    Notes: A flexural plate wave resonator was constructed by patterning current lines on a silicon nitride membrane suspended on a rectangular silicon frame. Eigenmodes of the rectangular membrane were excited using Lorentz forces generated between alternating surface currents and a static in-plane magnetic field. Preferential coupling to a particular membrane mode was achieved by positioning current lines along longitudinal mode antinodes. An equivalent-circuit model was derived which characterizes the input impedance of a one-port device and the transmission response of a two-port device over a range of frequencies near a single membrane resonance. Experiments were performed to characterize the device's response to changes in dc magnetic field strength, ambient gas composition, gas pressure, and input power. © 1998 American Institute of Physics.

    Notes: We assessed the influence of perinatal melatonin on reproductive development and adult responsiveness to melatonin. Testicular growth in an intermediate day length (14 : 10 h light/dark cycle) was substantially reduced in Siberian hamsters gestated by pinealectomised compared to pineal-intact females; gonadal development was normalised in offspring of pinealectomised dams that were pinealectomised at 3–4 days of age. Hamsters deprived of melatonin only during gestation, or both pre- and postnatally, underwent testicular involution during treatment with melatonin in adulthood. Photoperiodic histories acquired prenatally did not endure as long as those acquired by adult hamsters. Hamsters first exposed to melatonin in adulthood were not more proficient in acquiring photoperiodic histories than were normal males. These findings indicate that pre- versus postnatal differences in melatonin signal duration determine rates of testicular development. Exposure to melatonin perinatally does not appear to organise the neuroendocrine substrate that mediates effects of day length and melatonin on the gonads of adult hamsters.

    Notes: When heat treated, amorphous alumino-silicate fibres will devitrify into crystalline phases which on cooling become stable at room temperature. X-ray diffraction has been used to identify and quantify the phases present in such products; mullite and two distinct forms of cristobalite have been observed. One form of cristobalite has been identified as α-cristobalite, the second form has been designated as α′-cristobalite. An internal standard approach was used to enable calibration curves to be obtained for the three crystallite phases, including the α′- cristobalite for which no commercially available standard exists. The accuracy of the methods was estimated as ± 5 wt% for all three crystalline phases present. This was achieved by quantifying the proportion of α′-cristobalite in a standard alumino-silicate fibre after heat treatment for 48 h at 1773 K and then using this material as the `standard' for α′-cristobalite. This `standard' contained 32.9 wt% α′-cristobalite, 1.6 wt% α-cristobalite and 65.5 wt% mullite. The exact nature of the α′- cristobalite phase has also been examined using high-temperature X-ray diffraction and differential scanning calorimetry. It has been shown to be more similar in nature to α-cristobalite than β-cristobalite since it undergoes an α′ → β phase transition on heating. Compared with α-cristobalite it has a different lattice size, and both the temperature and enthalpy of the α′ → β phase transition are lower than for the α → β phase transition. It has been proposed that the α′-cristobalite observed is a defect form of α-cristobalite but with a constant amount of defects/substituted cations which gives it a well defined and consistent structure.

    Notes: Abstract A prominent hypothesis for the function of the glomerular structures in the primary olfactory neuropil of many groups of vertebrate and invertebrate animals is that they enable the processing and coding of information about the chemical compounds that compose complex odors. Previous studies have indicated that various degrees of glomerulus formation in the antennal lobes of the brain of the moth Manduca sexta can be effected by reducing the number of olfactory sensory axons that grow from the antenna into the antennal lobe during metamorphosis. To test the hypothesis that the presence of glomerular structure is necessary to process and identify odors, we substantially reduced, by surgery, the number of antennal segments in developing moths and upon metamorphosis we observed and quantified behavioral responses known to be elicited by odors. Intact and lesioned adult female moths were challenged to fly upwind to the source of an attractive host-plant odor in a wind tunnel. Some of the moths that had developed with reduced olfactory input flew upwind to the odor source. The flight behavior of these individuals was similar to the odor-mediated flight typically observed in moths that had developed normally. Histological analysis of the moths’ antennal lobes revealed that the lobes of more than half of the respondents that had been lesioned during development lacked normal glomerular organization. The neuropil of these abnormally developed antennal lobes was mostly aglomerular, but with a few isolated, clearly abnormal glomerulus-like structures. This suggests either that even a few abnormal glomeruli are sufficient to mediate this specific behavior or that “canonical” glomerular organization per se is not necessary for this odor-mediated behavior.

    Notes: Abstract  A wide variety of masses may be visualized in the posterior knee. Some are encountered incidentally on MRI obtained for seemingly unrelated symptoms; others are palpable and present for MRI characterization. We describe the appearance, significance and pathophysiology of some of the most common lesions found in the popliteal fossa as well as several of the less common entities we have encountered. Thorough understanding of the appearance and location of the more specific masses should augment precise diagnosis in a majority of cases. In turn, this will eliminate unnecessary additional investigations and enable prompt and appropriate treatment.

    Notes: Abstract  Amino-terminal degradation has been observed for many of the secreted heterologous proteins produced by S. lividans 66. We, therefore, set out to characterize the relevant proteinases and their genes. A tripeptide chromogenic substrate was used to identify a gene that was shown to encode a secreted protein which removed tripeptides from the amino terminus of extracellular proteins (tripeptidyl aminopeptidase, Tap; Butler et al. 1995). This activity was removed by a homologous gene deletion replacement and the ability of the S. lividans strain to remove N-terminal tripeptides was greatly reduced, but still significant. When the tap-deleted strain was used as a host for the rescreening of a S. lividans 66 genomic DNA library, a number of other genes encoding proteases with aminopeptidase activities were discovered. One clone (P5-4) produced a 45-kDa secreted protein (Ssp), which showed activity against Ala-Pro-Ala-β-naphthylamide (APA-βNH-Nap) substrate. Further analysis of the cloned DNA showed an open-reading frame encoding a protein larger than 45 kDa. Direct Edman degradation of the secreted protein confirmed that it was encoded within the cloned DNA and probably processed from a larger precursor. Protein sequence analysis revealed a striking homology to subtilisin BPN′ in three regions around the active-site residues suggesting that the protein is a serine protease. As expected, the protease activity was inhibited by phenylmethylsulphonyl fluoride. Mutant strains with most of the ssp gene deleted exhibited reduced activity against APA-βNH-Nap substrate compared to their non-deleted parental strains.

    Notes: Abstract Nuclear counts determined by crystal violet staining from samples of stationary or microcarrier cultures of hybridomas, CHO or Vero cells were consistently and significantly higher than cell concentrations determined by the trypan blue or Coulter counter methods. This difference was attributed to the presence of a significant proportion of binucleated cells, which are assumed to be 35% of the cell population in the stationary phase of Vero cultures. The proportion of such cells during exponential growth was variable. However, continuous sub-culture of these cells induced a degree of synchrony during growth which resulted in a cyclic variation of the difference between the cell and nuclei counting techniques. This data indicates that care should be taken in interpreting cell culture profiles based solely on crystal violet nuclei staining counts.
