    Notes: Bovine serum albumin (BSA) enhances nicotinic agonist-induced 86Rb+ efflux from synaptosomal fractions of the mouse thalamus, but how it does so is not understood. The experiments reported here indicated that BSA enhancement of nicotinic acetylcholine receptor function was rapid, reversible, depended on BSA concentration, and occurred at all points of the nicotinic agonist concentration-response curve. We hypothesized that BSA-extractable compounds, such as long-chain fatty acids, were responsible for inhibiting nicotinic responses in the absence of BSA. The hypothesis was tested by applying arachidonic, linolenic, or oleic acids in the absence of BSA after an initial prewash with BSA. All three fatty acids exhibited a rapid, concentration-dependent inhibition of nicotinic-agonist stimulated ion flux. Concentration-response curves produced after 30 s of pre-treatment with arachidonic acid were similar to those seen when BSA was completely absent. The effects of pre-treatment were reversed immediately by the introduction of BSA. Furthermore, no effects of fatty acids were observed when preparations were continuously exposed to BSA or when BSA was continuously absent. These results suggest that the removal of endogenous, inhibitory compounds is largely responsible for the rapid, potentiating action of BSA at nicotinic acetylcholine receptors expressed in the mouse thalamus.

    Notes: Background: The significance of serum concentrations of various antibodies and cytokines in the pathogenesis of early-onset periodontitis (EOP) is not well understood. Recent reports suggest differences between young blacks and whites in certain humoral responses, regardless of periodontal status. This study was undertaken to compare the serum concentrations of IgG, IgA, IgM, and IL-1β in EOP subjects with that of healthy controls, and to study the effect of race on these levels.Material and Methods: This case-control study included 228 individuals, 19–25 years old who were selected from a larger population examined in the National Survey of Oral Health of United States Children in 1986/1987. The subjects were classified by their EOP status and they included 166 subjects with EOP and 62 healthy controls. Blood samples were used to assess the serum concentrations of IgG, IgM, IgA, IgG subclass, and IL-1β.Results: The serum concentrations of IgG, IgG subclasses, IgA, and IgM in blacks were not significantly different in the generalized, localized and incidental EOP groups as compared to the healthy controls. The serum IL-1β concentration was slightly and uniformly lower in the EOP groups than in the control group, although not statistically significant. Blacks had significantly higher serum concentrations of total IgG, and of IgG1, IgG2 and IgG3 than whites and Hispanics. Hispanics had significantly higher serum concentrations of IgM and IgG4 than whites and blacks. Hispanics also had a significantly higher serum concentration of IL-1β than blacks.Conclusions: Total antibody response in blacks is not associated with EOP classification. Race has a significant effect on serum antibody concentrations irrespective of disease classification, with blacks having significantly higher serum concentrations of IgG1, IgG2 and IgG3 than whites and Hispanics.

    Notes: The 10 GHz ECR-I ion source at ATLAS is being modified to create a small inner volume within the main plasma chamber to reduce the residual helium background gas load. This effort is part of an experiment to measure the residual concentration of 3He in highly isotopically enriched 4He samples. The modification consists of a new extractor electrode on which is integrally mounted a 2.5-cm-diam quartz tube of approximately 8 cm length. Energizing just the extraction coil produces an ECR only inside the quartz volume. The quartz tube is pumped only through the 1-mm-diam extraction hole. rf power is provided by propagation through the main plasma tank acting as a multimode waveguide. The source must operate in the pressure regime of 10–200 mTorr in order to achieve sufficient sensitivity to measure a 3He/4He ratio of approximately 1×10−15. Results of these tests and the operation of the source in this highly unusual mode will be presented. © 2002 American Institute of Physics.

    Notes: Two techniques for the discrete injection of material into an Electron Cyclotron Resonance ion source (ECRIS) have been developed for the purpose of measuring the ionization and confinement times of ion species. Previously only solid materials in conjunction with a pulsed laser were used in these studies due to the discrete material introduction produced by this configuration. The first method replaces the pulsed laser with a fast high voltage pulse applied to a sputter sample. The high voltage pulse has a rise time of 100 ns, fall time of 80.0 μs, and variable pulse duration. The second method utilizes a fast-pulsed gas valve capable of producing a gas pulse 160 μs in width. These pulse widths are well below the ionization times of the lower charge states and thus allows for time measurements to be made of all charge states. Both of these techniques can be employed to study the effects of rf power, coil configuration, biased disk, and gas mixing on ionization and confinement times. Rise times for neon, argon, and gold will be presented. © 2002 American Institute of Physics.

    Notes: Rotationally resolved S1←S0 fluorescence excitation spectra of 2-chloronaphthalene (2ClN) are measured using a newly constructed ultraviolet (UV) laser/molecular beam spectrometer. More than 1000 well-resolved rotational lines are recorded at a resolution of ∼3 MHz for the two overlapping band origins of the 35Cl and 37Cl isotopomers (Δνorigin∼3 GHz) over a 3.5 cm−1 spectral region at a rotational temperature of 10 K. Both spectra are hybrid bands, having 18% a-/82% b-type character in accordance with 1La←1A1 excitations observed for other 2-substituted naphthalenes. Additionally, Cl nuclear quadrupole splittings are observed as broadened asymmetric line shapes with linewidths from 5 to 20 MHz. Least-squares fits of these line shapes coupled with high-precision Fourier-transform microwave measurements of the ground state have provided accurate S1 quadrupole coupling constants. Specified relative to the principal quadrupole tensor orientation of S0, the S0 and S1 values of eQqzz are −71.198(5) MHz and −67.8(30) MHz, and the asymmetry parameters, η=(eQqyy−eQqxx)/eQqzz, are 0.076 38(5) and 0.13(5), respectively. The experimental line shapes are fit to Voigt functions, yielding 4.9(3) MHz homogeneous (lifetime) and 3.0(2) MHz inhomogeneous (Doppler) contributions. All of the observed structure are well reproduced by more than 20 000 transitions predicted for the nuclear-quadrupole-split rotational spectra of the two hybrid-band origins. Furthermore, the homogeneous linewidths show no dependence on either the upper state energy or angular momentum quantum numbers. The absence of additional level structure at the resolution of 1 MHz or more, from heavy-atom-enhanced intersystem crossing mechanisms, is discussed within the framework of radiationless transition theory. The quadrupole tensor components are used in simple theoretical models to investigate the orbital character of the S1 state and its radiationless decay mechanisms. © 2001 American Institute of Physics.

    Notes: Progesterone receptors play a central role in neuroendocrine and behavioural regulation. To gain insight into the sex- and tissue-specific regulation of progesterone receptors, protein binding on a progesterone receptor-oestrogen response element and mRNA levels for progesterone receptor (PR)-A and PR-B were compared between female and male rats following oestradiol benzoate replacement treatment in hypothalamic and pituitary tissue. Both male and female pituitary protein extracts demonstrated an increase in nuclear protein binding activity to a progesterone receptor-oestrogen response element following oestradiol benzoate treatment. However, there was a greater difference in total binding activity seen in the female pituitary extracts compared to male pituitary protein extracts. In both cases, reflecting the binding data, oestradiol benzoate pretreatment led to an increase in pituitary PR-B messenger RNA, although this increase was significantly larger in females than in males. Oestradiol benzoate treatment also led to a significant increase in specific binding of hypothalamic nuclear proteins to the progesterone receptor oestrogen response element from both females and male hypothalamic extracts. In addition, PR-B messenger RNA was induced by oestradiol benzoate treatment in the female rat hypothalamus, under circumstances where no PR-A could be detected. The male also demonstrated an increase in PR-B messenger RNA following oestradiol benzoate treatment, with undetectable levels of PR-A, although to a lesser degree than that seen in the female. The predominance of PR-B over PR-A messenger RNA in rat hypothalamus and pituitary, and the quantitative differences between female and male rats, could both contribute to the greater responsiveness of female rats to progesterone with respect to control over luteinizing hormone release from the pituitary, and lordosis behaviour regulated by hypothalamic neurones.

    Notes: A high-energy Aptian–Albian platform margin in northern Oman fronted onto an open oceanic basin, making the area a valuable analogue for coeval guyot margins. Most similar aged carbonate margins described in the literature faced either intracratonic or minor oceanic basins. The studied margin is characterized by a stabilized outer rim, which, although it did not rise discernibly above the adjacent lagoonal deposits, flanked a steep upper slope (32–40°) basinwards with a relief of at least 30 m. Two main facies provided the rigidity of the outer margin: Lithocodium boundstones that constituted up to 50% of the rock volume; and marine fibrous cements that occluded up to 35% of primary pore space. In contrast, coral–rudist patches and other shelly sessile benthos were distributed irregularly, and the rudist bioherms of the outer margin were often disrupted, with shells being transported and redeposited. The inner margin is characterized by wedge-shaped storm layers that radiate from the platform top lagoonwards, where they interdigitate with carbonate sands and small rudist bioherms. Polygenetic discontinuity surfaces that bear evidence of both marine hardground and subaerial exposure stages are prominent features of the margin. Throughout the latest Aptian to Middle Albian, the platform succession recorded some 30 relative sea-level falls, of which seven reached amplitudes of many tens of metres. These seven high-amplitude falls in sea level are recorded across the entire south-eastern portion of the Arabian craton and are probably of eustatic origin.

    Notes: Three-spined sticklebacks Gasterosteus aculeatus, from the Chehalis River drainage. Washington, U.S.A., that have lost the mosaic nuptial signal were more likely to be found in tannin stained sub-drainages whereas those expressing the mosaic nuptial signal were more likely to be found in sub-drainages that were not tannin stained. This result is consistent with the sensory drive model.
