympathetic nervous system.

Notes: Summary Premitotic stages of experimentally induced vascular differentiation were investigated by confocal laser scanning-fluorescence and electron microscopy. Activation of former quiescent and highly vacuolated cortex cells, observed in living and in fixed tissue, involves a centripetal movement of the nucleus along newly-formed, thick cytoplasmic strands. These traverse the large vacuoles with angles different from the former polar axis, thereby anticipating the orientation of later developing phragmosomes, future divisions, and the course of wound-vascular elements. A premitotic increase in cellular activity is reflected by the number of mitochondria, rough ER-cisternae, active dictyosomes and dense cytoplasm as well as by shape of nuclei and development of “nucleolar vacuoles”. Microtubules occur in parallel arrays running through the transvacuolar strands and radiating from the nucleus towards the cell wall. They might be responsible for the centripetal movement of the nucleus through the vacuole and/or might participate on the orientation of the plane of a future division. Preprophase bands were not detected in cortex cells of the investigated root region, which prior to wounding were already fully elongated.
