    Notes: Abstract  We report on a 13-year old girl with severe aplastic anemia and hypertrophic cranial pachymeningitis. She was admitted to our hospital with severe headache and vomiting. A computerized tomographic (CT) scan of the brain on the third day of symptoms showed a hyperdense area in the tentorial region. Magnetic resonance imaging (MRI) showed iso-intensity in the same tentorial region in T1- and T2-weighted images, and gadolinium enhancement of this region suggested a thickened dura mater. Initially, a diagnosis of subdural or subarachnoid hemorrhage was made. Since her platelet count was low (3000/μl) making the patient a poor-risk candidate for surgery, and the area was limited to the dura mater, conservative therapy, including glycerol administration and platelet transfusion, was carried out. Despite clinical improvement 10 days after admission without specific therapy, the iso-intense region on the left side of the tentorial region remained unchanged on MRI. On the other hand, the iso-intense area on the right side of the tentorial region became hyperdense on T1-weighted MRI images and was also enhanced by gadolinium. Cerebrospinal fluid findings were normal except for slightly elevated protein at 62 mg/dl. A diagnosis of hypertrophic cranial pachymeningitis of the tentorial dura mater with hemorrhage on the right side was made. Although hypertrophic cranial pachymeningitis is a rare disease, it must be considered in the differential diagnosis of severe headache in a case of aplastic anemia.

    Notes: Abstract  The most important commercial coffee species, Coffea arabica, which is cultivated in about 70% of the plantations world-wide, is the only tetraploid (2n=4x=44) species known in the genus. Genomic in situ hybridization (GISH) and fluorescent in situ hybridization (FISH) were used to study the genome organization and evolution of this species. Labelled total genomic DNA from diploid species (C. eugenioides, C. congensis, C. canephora, C. liberica) closely related to C. arabica was separately used as a probe in combination with or without blocking DNA to the chromosome spreads of C. arabica. GISH discriminated between chromosomes of C. arabica only in the presence of an excess of unlabelled block DNA from the species not used as a probe. Among the range of different species combinations used, DNA from C. eugenioides strongly and preferentially labelled 22 chromosomes of the tetraploid C. arabica, while the remaining 22 chromosomes were labelled with C. congensis DNA. The similarity of observations between C. arabica and the two diploid species using two ribosomal genes with FISH with respect to metaphase chromosomes provided additional support to the GISH results. These results confirm the allopolyploid nature of C. arabica and show that C. congensis and C. eugenioides are the diploid progenitors of C. arabica.

    Notes: Abstract The value of measuring soluble interleukin-2 receptor (sIL-2R) in the sera of patients with joint pain as a predicting parameter for the future development of rheumatoid arthritis (RA) was examined. sIL-2R was measured by the ELISA method. Sixty-four patients with joint pain (suspected RA: sus-RA) but no bone or joint destruction were enrolled over 2 years and 47 were selected for the study. Eleven patients whose diagnosis was sus-RA after a year of observation were successively followed-up for 5 years. Two-thirds of the patients whose sIL-2R levels were higher than those of normal healthy adults (〈82 pmol/l; mean+2SD) developed RA within a year. On the other hand, one-quarter of the patients with normal levels of sIL-2R also developed RA within a year. The presence of two or three of the following three items in patients with joint pain without any bone and joint destruction was thus indicated to be useful for the early diagnosis of RA: elevated CRP level (≥1.0 mg/dl), positive rheumatoid factor (RF) (≥30 IU/ ml) and an elevated sIL-2R level (≥100 pmol/l). Sensitivity and specificity were 72.7% and 96.0%, respectively. The probability of development of RA is expressed asP=1/[1+exp(2.673−0.01784×sIL-2R−0.4398*#x00D7;CRP − 0.004835 × RF)], withR 2=0.3083 andp〈0.0005. On the other hand, the sIL-2R levels did not correlate with any future bone or joint changes within a year of observation. The above criteria may therefore hopefully justify the early treatment of patients with joint pain using drugs that can modify the patients' immune fuction. However, the validity of these criteria still need to be examined more thoroughly in the future.

    Notes: Abstract The expression of metallothionein, an intracellular heavy-metal-binding protein, and p-glycoprotein, an energy-dependent drug efflux pump, was examined to study the mechanism of cell resistance to gold sodium thiomalate (GST). THP-1, one of the monocyte-derived cell lines, was cultured for 6 months and resistance to 25 μg/ml of GST (GST-resistant cells) was thus induced. The GST-resistant cells were then cultured with bucillamine to examine the presence of cross-resistance. The intracellular GST concentration was examined by flameless atomic absorption spectroscopy. The cell viability was determined by the uptake of 3–4,5 dimethylthiazole-2,5 diphenyl tetrazolium bromide (MTT). The expression of p-glycoprotein was detected by Western blotting using monoclonal anti-p-glycoprotein antibody. The expression of metallothionein was detected using the indirect immunofluorescence technique. GST-resistant cells did not show any cross-resistance to bucillamine. The rate of cytoplasmic GST accumulation decreased in the GST-resistant cells, while the rate of GST efflux also decreased. The expression of p-glycoprotein in the GST-resistant cells was not significantly different from that in the cells not treated with GST. On the other hand, the GST-resistant cells showed a higher expression of metallothionein than cells not treated with GST. These findings suggest that the induced resistance to GST might partly be due to an induction of metallothionein.

    Notes: The link between childhood experiences (before age of 16) and later onset of depression was examined among 98 young Japanese women who had all been newly employed by a company in Tokyo, Japan. We compared three groups: (a) 15 women who had reported a single episode of DSM-III-R Major Depression of less than two years duration (single episode; S.E.); (b) four women who had reported either more than one episode or any episode of two years or more duration meeting the criteria of Major Depression (recurrent or chronic; R.C.) and; (c) 53 women who had never experienced any major DSM-III-R Axis I disorders (normal control). The three groups did not differ significantly in terms of any parental loss experiences (either death or separation for 12 months or longer). The S.E. group perceived the father to be less affectionate than the other two groups. The R.C. group reported having been punched with a fist by the mother more frequently, and bullied at school. Among early life events (other than being bullied), parental divorce and own illness were reported more frequently by the R.C. group, and not being appointed as a "class leader" by the S.E. group. These findings suggest that early human experiences are linked to later depression and that single episode and recurrent/chronic depressions are discrete in their life history profiles. In order to screen women who need prevention and intervention (R.C. in particular) in community or school settings, it may be useful to tap their life history.

    Notes: Abstract Atmospheric concentration and total (wet plus dry) and dry deposition of210Pb and7Be have been measured at our laboratory in Ishikawa Prefecture in Japan which faced the Sea of Japan during 1993–1994. At the same time inventories of excess210Pb in soil samples were measured from a mixed pine forest. On the basis of the data obtained, an attempt was made to evaluate the dry deposition velocity as a result of the interception by the canopies of airborne210Pb in a pine stand The dry deposition velocity of210Pb in the pine stand was roughly estimated to be 0.9–0.8 cm/s, higher by a factor of about 10 than the value (mean: 0.12±0.09 cm/s; range: 0.06–0.28 cm/s) in the open area measured on the roof of our laboratory.
