    Notes: We have proposed an equation for estimating the real-time mixed venous blood concentration (MVBC) of isoflurane in cardiac anaesthesia. However, information related to the application of our method to sevoflurane is lacking. We studied 12 patients undergoing cardiac surgery and anaesthetised with sevoflurane. At different time points, pulmonary arterial blood samples were collected for gas chromatography-mass spectrometry (GC-MS) to determine the real mixed venous concentrations of sevoflurane. The inspired and expired concentrations of sevoflurane, measured by a gas monitor, were used for the MVBC calculations. Using Bland-Altman analyses, we found that the calculated MVBCs accurately represent the actual concentrations of sevoflurane in pulmonary arterial blood, as shown by a near-zero percentage bias with a 0.14% precision between the two concentrations. The results demonstrated that our equation could be a useful method for estimating the pulmonary blood concentration of sevoflurane.

    Notes: Background:  Forcipomyia taiwana is a tiny, blood-sucking midge that cause intense pruritis and swelling in sensitive individuals. It is distributed island-wide in rural Taiwan and Southern China.Objective:  This study aimed to study the allergic immune responses and identify F. taiwana allergens.Methods:  Crude whole body F. taiwana extracts were prepared with phosphate-buffered saline. The specific IgE antibody was determined by enzyme-linked immunoassay and immunoblotting. Protein was analyzed by electrospray ionization tandem mass spectrometry.Results:  Among the 372 subjects that were exposed to F. taiwana bites, 179 (48%) reported an immediate skin reaction with/without delay reaction and 41(11.1%) reported a solely delay reaction. The skin of 21 subjects was tested with F. taiwana extract. Of these 21 subjects, 12 (57.1%) produced immediate skin reactions and contained high levels of specific IgE antibody against F. taiwana. Immunoblotting revealed that 11 allergenic components are able to bind specific IgE. Allergens of 22, 24, 35, 36, and 64 kDa bound 50, 50, 75, 66.7, and 75% of IgE-containing sera tested, respectively. Tryptic fragments of the 24, 35, 36, and 64 kDa allergens were analyzed by ESI-MS/MS. Selected tryptic peptides of 24, 35, and 36, and 64 kDa allergens exhibited significant sequence identity with triosephosphate isomerase of Anopheles merus,Tenebrio molitor,Ochlerotatus togoi, and Chrysops vittatus, fructose 1,6-bisphosphate aldolase of Antheraea yamamai and Homalodisca coagulata, and a slow muscle myosin S1 heavy chain of Homarusamericanus and a protein with unknown function from A. gambiae, respectively. The 35 and 36 kDa proteins may represent different isoforms of the fructose 1,6-bisphosphate aldolase.Conclusion:  We conclude that immediate reaction to F. taiwana bites is IgE mediated and the 24 (For t 1), 35 (For t 2), and 64 kDa (For t 3) proteins are candidates for major F. taiwana allergens. Further studies are needed to confirm these allergens.

    Notes: Background  Tacrolimus (FK506) ointment has been used for treatment of inflammatory dermatoses with remarkable success. Our previous studies have indicated that direct modulation of tacrolimus on keratinocytes (KCs) may have an impact on its therapeutic effect. The use of monoclonal antibody specific for tumour necrosis factor (TNF)-α has shown efficacy in treating both psoriasis and Crohn disease. Topical tacrolimus has also been shown to be effective for treating cutaneous manifestations of both diseases.Objectives  To explore the effects of FK506 on human KCs in terms of TNF-α secretion and to investigate the regulatory pathway involved.Methods  Ultraviolet (UV) B-irradiated cultured KCs were treated with various concentrations of FK506. At indicated time points after UVB irradiation we determined: (i) the TNF-α concentrations present in the culture supernatants; (ii) the activation and translocation of nuclear factor (NF)-κB in the cell nucleus; and (iii) the protein expressions of IκB kinase (IKK) and IκB in the cell lysates. In addition, a mouse model was used to corroborate our in vitro findings in vivo. More specifically, topical tacrolimus was applied on to mouse skin unilaterally after UVB irradiation. The effects of FK506 on nuclear NF-κB expression of UVB-irradiated mouse skin were determined.Results  Our results showed that FK506 dose-dependently downregulated the secretion of TNF-α from UVB-irradiated KCs. The activation and translocation of NF-κB in UVB-irradiated KCs were also dose-dependently suppressed by FK506. The degradation of IκB induced by UVB was also inhibited by FK506, while no change in IKK expression was noted regardless of UVB and FK506 treatment. Murine skin biopsies showed that nuclear NF-κB expression induced by UVB was inhibited by topical tacrolimus treatment.Conclusions  Our results indicate that FK506 inhibits TNF-α secretion in human KCs via direct regulation of NF-κB. This modulatory effect of FK506 on KCs offers a possible mechanism for how topical tacrolimus regulates cutaneous inflammatory conditions.

    Notes: Background  Vitiligo is an acquired pigmentary disorder characterized by depigmentation of skin and hair. As the pathogenesis of this disease is still obscure, the treatment of vitiligo has generally been unsatisfactory and often disappointing. Topical tacrolimus (FK506) ointment has recently been added to the armamentarium against this pigmentary disorder. Despite its clinical efficacy, the underlying mechanisms of how topical tacrolimus induces repigmentation in vitiligo have rarely been investigated. As tacrolimus ointment is applied directly to the skin, its impact on keratinocytes (KCs) requires thorough investigation.Objectives  To investigate the effects of FK506 on melanocyte (MC) and melanoblast (MB) growth via KCs.Methods  Cultured MCs and MBs were treated with supernatant of KC cultures conditioned with various concentrations of FK506. The impact of supernatant on MCs and MBs was assessed in terms of its effect on MC/MB proliferation, melanin formation and cell migration. The activities of matrix metalloproteinase (MMP)-2 and MMP-9, known for their influence on cell migration, were evaluated. The concentrations of MC/MB growth factors in the KC supernatant were also determined.Results  Results demonstrated that proliferation of both MCs and MBs was significantly enhanced by FK506-treated KC supernatant. In addition, the concentration of stem cell factor in KC supernatant increased dose-dependently with FK506 treatment. The supernatant from FK506-treated KC culture showed a significant increase in MMP-9 activity.Conclusions  Our study provides in vitro evidence demonstrating that direct interaction between FK506 and KCs creates a favourable milieu for MC growth and migration. Furthermore, our findings provide a possible mechanism explaining how tacrolimus ointment induces repigmentation in patients with vitiligo.

    Notes: Currently, there are adequate guidelines available for FRP retrofitting RC structures against static and seismic loads. However, there is still limited information on retrofitting RC structures against short-duration dynamic loading effects such as blast loading. Due to the increasing threat of terrorism in recent years, retrofitting of RC structures against blast loading is of paramount importance in structural engineering. In this paper, a dynamic model that is based on single-degree-of-freedom (SDOF) approach is developed for the analysis of the response of retrofitted fixed end supported RC slabs subjected to blast loads. A previously validated layered capacity analysis method is used to determine the yielded and ultimate blast resistant capacity of a cross-section of a RC slab which allows varying strain rates with time along the depth of the member. The corresponding deflections are determined by plastic hinge analysis. To simplify the calculation process, a tri-linear resistance-deflection function which consists of elastic, elasto-plastic and plastic region for fixed end supported RC slabs is converted to an equivalent bilinear function. This developed model can adequately predict the retrofitted members’ response to blast loading. It is then is used to conduct a parametric study to optimise the retrofitting of RC slabs subjected to blast loading by varying the quantity, material type and technique of retrofitting

    Notes: Using environment-friendly cutting fluid in the process of metal cutting is one of the validpaths to carry out the green manufacturing. Using metacrylic acid ester, castor oil and boride as themain raw materials, a new type of environment-friendly water-base cutting fluid had been obtained.The main components of this cutting fluid are some kind of high molecular polymer and some kind ofB-containing castor oil EP agents. The test results show that the cutting fluid has good stability,antirust property and biodegradability. The maximal non-chucking load value (PB) of the cuttingfluid, whose consistency is 5%, can be 1068N. In the comparison with dry cutting, cutting force usingenvironment-friendly cutting fluid is reduced by 23% in a certain condition. Using this cutting fluidcan raise stock-removing efficiency and cutting quality, and extend the service life of cutting-tool.And this cutting fluid is harmless to operators and free from pollution to the environment. Expansionand application of the environment-friendly cutting fluid are advantageous to promote thedevelopment of the green manufacturing

    Notes: This paper presents that the high-speed steel tools with solid lubrication material coatingcan cut without fluid because it has the quality of self-lubrication. With the help of M-2 friction andwearing testing machine and KYKY2800 scan electron microscope, the coating preparationtechnology and mixture ratio of solid lubrication material in complex coating can be optimized. Theresults show: phosphate treatment can increase the power of the tools’ surface adsorbing solidlubricant, and the frictional behaviour of solid lubrication coating is excellent when surfaceroughness of phosphate coating is Ra 2μm~Ra 4μm; molybdenum bisulfide, graphite and trioxidaluminium are mixed together and form a kind of complex coating which has better antifrictioneffect. The cutting test shows that the solid lubricant coating on the high-speed steel tools can behelpful in increasing the service life of the tools, decreasing the cutting resistance, and reducing thechip deformation and surface roughness of finished work
