    Notes: Abstract— The β-adrenergic agonist, isoproterenol and the α- and β-adrenergic agonist. NA. raise the intracellular concentration of cyclic AMP in cultures of dissociated perinatal mouse brain. This rise is prevented by a β- but not by an α-adrenergic antagonist. The maximal level of cyclic AMP reached in the presence of isoproterenol is markedly higher than that found after exposure to NA. However, if NA is used along with an α-adrenergic antagonist, cyclic AMP levels as high as those after isoproterenol are measured. Agonists with α-adrenergic activity including NA decrease the response to isoproterenol. The decrease is blocked by α-adrenergic antagonists. From this and additional evidence it is concluded: (1) The increase in the level of cyclic AMP caused by β-adrenergic agonists is due to β-receptor-mediated stimulation of adenylate cyclase; (2) the inhibition of this effect by α-adrenergic agonists is mediated by adrenergic α-receptors; (3) the α- and β-adrenergic receptors are likely to be located on the same cells, probably the most abundant putative glial precursor cells. The simultaneous stimulation of α- and β-adrenergic receptors on the same cell may be of significance in the regulation of the response to NA.

    Notes: Two novel interleukin-24 (IL-24) splice variants were identified in normal human melanocytes by sequencing cloned polymerase chain reaction (PCR) products that are not expressed in metastatic melanoma. These gene products have been generated by differential skipping of exons 3 (IL-24 delE3) and 5 (IL-24 delE5). IL-24 delE3 has limited sequence identity to the IL-24-interacting protein mda-7s, and IL-24 delE5 is homologous to IL-24.

    Notes: The study was undertaken to determine the amount and nature of condensed tannins in rapeseed meal. No condensed tannins were found in rapeseed meats (endosperm plus germ). The hulls, however, yielded 0.1% condensed tannins by 70% aqueous acetone extraction, the bulk of the condensed tannins in the hulls being unextractable by the extractants used. The basic unit of the isolated polymeric flavanols appeared to be leucocyanidin.

    Notes: Background : Local treatment with foams in patients suffering from ulcerative proctitis or proctosigmoiditis is considered a rational treatment option.Aims : To investigate colonic spread, safety, tolerability and acceptance of a newly developed budesonide foam formulation.Methods : Twelve patients (four females, eight males) with acute proctosigmoiditis or left-sided ulcerative colitis were rectally administered a single dose of [99Tcm]-labelled budesonide foam (Budenofalk; Dr Falk Pharma GmbH, Freiburg, Germany) containing 2 mg budesonide in 20 mL foam after diagnostic colonoscopy. Thereafter, the colonic spread was assessed by means of γ-scintigraphy for 6 h. Serum samples were taken simultaneously.Results : Budesonide foam spread with a maximum between 11 and 40 cm, thus reaching the sigmoid colon in all patients. In some patients, the foam even extended into the distal third and the middle of the descending colon with maximum radioactivity at 4 h. Systemic budesonide absorption was rapid and pharmacokinetic data were comparable with published data on marketed budesonide enemas, with mean serum Cmax and AUC0-8h values of 0.8 ± 0.5 ng/mL and 3.7 ± 1.9 ng h/mL, respectively. The new formulation was well accepted by all patients, who could retain the foam for at least 4 h.Conclusions : In the majority of patients, budesonide foam effectively spread up to the left-sided colon and thus qualifies for the local treatment of proctosigmoiditis.

    Notes: Neutron diffraction has been used to study the structural disorder in γ-CuCl at 25 and 366°C. Powder patterns of 63CuNCl, NCuNCl and 65CuNCl (N= natural abundance) were treated by integrated intensity (II) and radial density (RD) analyses. Using II, a satisfactory fit to these 366°C data was made with the anharmonic model described by Sakata, Hoshino & Harada [Acta Cryst. (1974). A30, 655-661]. This fit resulted in an anharmonic parameter βCu = 1.2 ± 0.6 × 10-12 erg Å-3, which is in excellent agreement with the value of βCu = 1.15 ± 0.66 × 10-12 erg Å-3 obtained from a single-crystal study at room temperature by Sakata et al. (1974). To eliminate the indistinguishability of the anharmonic and statistically disordered models inherent in II, an energy analysis was made of the diffuse scattering. It was found that at 25 °C, 50 to 70% of the diffuse scattering is elastic. The non-monotonic κ dependence of this elastic diffuse scattering implies a model of correlated static displacements of Cu atoms and is inconsistent with the anharmonic model. Radial density analyses of the three spectra were fitted to a model based on harmonic vibration to obtain partial radial densities QCuCu,QClCl and QCuCl,. Although theQClCl, was typical of that expected for a solid structure, QCuCu,, at 366°C was found to be more 'liquid-like'. Quantitative conclusions from this radial density analysis are limited by the strong dependence of the results on instrumental resolution. An estimate of the effect of instrumental resolution on the measured radial density function is contained in Appendix B.

    Notes: The superconductor Th4D15 has been investigated by neutron diffraction with the spallation neutron source (ZING-P) at Argonne National Laboratory. Profile refinement of the powder data for d spacings between 0.65 and 1.95 Å, corresponding to a neutron wavelength range of 1.26–3.77 Å, confirms the structure suggested by Zachariasen [Acta Cryst. (1953), 6, 393–395]. The b.c.c. cell, I\bar 43 d (a0 = 9–11 Å), of Th4D15 contains 16 Th atoms at 16(c) [xTh = 0.2066 (3)] and 60 D atoms at 12(a) and 48(e) [x = 0.3706 (7), y = 0.2155 (7), and z = 0.4048 (7)]. The Th atoms are equally spaced (3.95 Å) along four non-intersecting chains that are parallel to the four body diagonals. The general position D atoms, 48(e), form small equilateral triangles along the chains approximately equidistant from each Th atom. Neighboring triangles along a chain are rotated ±19.5 (2.0)° with respect to each other. The D atoms at 12(a) are centered in near-regular tetrahedra of Th atoms, consisting of one member from each chain and sharing only corners with adjacent tetrahedra. Each Th atom is found in a cage of three 12(a) plus nine 48(e) D atoms. Results for Debye–Waller factors of the Th and D atoms at 295 and 92 K are presented and compared with values obtained for the Debye–Waller factors in Pd–H.

    Description / Table of Contents: Summary In peripheral human blood lymphocytes the uptake and metabolism of adenine, guanine, and hypoxanthine was investigated. This was achieved by incubation of purified lymphocytes with14C-purine bases, separation of cells from the incubation medium by a rapid filtration technique, and subsequent separation of the acid soluble material by thin-layer chromatography. No preferential uptake for one of the purine bases was observed. In all cases only traces of14C-purine bases not added originally and labeled nucleosides could be demonstrated. Approximately 2/3 of adenine and 1/2 of guanine or hypoxanthine were converted to nucleotides. Separation of formed nucleotides showed that adenine and guanine were metabolized mainly to their corresponding nucleotides; hypoxanthine was converted to a considerable amount to adenine nucleotides and only to a small proportion into its own nucleotides. These results demonstrate the predominance of adenine nucleotide formation in normal human lymphocytes.
    Notes: Zusammenfassung In peripheren menschlichen Lymphozyten wurden die Aufnahme und der Stoffwechsel von Adenin, Guanin und Hypoxanthin untersucht. Die gereinigten Lymphozyten wurden mit14C-markierten Purinbasen inkubiert und anschlie\end vom Inkubationsmedium mittels rascher Filtrationstechnik getrennt. Eine bevorzugte Aufnahme einer der Purinbasen konnte nicht beobachtet werden. Die dünnschichtchromatographische Auftrennung der säurelöslichen Metabolite ergab kaum eine Bildung von Nukleosiden und anderen als den zugesetzten Purinbasen. Adenin wurde zu ca. 2/3, Guanin und Hypoxanthin hingegen zu etwa der Hälfte zu Nukleotiden aufgebaut, wobei Adenin und Guanin hauptsächlich zu ihren eigenen, Hypoxanthin jedoch zu Adeninnukiektiden metabolisiert wurde. Die Ergebnisse weisen auf die bevorzugte Bildung von Adeninnukleotiden in normalen menschlichen Lymphozyten.
