    Notes: Cerebellar long-term depression (LTD), a candidate cellular mechanism of motor learning, is induced by conjunctive activation of parallel fibres and a climbing fibre. Previous studies have shown that combinatorial application of high potassium and glutamate (K/glu) to cultured cerebellar neurons can mimic this conjunctive stimulation of presynaptic fibres and induces the LTD of miniature excitatory postsynaptic current (mEPSC) amplitudes lasting for more than 24 h. The late phase of this LTD (LLTD, 〉 3 h) depends on de novo transcription induced by prolonged conditioning. Here, the role of Calcineurin in the LLTD induction was examined. Application of a Calcineurin inhibitor FK506 mimicked the effect of K/glu-treatment by decreasing mEPSC amplitudes for more than 24 h. FK506-induced depression, as well as the K/glu-induced LLTD, was blocked by inhibitors of either mRNA synthesis or Ca/Calmodulin dependent kinase. In addition, the FK506-induced depression and K/glu-induced LLTD occluded each other, suggesting that they share the same mechanism. On the other hand, misexpression of the constitutively active form of Calcineurin in the Purkinje neuron nucleus blocked the LLTD induction by the K/glu-treatment. These results suggest that Calcineurin is involved in the induction of LLTD as a negative regulator. Furthermore, it was found that trapping superoxide, which is increased by neuronal activity and inactivates Calcineurin, suppressed the LLTD induction. Taken together, these results suggest that the LLTD might be induced by down-regulation of Calcineurin activity through superoxide in cultured Purkinje neurons.

    Notes: Abstract The Ryoke Belt in the Ikoma Mountains, Nara Prefecture, Japan, is composed mainly of various granitic, intermediate and gabbroic rocks. Igneous activity in this area is divided into two periods, early–middle Jurassic and late Cretaceous, based on isotopic dating. The intermediate plutonic rocks in the Fukihata area are composed of two rock types: Kyuanji quartz diorite and Fukihata tonalite. Rb–Sr whole-rock isochron ages have been determined for both plutonic rocks. Their ages and initial 87Sr/86Sr ratios are as follows: the Kyuanji quartz diorite has an age of 161.0 ± 17.9 Ma with an initial 87Sr/86Sr ratio of 0.70727 ± 0.00007, while the Fukihata tonalite has an age of 121.4 ± 24.6 Ma with an initial 87Sr/86Sr ratio of 0.70753 ± 0.00020. Our chronological results indicate that the Kyuanji quartz diorite belongs to the Jurassic mafic rocks, such as the Ikoma gabbroic mass, while the Fukihata tonalite belongs to the early Cretaceous granitic rocks. Both these intermediate plutonic rocks have different chemical characteristics and were derived from different magmas.

    Notes: Abstract  Autoimmune diseases involve multiple genes. While functions of these genes are largely unknown, some may be related to an intrinsic hyperresponsiveness of B cells. B-cell responses are controlled by signaling thresholds through the B-cell antigen receptor (BCR) complex. The B1 isoform of type II IgG Fc receptors (FcγRIIB1) is exclusively expressed on B cells and serves as a negative regulator for inhibiting BCR-elicited activation. Thus, its allelic variants associated with functional deficits could be examined for possible associations with susceptibility to autoimmune diseases. We found that there are three types of polymorphisms in the reported FcγRIIB transcription regulatory regions in mouse strains. Compared to normal healthy mouse strains (group III), autoimmune disease-prone strains (group I) share three deletion sites: two in the promoter region and one in the third intron. Strains (group II) that per se are not autoimmune-prone, but have potentials to accelerate autoimmune diseases share two deletion sites in the third intron: one identical to that in group I and the other unique to group II. These polymorphisms correlated well with extents of down-regulation of FcγRIIB1 expression in germinal-center B cells upon stimulation with antigens and up-regulation of IgG antibody responses. Our data imply that these FcγRIIB polymorphisms are selected evolutionarily for natural defense against pathogens, and that such polymorphisms may, in turn, form the basis of one aspect of autoimmune susceptibility.

    Notes: Abstract The immune response to bovine insulin (BI) in the rat is controlled by the major histocompatibility complex (Mhc)-linked immune response gene (Ir-BI) and immune suppression gene (Is-BI). In the present study, we investigated the low responsiveness to BI in the WKAH rat (RT1 k ) and attempted to explore the functional link between Is-BI and Mhc class II molecules. Lymph node cells (LNC) from the low responder (WKAH) rats responded well to BI when a large amount of antigen was added to the culture in vitro or after OX8-bearing (OX8+) T cells were eliminated. These LNC, after the elimination of OX8+ cells, could show the RT1.Dk-restricted proliferative response upon in vitro challenge with BI, BI-B chain, or pork insulin. In addition, OX8+ T cells, which were activated with BI and antigen-presenting cells (APC) in vitro, suppressed the anti-BI response of W3/25-bearing proliferating T cells from BI-immunized rats. The results have demonstrated that proliferating T-cell repertoires do exist to BI, which recognize BI-B chain in the context of RT1.Dk molecules in the WKAH rat, and that the state of low responsiveness is mediated to a great extent by antigen-specific OX8+ suppressor T (Ts) cells. Furthermore, the elimination of APC or the addition to RT1.Bk-specific monoclonal antibody in the in vitro secondary activation culture of Ts cells diminished the suppressive activity of OX8+ Ts cells. In the induction phase of Ts cells it therefore seems to be necessary for these cells to recognize BI together with RT1.Bk molecules on APC.

    Notes: Abstract Ionic transport properties of perovskite-type oxides based on LnScO3, Ln1- xCaxScO3-α (Ln=La, Nd, Sm and Gd) and LaSc1-xMgxO3-α were studied using an electrochemical method at elevated temperatures. Conductivity in these oxide systems increased by more than three orders of magnitude upon doping with divalent atoms such as Ca or Mg. However, when x≥0.1, conductivities are almost independent of x in La1- xCaxScO_3-α and LaSc1- xMgxO3-α. Gd0.9Ca0.1ScO3-α showed lower conductivity than Ln1-xCaxScO3-α (Ln=La, Nd and Sm). Protonic conduction in these oxides under hydrogen containing atmospheres was confirmed by emf measurements of hydrogen concentration cells and by electrochemical hydrogen pumping using these oxides as a diaphragm. In LaSc1-xMgxO3-α the transport number of protons under hydrogen containing atmosphere was unity, suggesting that electronic conduction never became dominant, even under strong reducing conditions. Under condition of high oxygen partial pressure, the transport number of ions was less than 0.1, suggesting that the majority conductive carriers under such conditions were holes.

    Notes: The activated sludge that had been sufficiently cultivated with DMF-containing waste water was entrapped and immobilized in spherical poly(vinyl alcohol) (PVA) hydrogel particles. Features of numbers and distribution of bacteria in the resultant PVA gel were estimated. The DMF-decomposing bacteria named Bacillus cereus D-1 was isolated. Several morphological and physiological responses of the bacteria were revealed. Spherical PVA gel with the DMF-decomposing bacteria prepared through of freezing and thawing was tested for treatment of DMF-containing effluent. Test results have shown the ability of stable operation and maintenance at the same capacity of 1 kg/m3/day by both a basic study and a bench plant test. © 1995 John Wiley & Sons, Inc.

    Notes: The structures and properties of poly(viny1 alcohol) (PVA) hydrogel prepared by the freezing and thawing method and poly(ethy1ene glycol) and polyacrylamide hydrogels prepared by polymerization were compared in view of immobilizing microorganisms. Observation in a scanning electron microscope and measurement of the physicochemical properties revealed that the frozen PVA gel is superior in water content and oxygen permeability than the other gels. The PVA hydrogel has a high tensile and folding strength and good handleability and processability. The durability against abrasion and chemical resistance of PVA gel were also studied. Activated sludge was immobilized and fixed on PVA gel particles and incubated. This gel had a treating capacity for synthetic sewage of about two to three times that with the standard activated sludge process. © 1995 John Wiley & Sons, Inc.
