    Notes: Summary 3′-Deamino-3′-(4-morpholinyl)adriamycin (MRA) and 3′-deamino-3′(3-cyano-4-morpholinyl)adriamycin (MRA-CN) were compared with adriamycin (ADR) in ADR-sensitive (P388/S) and-resistant (P388/ADR) murine leukemia cell lines with respect to cytotoxicity and cellular accumulation. MRA is only two- to threefold more cytotoxic to P388/S in culture than ADR, whereas MRA-CN is 500-fold more cytotoxic than ADR to this cell line. Yet both MRA and MRA-CN retain their potency against P388/ADR in spite of a 150-fold decrease in potency for ADR. The observed noncross-resistance of both MRA and MRA-CN in P388/ADR correlates with their increased cellular uptake and retention relative to ADR and the inability of P388/ADR to exclude these analogs as readily as it does ADR. The decreased uptake of MRA and MRA-CN in P388/ADR relative to P388/S (1.5 to 2.0-fold), the increased efflux, and the ability of verapamil to enhance cellular uptake of these analogs in P388/ADR, as it does with ADR, all indicate that the mechanism of ADR-resistance effects ADR and the morpholino analogs in a similar manner but to far different extents. The potent cytotoxicity of MRA-CN appears to be related to strong cellular interactions of the drug with macromolecules that are characterized by its nonextraction from cells by chloroform: methanol or 10 M urea and may therefore represent covalent binding.

    Notes: Summary Urine of twenty hospital workers was monitored for the excretion of the cytostatic drug cyclophosphamide using GC-MSD. The drug was found to be present above the detection limit of 0.5 μg/24 h urine in five cases. A clear relationship between cyclophosphamide handling and the detectability of excretion existed. This method developed can be of use for biological monitoring studies directed toward the finding of exposure hazards.

    Notes: Abstract An automated continuous flow process has been developed for the synthesis of11C-ethanol and11C-butanol. These alcohols were synthesized via the same route. The reaction of11CO2 with methylmagnesium bromide or with n-propylmagnesium chloride, followed by a lithium aluminum hydride reduction and hydrolysis produced respectively11C-ethanol and11C-butanol. Preparation can be completed in 25 min. In each case the radiochemical purity, as determined by high pressure liquid chromatography /HPLC/ was greater than 98%. Biological quality control shows that the products are suitable for human use. The process has been completely automated to limit radiation exposure to personnel, reduce preparation time, and increase reproducibility.

    Notes: Abstract Position emission tomography can picture the distribution of flow tracers as well as of metabolic substrates or analogs. Studies of the distribution of these tracers allow to infer information about regional myocardial clearance (flow x extraction) and substrate utilization. In a study of 32 patients after myocardial infarction, we have contrasted flow and substrate utilization to demonstrate ischemic but viable myocardium in the arterial territory of the infarct in a number of patients also specially after fibrinolytic reperfusion. Restoration of blood flow to the ischemic but viable myocardium through coronary bypass or dilatation improves flow from 56.3% to 84.2% of control and restores substrate utilization. In another group of 32 patients studied with the Strontium-82/Rubidium-82 generator, we have demonstrated perfusion changes both in the myocardial infarct area and at a distance. These changes predominate in patients with multiple vessel disease. Combined PET studies of flow and substrate utilization are new tools to study early intervention after myocardial infarction and to document the benefits of revascularization.

    Notes: Abstract 57Fe Mössbauer effect spectroscopy is employed to determine the relationship between the microstructure and the mechanical properties of martensitic steels with base composition Fe-10wt%Cr-0,26wt%C. The microstructure consists predominantly of two phases: martensite and austenite. The effect of low concentrations of both Mn and Ni on the structure and the mechanical properties of these steels is studied. The results indicate that Mn and Ni additions are equally effective in increasing the fraction of retained austenite. The austenite is an important phase since it is considered to be beneficial to the toughness of steel. However, we find that the impact toughness first decreases and then increases as a function of the fraction of austenite.

    Notes: Abstract We have investigated the phagocytic properties of the macrophage-like cell line DCH-7, derived from fusion of mouse macrophages with a mouse T-lymphoma cell line. These cells phagocytosed opsonized bacteria. IgG appeared to be the major opsonin forStaphylococcus aureus Wood 46 as well as for threeEscherichia coli strains; complement components were not required as opsonins. Intracellular bacteria survived to a large extent. This model system should be a useful tool for studying the process of phagocytosis and phagocytic killing of bacteria.
