    Notes: To shed more light on the controversial findings concerning the functional participation of the highly conserved nut-like leader box A sequence element in ribosomal RNA transcription antitermination we have carried out a mutational study. We have substituted the box A and combined this mutation with several deletions comprising the rRNA leader elements box B, box C and the tL region. The mutations are located within the genuine rrnB operon cloned on multicopy plasmids. We determined the effects of the mutations on cell growth, rRNA accumulation and ribosomal subunit stoichiometry. Cells transformed with the mutated plasmids were affected in their growth rate, and showed a surprising deficiency of the promoter-proximal 16S compared to the 23S RNA, indicative of a post-transcriptional degradation event. Accordingly, we could demonstrate a reduced amount of free 305 relative to 50S ribosomal subunits in exponentially growing cells. Similar stoichiometric aberrations in the ribosome pool were detected in conditionally Nus factor-defective strains. The results show that the leader box A sequence within rRNA operons has important post-transcriptional functions for 16S RNA stability and ribosomal subunit stoichiometry. A model is proposed, describing the biogenesis and quality control of ribosomes based on rRNA leader and Nus-factor interactions. It is compatible with the previously observed effects of box A in antitermination.

    Notes: Abstract In 50 patients, 1 mCi123I phenylpentadecanoic acid (IPPA) was injected at peak ergometric stress and 1500 frames were acquired (1 frame/s) with a high count rate gamma camera. Parametric images of rates of decrease and increase for different time intervals after stress were compared with coronary angiography and LV ventriculography, separately evaluating the 3 main coronary territories: 18/150 territories supplied by normal coronaries presented rather homogeneous regional clearing rates, whereas a gradual decrease in clearing rates towards the end of the territory (frequently with peripheral defects) was seen in all 87/150 territories with significant coronary narrowing. In local correspondence to clearing defects, initial IPPA accumulations could be observed with later onset of clearing between 10 and 25 min. 44/150 territories presented abnormal clearing rates, mostly with a patchy pattern, with normal coronary anatomy, but all except one had LV dysfunction and a clinical diagnosis of Cadomyopathy, diabetes mellitus or hypertensive disease. Twenty four of the 41 patients with CAD had, in correspondence to a prior myocardial infarction, minimum or missing metabolic activity frequently in circumscribed zones, partly separated by bridges of still viable tissue with preserved but reduced clearing rates.

    Description / Table of Contents: Linear poly(dimethyl siloxanes) bearing vinyl groups can be converted to crosslinked products by photoinitiated formation of radicals. In chloroform solution the quantum yield indicated by the decrease in double bonds (ΦP) assumes values of 0.9 to 2.5, depending on the type of initiator. It decreases with rising silicon concentration. A ΦP-value of 0.01can be extrapolated for solvent free systems. Under these conditions network formation is particularly efficient with benzoin derivatives. As the content of vinyl groups increases the network density also rises. As is demonstrated by IR spectroscopic studies on benzoin derivatives as initiators and by characterization of the networks built up this way, network formation is almost exclusively due to the vinyl groups.

In CHCI3-Lösungen erreichen die Quantenausbeuten der Doppelbindungsabnahme (Φp) Werte von 0,9 bis 2,5 Sie hängen vom Initiatortyp ab und sinken bei Erhöhung der Siliconkonzentration. Für lösungsmittelfreie Systeme kann ein ΦP-Wert von 0,01 extrapoliert werden. Unter solchen Bedingungen erfolgt die Netzwerkbildung besonders effektiv mit Benzoinderivaten. Mit steigendem Vinylgruppengehalt der Silicone erhöht sich die Netzwerkdichte. IR-spektroskopische Untersuchungen mit Benzoinderivaten als Initiator und die Charakterisierung der entstandenen Netzwerke zeigen, daß die Netzwerkbildung fast ausschließlich über die Vinylgruppen abläuft.
