    Notes: Tumour necrosis factor-alpha (TNFα) has been suspected of playing an important role in the pathogenesis of inflammatory bowel diseases, and has become a target for the treatment of these diseases. Open-label, placebo controlled studies have shown that engineered CDP571 and chimeric anti-TNF antibody (cA2) provide a significant benefit in Crohn’s disease. Since these antibodies have to be used repeatedly to maintain remission in inflammatory bowel disease, there is a concern that their use may compromise host defence and produce toxic side-effects.〈section xml:id="abs1-2"〉〈title type="main"〉Methods:We evaluated the combined use of mouse specific TNFα mab (25 μg/mouse, Endogen) and pentoxifylline (PF, 100 mg/kg/day, p.o., TNFα release inhibitor) in the DSS (3% dextran sulphate solution) model of mouse colitis. Colitis was induced by the feeding of 3% DSS for three cycles. The study groups were: Group I: single injection of rat anti-mouse IgG, Group II: single injection of TNFα mab, Group III: daily PF for three cycles, Group IV: single injection of TNFα mab + PF for three cycles, Group V: TNFα mab at the beginning of each cycle (three injections) and Group VI: TNFα mab (three injections) + daily PF for three cycles. Daily disease activity (DAI) was measured throughout the study. At the end of each cycle, colon tissue was processed for histology, myeloperoxidase (MPO) and plasma TNFα.〈section xml:id="abs1-3"〉〈title type="main"〉Results:Mice treated with a single injection of TNFα alone or TNFα mab + PF showed significantly lower DAI, inflammation scores and ulcer index compared with the IgG treated group. Mice treated with TNFα mab + PF had no ulcers. Multiple injections of TNFα mab or TNFα mab + PF showed greater inhibition in DAI and cytokines in the first two cycles. However, in the third cycle, multiple injections of TNFα mab showed adverse proinflammatory effects.〈section xml:id="abs1-4"〉〈title type="main"〉Conclusion:The simultaneous administration of pentoxifylline and TNFα mab may enhance therapeutic outcomes in inflammatory bowel disease and reduce the side-effects associated with the repeated use of TNFα mab.

    Notes: Spin-polarized intersubband lasers based on optically pumped type-II antimonide quantum wells in a magnetic field are proposed. Complete discretization of the electron energy spectrum is predicted to extend the electron lifetime considerably. Continuous-wave room-temperature operation is projected for both interband and intersubband pumping configurations lasing at λ=16–24 μm. Furthermore, the parasitic capacitances associated with electrical injection are eliminated, and the large differential gain and fast intrinsic time scale should give modulation bandwidths in excess of 100 GHz. © 1999 American Institute of Physics.

    Notes: Electron acceleration by intense laser pulses is studied in the presence of a stochastic field representing a background plasma. Electron distributions are generated peaked in the direction of laser propagation and having a quasi-thermal energy spectrum. Effective temperatures are obtained above the ponderomotive energy. They scale with laser intensity I0 and interaction time t0 proportional to I01/2t0α with α(approximate)0.5−1.0. © 1999 American Institute of Physics.

    Notes: Vortex formation and merging are investigated in the near field of a driven axisymmetric jet. Acoustic forcing is used to obtain repeatable vortex pairing events, and simultaneous passive scalar and cold-chemistry planar laser-induced flourescence are used to obtain instantaneous images of molecularly mixed jet fluid fraction. The time-varying scalar dissipation field and area-averaged stirredness of the vortex core region are measured at various stages of vortex interaction. These mixing properties are analyzed in conjunction with the observed vortex dynamics, such as the time-dependent vortex convection velocity. The results indicate that there are several phases of the pairing event with distinct mixing characteristics, including vortex roll-up, interaction, coalescence, and reentrainment. Vortex roll-up is nearly laminar with molecular diffusion between the layers of jet and co-flow fluid. The most dramatic change in the mixing state of the leading vortex, which includes the appearance of a uniformly mixed core region, occurs as the trailing vortex approaches and interferes with co-flow fluid entrainment. Vortex coalescence is marked by gross deformation and stretching of the trailing vortex, and rapid homogenization of the diffusion layers. Finally, re-entrainment of pure fluid after the pairing event results in an elongated, nonrotating structure. These stages of vortex pairing correspond to the temporal evolution of vorticity observed in previous studies. © 1999 American Institute of Physics.

    Notes: Energy spectra of ions and fast electrons accelerated by a channeling laser pulse in near-critical plasma are studied using three-dimensional (3D) Particle-In-Cell simulations. The realistic 3D geometry of the simulations allows us to obtain not only the shape of the spectra, but also the absolute numbers of accelerated particles. It is shown that ions are accelerated by a collisionless radial expansion of the channel and have nonthermal energy spectra. The electron energy spectra instead are Boltzmann-like. The effective temperature Teff scales as I1/2. The form of electron spectra and Teff depends also on the length of the plasma channel. The major mechanism of electron acceleration in relativistic channels is identified. Electrons make transverse betatron oscillations in the self-generated static electric and magnetic fields. When the betatron frequency coincides with the laser frequency as witnessed by the relativistic electron, a resonance occurs, leading to an effective energy exchange between the laser and electron. This is the inverse free-electron laser mechanism. Electrons are accelerated at the betatron resonance when the laser power overcomes significantly the critical power for self-focusing. © 1999 American Institute of Physics.

    Notes: A technique for simultaneous energy distribution and ion fraction measurements using a linear time-of-flight analyzer with a floatable drift tube is described. Analytical expressions for the relative collection efficiency and viewing region of the apparatus are developed as functions of the analyzed particle reduced energy and dimensionless apparatus parameters. The method was applied to studies of large-angle scattering of singly charged oxygen ions incident on Au(110), and carried out at the Oak Ridge National Laboratory's Multicharged Ion Research Facility. Energy distributions of the scattered projectiles and the negative ion fraction are presented as a function of scattered projectile energy. As a by-product of the measurements, the relative ion detection efficiency of the particle detector was reconstructed as a function of ion impact energy on the detector. © 1999 American Institute of Physics.

    Notes: As the focus of health promotion moves from individuals to organizations, communities and broader social policy, the models that guide public health program planning and development need reexamination. Public health nurses are gaining experience in strengthening and supporting the ability of communities to grow and change. This study aimed to illuminate the process. Data, gathered as part of an action research project to develop individual and community-based strategies to decrease isolation in frail older adults, provided a rich source of qualitative data. Analysis was directed toward identifying the factors and processes of capacity-building. The emerging model was clarified and partially validated with academics and practitioners from health promotion programs across the age span. Four stages of building collective capacity were identified: identifying common ground, working cooperatively, working in partnership, and working across the community. At each stage, processes of relationship building, project management and capacity-building resulted in stage specific products. A model of building collective capacity, grounded in community health practice and supported by the literature provides a base for developing practice indicators, and practice guidelines which will strengthen the ability to reach health goals.

    Notes: Abstract : Basic properties of noradrenaline release were studied in primary cultures of thoracolumbar postganglionic sympathetic neurons taken from 1-3-day-old NMRI mice. After 7 days in vitro, the cultures were preincubated with [3H]noradrenaline and then superfused and stimulated electrically. Conventional trains of pulses (for example, 36 pulses at 3 Hz) as well as single pulses and brief high-frequency trains (for example, four pulses at 100 Hz) elicited a well-measurable overflow of tritium, which was abolished by 0.3 μM tetrodotoxin or omission of Ca2+, but not changed by 1 μM rauwolscine. In trains of one, two, four, six, eight, or 10 pulses at 3 Hz, the evoked overflow of tritium remained constant from pulse to pulse at 1.3 mM Ca2+, but declined slightly at 2.5 mM Ca2+. Tetraethylammonium at 10 mM selectively increased the overflow elicited by small pulse numbers and especially by a single pulse. In trains of 10 pulses delivered at 0.3, 1, 3, 10, 30, or 100 Hz, the evoked overflow of tritium increased from 0.3 to 30 Hz and then declined at 100 Hz. This relationship was particularly pronounced at low Ca2+ concentrations (for example, 0.3 mM) Tetraethylammonium at 10 mM selectively increased the overflow elicited by low frequencies of stimulation. It is concluded that primary cultures of mouse postganglionic sympathetic neurons can be used to investigate release of [3H]noradrenaline. The release is well measurable, even upon a single electrical pulse. It agrees with release in intact sympathetically innervated tissues in a number of fundamental properties, including the pulse number and frequency dependence. The preparation may be of special interest in conjunction with genetic manipulations in the donor animals.

    Notes: Abstract : The possible existence of α2-autoreceptors, P2-autoreceptors, and adenosine A1- or A2A-receptors was studied in cultured thoracolumbar postganglionic sympathetic neurons from mice. The cells were preincubated with [3H]noradrenaline and then superfused. The selective α2-adrenoceptor agonist UK 14,304 reduced the electrically evoked overflow of tritium. When the cultures were stimulated by trains of increasing pulse number, ranging from a single pulse to 72 pulses at 3 Hz, the concentration-inhibition curve of UK 14,304 was shifted progressively to the right and the maximal inhibition obtainable became progressively smaller. Six α-adrenoceptor antagonists shifted the concentration-inhibition curve of UK 14,304 in a parallel manner to the right. Neither ATP (3-300 μM), adenosine (0.01-100 μM), the selective A1-receptor agonist cyclopentyladenosine (1-1,000 nM), nor the selective A2A-receptor agonist CGS-21680 (1-10,000 nM) changed the basal or the electrically evoked overflow of tritium. It is concluded that the cultured neurons possess presynaptic, release-inhibiting α2-autoreceptors. As in intact tissues, the effectiveness of presynaptic α2-adrenergic inhibition depends on the “strength” of the releasing stimulus. The pKD values of the six antagonists against UK 14,304 indicate that the autoreceptors belong to the pharmacological α2D and hence the genetic α2A/D subtype of α2-adrenoceptor. Neither P2-autoreceptors nor receptors for adenosine, the degradation product of ATP, were detected.
