    Notes: We are incorporating a system of fast, variable apertures in our designs for the x-ray, neutral, and ion spectrometers for plasma diagnostics on Mirror Fusion Test Facility B (MFTF-B). The purpose of these apertures is to maintain optimized count rates, and thus optimized measurement statistics during an experimental shot, where flux variations as large as 104–105 may occur in periods as short as 0.1 s. We shall discuss the application of the fast apertures to the x-ray spectrometer in particular, and to the other diagnostic instruments in general. The particular design requirements for the optimized aperture system will be discussed, as will be the influence the system has on the data-handling process. We have performed experiments to test this new concept and shall present the results, including the details of the closed-loop feedback circuit controlling the apertures, optimization of the loop gain, and the engineering issues of the system, such as the effect of vacuum feedthrough, position sensitivity, and alignment criteria.

    Notes: The Tandem-Mirror Experiment-Upgrade (TMX-U) has a large and complex system of Bayard–Alpert, magnetron, and Penning gauges, in addition to mass spectrometers (RGA), all of which measure neutral pressures in the many internal regions of TMX-U. These pressure measurements are used as part of the confinement physics data base as well as for management of the TMX-U vacuum system. Dynamic pressures are modeled by a coupled-volumes simulation code, which includes wall reflux, getter pumping, and plasma pumping.

    Notes: Summary A DNA marker C7, localised Xp21.1-Xp21.3, has been studied in kindreds segregating for Duchenne muscular dystrophy (DMD) and Becker muscular dystrophy (BMD). In DMD families four crossovers were observed in 38 informative meioses between C7 and the DMD locus (θ=0.12, z max=+2.72). In BMD families no recombinants were observed in the 16 informative meioses studied. These data are consistent with the localisation of the mutations in these disorders being in the same region of Xp21. Studies in families also segregating for the DNA marker 754 support the previously reported physical order of these loci as X centromere-754-DMD-BMD-C7-X telomere. A recombination fraction of 0.11 (z max=+5.58) was found between DMD-754 by combining our previously published data with the data presented here. C7 and 754 thus provide good bridging markers for the diagnosis of DMD and BMD.

    Notes: We investigated the actions of human placental lactogen (HPL) and human growth hormone (HGH) on [3H]thymidine incorporation and the release of immunoassyable somatomedin-C (SM-C) by isolated myoblasts, dermal fibroblasts, and costal cartilage explants taken from human fetuses, at 11-21 weeks of gestation. The incorporation of [3H] thymidine by myoblasts and fibroblasts was significantly increased after incubation for 20 hr or 44 hr, and cell number after incubation for 7 days, in the presence of 50-250 ng/ml HPL. Incubation with HPL did not increase [3H]thymidine incorporation into cartilage explants, whereas incubation with HGH failed to enhance the uptake of this isotope by any of the tissues. Following extraction with acid-ethanol, culture medium conditioned by exposure to myoblasts or fibroblasts for 44 hr, and to cartilage explants for 7 days, contained radioimmunoassayable SMC. Myoblast-conditioned medium contained significantly more SM-C [1,609 ± 953 mU/mg cell protein (mean ± SD) n = 10] than did that conditioned by fibroblasts (637 ± 323; n = 5; P 〈 0.02). In 1 week of culture, cartilage explants released 4.1 ± 1.1 mU/mg wet weight (n = 7). The release of immunoassayable SM-C from cultured cells was significantly increased in the presence of 250 ng/ml HPL in five of eight experiments with myoblasts and two of four experiments with fibroblasts. Neither fibroblasts or myoblasts showed increased SM-C release following exposure to HGH.The results suggest that HPL, but not HGH, is growth-promoting for some human fetal tissues in vitro and that this action is mediated, at least in part, by an increased release of somatomedins.
