    Notes: Polyamines are a ubiquitous group of amines that play diverse biological roles. In the anterior pituitary, intracellular polyamine levels are reported to show diurnal changes, although the biological significance remains to be elucidated. In this study, we examined the effects of polyamines on the transcriptional activity of the rat pro-opiomelanocortin (POMC) gene using AtT20PL, a clone of the AtT20 cell line in which an approximately 0.7 kb of the rat POMC 5′ promoter-luciferase fusion gene was stably incorporated. The results showed that three representative polyamines (putrescine, spermidine and spermine) all stimulated POMC promoter activity in a time- and dose-related manner, spermine showing the most potent effect (maximum approximate three-fold increase). This effect was not observed under treatment with actinomycin D, suggesting the effect of polyamine at the transcriptional level. On the other hand, methylglyoxal bis (guanylhydrazone), an inhibitor of polyamine synthesis, showed the opposite effect, further supporting the positive role of intracellular polyamines. Taken together, our findings suggest that polyamines are involved in the regulation of POMC gene expression (especially in terms of diurnal changes) in corticotroph cells. The precise molecular mechanisms of polyamine effects await further research.

    Notes: Background Collagen type I is one of the major deposits in thickening of the reticular basement membrane of asthma.Objective and Methods In this study, we assessed turnover of collagen type I in asthma by measuring procollagen type I C-terminal peptide (PICP) and collagen type I C-terminal telopeptide (ICTP) in induced sputum.Results PICP but not ICTP was found to be significantly higher in asthma subjects than in normal volunteers (P 〈 0.05). In asthma, PICP was inversely correlated with %FEV1.0 (r = −0.539), and its levels significantly increased upon exacerbation (P 〈 0.05), indicating that collagen synthesis increases during asthma exacerbation. Additionally, PICP was found to significantly correlate with eosinophil counts in sputum (r = 0.539), indicating that eosinophils stimulate collagen turnover. Because eosinophils can produce TGF-β, a potent stimulator of collagen synthesis, we immunocytochemically examined TGF-β-positive cells in sputum. TGF-β-positive cells significantly correlated with eosinophil counts (r = 0.811) and PICP (r = 0.569), suggesting that TGF-β released from eosinophils is involved in collagen synthesis.Conclusions The results of the present study suggest that collagen synthesis is stimulated in asthmatic airways by eosinophils through TGF-β, while collagen degradation is not, and that PICP in sputum can act as a new marker for airway inflammation in asthma.

    Notes: Abstract  Fifteen strains of aerobic bacteria and two actinomycetous strains were isolated from various soil samples, using cis-propenylphosphonic acid as the sole source of carbon. Taxonomic studies showed that these belonged to the Pseudomonas, Alcaligenes, Flavobacterium, Aeromonas, Corynebacterium and Streptomyces genera. Most of these strains produced the antibiotic fosfomycin [(—)-(1R, 2S)-1,2-epoxypropylphosphonic acid] from cis-propenylphosphonic acid by stereoselective epoxidation. Under the culture conditions tested, the production of fosfomycin reached a level of 2.0 mg/ml and 0.75 mg/ml by F. esteroaromaticum IFO 3751 and P. putida IK-8 respectively. Purified fosfomycin was an active stereoisomer and its optical purity was a little higher than that of the synthetic compound. Although the stereoespecificity of each enzyme participating in the epoxidation reaction has not yet been clarified, the epoxide formation proceeded through a bromohydrin intermediate produced by bromoperoxidase, finally yielding fosfomycin by the action of halohydrin epoxidase.

    Notes: Abstract. We evaluated the apoptosis of neutrophils during the resolution of acute pulmonary inflammation induced by exposure to ozone. The inflammatory response was assessed in rat lungs 0, 1, 3, and 7 days after 4-h exposure to air or 2 ppm ozone. Analysis of bronchoalveolar lavage fluid demonstrated significant increases in albumin concentrations on days 0 and 1 and in the number of lavageable neutrophils on days 0, 1, and 3, indicating the presence of acute pulmonary inflammation. These parameters returned to control values on day 7, which suggests that the acute pulmonary inflammation induced by ozone was reversible. On days 1 and 3, but not on day 0, the neutrophils showed morphologic evidence of apoptosis. Based on morphologic analysis, the proportion of apoptotic neutrophils was 23.3 ± 2.2% on day 1 and 55.7 ± 3.2% on day 3. Terminal deoxynucleotidyl transferase-mediated dUTP end labeling (TUNEL), in contrast, revealed that the proportion of apoptotic cells was 59.7 ± 9.1% on day 1 and 68.0 ± 4.3% on day 3. On day 3, light microscopy and electron microscopy demonstrated engulfment of the neutrophils by macrophages. These findings indicate that the apoptosis of neutrophils followed by their engulfment by macrophages contributes to the clearance of neutrophils from the sites of inflammation. Moreover, TUNEL detected apoptotic neutrophils with greater sensitivity compared with morphologic assessment.

    Notes: Abstract We investigated the quantitative expression of the human glucose-6-phosphate translocase gene (G6PT1) and its splicing variants in human tissues. The G6PT1 gene was strongly expressed in liver, kidney and haematopoietic progenitor cells, which might explain major clinical symptoms such as hepatomegaly, nephromegaly and neutropenia in glycogen storage diseases type Ib. Reverse transcriptase-mediated PCR amplification of G6PT1 cDNA revealed several splicing variants in tissue-specific manners. The brain-specific isoform, which has an additional 22 amino acids between exons 6 and 8, was also identified in heart and skeletal muscle. A new splicing variant, although less prominent in quantity and lacking polypeptide loops corresponding to exons 2 and 3, may have a distinct substrate affinity or specificity in leukocytes and haematopoietic progenitors. In conclusion, the G6PT1 gene was expressed in various tissues, and alternative splicing variants exist in tissue-specific manners.

    Notes: Abstract. We studied the effects of ebselen on rat lung inflammatory responses against ozone exposure. Rats were treated with ebselen every 12 h from 1 h before a single 4-h exposure to 2 ppm ozone. Treatment with ebselen (10 mg/kg) significantly decreased pulmonary inflammation as indicated by the albumin concentration and the number of neutrophils in the bronchoalveolar lavage fluid 18 h after the ozone exposure. Although treatment with ebselen did not alter the macrophage expression of inducible nitric oxide synthase after the ozone exposure, it did markedly inhibit the nitration reaction of tyrosine residues, suggesting that ebselen scavenges peroxynitrite during ozone-induced pulmonary inflammation. Treatment with ebselen also enhanced the pulmonary expression of both copper, zinc, and manganous superoxide dismutases at the same time point. These enzymes may also contribute to a decrease in the formation of peroxynitrite by lowering the concentration of superoxide. Thus, ebselen represents a useful compound for protecting against certain acute lung injuries by modulating the oxidant-related inflammatory process.

    Notes: Summary A method of supercritical fluid extraction (SFE) of triazine herbicides from soil has been developed. The recoveries of triazine herbicides increased with increase in both extraction pressure and temperature when supercritical carbon dioxide was used as the extraction medium alone. Because the recovery by SFE was influenced by the concentration of triazine herbicides in the soil matrix, the addition of methanol to carbon dioxide as a polar modifier was required for the satisfactory extraction of trace levels of the content of the herbicides. The moisture content of the soil also affected the recoveries by SFE. SFE was applied to the analysis of real samples. The results by SFE were almost the same as those from soxhlet extraction.

    Notes: Abstract Spinel iron oxide (Fe3O4-γ-Fe2O3) particles were supported on microbeads of silica gel by the calcination of the silica gel base adsorbing citric acid and Fe3+ ions. The X-ray diffraction patterns and the57Fe Mössbauer spectra measured for the spinel iron oxide indicated that the particle size of the oxide was regulated by the mean pore diameter (4–82 nm) of the silica gel support employed. In the case of α-Fe2O3 particles prepared by using the same silica gel beads, it was revealed by the Mössbauer spectra and the electron micrographs that there were relatively large particles of the oxide on the surface of the beads, in addition to the particles in the silica gel micropores.
