    Notes: In the E-157 experiment now being conducted at the Stanford Linear Accelerator Center, a 30 GeV electron beam of 2×1010 electrons in a 0.65-mm-long bunch is propagated through a 1.4-m-long lithium plasma of density up to 2×1014 e−/cm3. The initial beam density is greater than the plasma density, and the head of the bunch expels the plasma electrons leaving behind a uniform ion channel with transverse focusing fields of up to several thousand tesla per meter. The initial transverse beam size with σ=50–100 μm is larger than the matched size of 5 μm resulting in up to three beam envelope oscillations within the plasma. Time integrated optical transition radiation is used to study the transverse beam profile immediately before and after the plasma and to characterize the transverse beam dynamics as a function of plasma density. The head of the bunch deposits energy into plasma wakes, resulting in longitudinal accelerating fields which are witnessed by the tail of the same bunch. A time-resolved Cherenkov imaging system is located in an energy dispersive plane downstream of the plasma. It images the beam onto a streak camera allowing time-resolved measurements of the beam energy spectrum as a function of plasma density. Preliminary experimental data from the first three runs are compared to theory and computer simulations. © 2000 American Institute of Physics.

    Notes: Abstract: Rat PC-12 pheochromocytoma cells respond to stimulation with bradykinin, angiotensin II, and carbachol with an increased formation of labeled inositol phosphates after preincubation of the cells with [3H]inositol. Li+ potentiates greatly the agonist-induced increase in amount of nositol mono-, bis-, and trisphosphate but not the increase in amount of inositol tetrakisphosphate. Separation of the isomers of inositol trisphosphate shows that the lithium-induced increase in amount of inositol trisphosphate is due to potentiation evoked by lithium of the accumulation of inositol-1,3,4-trisphosphate.

    Notes: Introduction: The sterol carrier protein-2 gene encodes two functionally distinct proteins: sterol carrier protein-2 (SCP2, a peroxisomal lipid carrier) and sterol carrier protein-x (SCPx, a peroxisomal thiolase known as peroxisomal thiolase-2), which is involved in peroxisomal metabolism of bile acids and branched-chain fatty acids. We show in this study that mice deficient in SCP2 and SCPx (SCP2null) develop a cardiac phenotype leading to a high sudden cardiac death rate if mice are maintained on diets enriched for phytol (a metabolic precursor of branched-chain fatty acids). Methods and Results: In 210 surface and 305 telemetric ECGs recorded in wild-type (C57BL/6; wt; n = 40) and SCP2 null mice (n = 40), no difference was observed at baseline. However, on diet, cycle lengths were prolonged in SCP2 null mice (262.9 ± 190 vs 146.3 ± 43 msec), AV conduction was prolonged (58.3 ± 17 vs 42.6 ± 4 ms), and QRS complexes were wider (19.1 ± 5 vs 14.0 ± 4 ms). In 11 gene-targeted Langendorff-perfused hearts isolated from SCP2 null mice after dietary challenge, complete AV blocks (n = 5/11) or impaired AV conduction (Wenckebach point 132 ± 27 vs 92 ± 10 msec; P 〈 0.05) could be confirmed. Monophasic action potentials were not different between the two genotypes. Left ventricular function studied by echocardiography was similar in both strains. Phytanic acid but not pristanic acid accumulated in the phospholipid fraction of myocardial membranes isolated from SCP2 null mice. Conclusion: Accumulation of phytanic acid in myocardial phospholipid membranes is associated with bradycardia and impaired AV nodal and intraventricular impulse conduction, which could provide an explanation for sudden cardiac death in this model.

    Notes: High variability of the clinical appearance of malignant melanoma (MM) and its metastases render the differential diagnosis of solid amelanotic tumours difficult. We report a 71-year-old woman with several unusual cutaneous tumours of cerebriform morphology, suggesting skin metastases from occult internal cancer. Histopathological findings and thorough investigations, however, revealed a late-stage metastatic MM. We discuss the differential diagnosis of skin metastases of various origin and underline the difficulties for early detection of MM.

    Notes: Abstract The impact of calcium fortification in the form of calcium citrate malate (CCM) on sensory attributes of an orange flavored beverage was investigated. It was hypothesized that the sensory properties would be negatively influenced by increasing the calcium concentration. Samples of an orange flavored beverage were prepared containing five levels of CCM that had 20% to 60% of the daily Adequate Intake (AI) for calcium. The influence of calcium fortification with CCM on sensory attributes of the beverage was determined using descriptive sensory analysis. In addition, a consumer acceptance test evaluated the acceptability of the sample beverages. The astringency of the beverage increased with higher levels of calcium. However, no further influence on sensory properties of the beverage was observed. There were minimal differences in consumers’liking for all five samples. The acceptability by consumers was not influenced by CCM concentration. Calcium fortification up to 60% of the AI did not reduce consumers’acceptability, however, it influenced the astringency of the orange flavored beverage.

    Notes: Abstract Observational studies provide overwhelming evidence that a low high-density lipoprotein (HDL)-cholesterol level increases the risk of coronary events, both in healthy subjects and in patients with coronary heart disease. Based on in vitro experiments, several mechanistic explanations for the atheroprotective function of HDL have been suggested. However, few of these were verified in vivo in humans or in experiments with transgenic animals. The HDL functions currently most widely held to account for the antiatherogenic effect include participation in reverse cholesterol transport, protection against endothelial dysfunction, and inhibition of oxidative stress. This review summarizes current views on the molecular mechanism underlying these atheroprotective effects of HDL.

    Notes: Abstract Environmental stresses can decrease photosynthesis by a direct effect on photosynthetic capacity of the mesophyll or by a CO2 limitation resulting from stomatal closure. In the present study, a ‘path-dependent method’ (Jones, 1985) for the partitioning of a stress-related decline in assimilation rate between non-stomatal and stomatal factors was evaluated, using light quality as a ‘stress’. Kinetic data on assimilation rate and conductance of Phragmipedium longifolium following a change in light quality from 95 μmol m−2s−1 white light to 95 μmol m−2s−1 red light failed to generate a smooth response curve for conductance. Partitioning of limitations on assimilation by a path-dependent method that utilizes the actual trajectories of conductance and assimilation was therefore not feasible. A simplified path-dependent method (Jones, 1985) which assumes that either mesophyll cells or guard cells respond first to a stress was applied to steady-state measurements of assimilation and conductance under red and white illumination. Either 5% or 23% of the observed reduction in assimilation rate under white light was attributable to stomatal factors, depending on whether the ‘stomatal first’ or the ‘mesophyll first’ path was assumed. In the absence of additional information indicating the appropriate choice of path, arbitrary choice may therefore lead to widely divergent estimates, and potentially erroneous conclusions. An alternative approach to the evaluation of the importance to carbon assimilation of stomatal and non-stomatal factors is suggested.

    Notes: In most plant species, a decrease in atmospheric humidity at the leaf surface triggers a decrease in stomatal conductance. While guard cells appear to respond to humidity-induced changes in transpiration rate, as opposed to relative humidity or vapour pressure difference, the underlying cellular mechanisms for this response remain unknown. In the present set of experiments, abscisic acid (ABA)-deficient (aba1) and ABA-insensitive (abi1-1 and abi2-1) mutants of Arabidopsis thaliana were used to test the hypothesis that the humidity signal is transduced by changes in the flux or concentration of ABA delivered to the stomatal complex in the transpiration stream. In gas exchange experiments, stomatal conductance was as sensitive to changes in vapour pressure difference in aba1, abi1-1 and abi2-1 mutant plants as in wild-type plants. These experiments appear to rule out an obligate role for either the concentration or flux of ABA or ABA conjugates as mediators of the guard cell response to atmospheric water potential. The results stand in contrast to the well-established role of ABA in mediating guard cell responses to decreases in soil water potential.
