    Notes: Summary An attempt at cytochemical demonstration of acidification proton-translocating ATPase (H+-ATPase) of Golgi complex in rat pancreatic acinar cells has been made by using p-nitrophenylphosphatase (NPPase) cytochemistry which is used for detecting of Na+-K+-ATPase (Mayahara et al. 1980) and gastric H+-K+-ATPase (Fujimoto et al. 1986). K+-independent NPPase activity was observed on the membrane of the trans cisternae of Golgi complex, but not inside of cisternae. The localization of NPPase activity is different from that of acid phosphatase activity where reaction products were seen on the inside of the trans Golgi cisternae. Since this activity was insensitive to vanadate, ouabain and independent of potassium ions, it was distinct from plasma membranous ATPases such as Na+-K+-ATPase and Ca2+-ATPase. The K+-independent NPPase activity was diminished by the inhibitors of H+-ATPase such as N-ethylmaleimide (NEM) and 4,4′-diisothiocyanostilbene-2,2′-disulfonic acid (DIDS). The NPPase reaction products were also seen on the membranes of other acidic organelles, i.e., lysosomes, endosomes, autophagosomes and coated vesicles. These results suggest that NPPase activity on the membrane of the Golgi complex and other acidic organelles corresponds with H+-ATPase which plays a role in acidification.

    Notes: Summary A frame-shifttat gene mutant of human immunodeficiency virus type 1 (HIV-1), which showed no detectable trans-activation potential, was constructed in vitro. Upon transfection, this clone directed the synthesis of virus mRNAs,gag proteins and virion-associated reverse transcriptase (RT) at a low level as was observed with thetat mutants of HIV-2 and simian immunodeficiency virus isolated from African green monkey (SIVAGM). Using these mutant viruses, trans-activation efficiency of viral gene expression bytat was compared among HIV-1, HIV-2, and SIVAGM. SIVAGM seemed to be less dependent ontat for RT production than HIV-1 and HIV-2.

    Notes: Abstract Composite spherical powders made of aluminium and yttria were prepared by the mechanical shock method. This unique idea for composite powder products, without the use of heat, and making use of the ductility of the metal and the brittleness of ceramics, has a preparation time of only 3 min. For analysis of the cross-sections of an Al-Y2O3 composite particle, electron probe microanalysis was used. In the composite particle, a diffused layer is formed at the interface between Al and Y2O3; and the boundary between these components is thought to be quite strong. These composite spherical powders are excellent regarding fluidity and their spray capability. The Al-Y3O3 composite powder was initially plasma sprayed on to an aluminium alloy substrate, and then coated with partially stabilized zirconia. The tensile strength of these test pieces was studied as a function of compositional changes in the Al-Y2O3 system. For 0.2 to 1 mol% Y2O3, the tensile strength is much higher, values up to 75.5 M Pa being attainable. The heat-cycle number of test pieces with the above composition range at the present time has been measured at over 2000.

    Notes: Abstract Studies of the dHvA effect have been made on NbO single crystals grown by a zone melting techniques and 13 separate frequency branches ranging from 5.2×106 G to 1.2×108G have been observed. To understand the origin of the dHvA frequencies, we have made a self-consistent band structure calculation. Most experimental frequencies are found to be assigned to the extremal orbits on the calculated Fermi surfaces. There is good qualitative agreement between experiment and calculation, but some quantitative discrepancies still exist. We also report some physical properties, i.e., electrical resistivity, superconducting transition temperature, and electronic specific heat, of this high-quality NbO crystal.

    Notes: Summary We measured the amount of 1,2-diacylglyccrol (DAG) and its fatty acid composition in hamster hearts of various ages because 1,2-DAG has been recognized to be onc of the intracellular second messengers. The highest level in 1,2-DAG was detected in 30-day-old hamster hearts. The heart weight increased about 2.5 times between 30 and 90 days of age. Hamster hearts at 90 days of age had 72% of the 1,2-DAG content at 30 days of age. The amount of 1,2-DAG at 240 days of age was a little lower than that in 90-day-old and 160-day-old hamsters, whereas there was no significant increase in heart weight and only a little gain in body weight between 90 and 240 days of age. The analysis of the fatty acid composition of 1,2-DAG showed an increase in the percentage of 18∶0 accompanied by a change in the percentages of other fatty acids with the growth of hamsters. The largest amount of arachidonic acid expressed as 20∶4 (n-6) was found at 30 days of age. On the other hand, triglyceride, cholesterol, and major phospholipid components of hamster hearts increased in accordance with growth. In particular, the increase in triglyceride content was remarkable, indicating that myocardial lipidosis may be induced by aging. These results demonstrate that 1,2-DAG may be involved in the synthesis of protein during heart growth and that the alteration of its fatty acid composition is related to the growth of hamsters.
