    Notes: Abstract. When DNA topoisomerase II (topo II) activity is inhibited with a non-DNA-damaging topo II inhibitor (ICRF-193), mammalian cells become checkpoint arrested in G2-phase. In this study, we analyzed chromosome structure in cells that bypassed this checkpoint. We observed a novel type of chromosome aberration, which we call Ω-figures. These are entangled chromosome regions that indicate the persistence of catenations between nonhomologous sequences. The number of Ω- figures per cell increased sharply as cells evaded the transient block imposed by the topo II-dependent checkpoint, and the presence of caffeine (a checkpoint-evading agent) potentiated this increase. Thus, the removal of nonreplicative catenations, a process that promotes chromosome individualization in G2, may be monitored by the topo II-dependent checkpoint in mammals.

    Notes: The thermal resistance in both superfluid phases of 3He has been measured at 20.0 and 29.6 bar in zero magnetic field. Heat conduction in 3He-B is shown to be primarily hydrodynamic, and a regime of reproducible heat flow behavior in the A phase is reported. The viscosity of each phase as a function of temperature is calculated using an equation of the two-fluid model, and critical velocity effects are discussed.

    Notes: The high-temperature electrical conductivity and thermal decomposition characteristics of Sylgard® 184 with and without hollow microspheres of glass, silica, and ceramic have been determined to 600 to 700°C in air and nitrogen environments. The materials are silicone-based dielectrics and are used as electronic encapsulants. Results show that a peak in the conductivity temperature dependence at ∼300°C results principally from volatilization of [Si(CH3)2O]n with some evolution of water, that oxygen accelerates decomposition, and that the microspheres may help form a network of interconnected conductive pathways in the residual material. There is a good correlation between thermal stability and temperature-dependent electrical properties.

    Notes: Electrical conductivity changes during thermal decomposition of several phenolic and silicone materials have been measured while increasing temperature from 25 to ∼ 700°C at a rate of 10°C per minute in a nitrogen atmosphere, The materials are based on phenolic and silicone resins and are reinforced with glass chopped fabric or cloth. The electrical results are correlated with mass loss and thermal decomposition product data obtained using mass spectroscopy and thermal gravimetric analysis. Peaks in the conductivity temperature dependence and deviations from ohmic behavior are found to be associated with material decomposition and/or outgassing. An excellent correlation is obtained between thermal stability and temperature-dependent electrical properties. Results suggest that electrical conductivity can be used as a thermal analytical tool in characterizing these materials.

    Notes: UV damage to CHO cell DNA, measured by formation of thymine-containing dimers, increases from mitosis to early S phase. Computer simulation of UV absorption by the DNA of an idealized CHO cell at different stages in the cell cycle resembles the cycle dependence of UV damage.Incision at UV damage sites, measured by the accumulation of breaks in preexisting DNA during 30 minutes' post-irradiation incubation with the DNA synthesis inhibitors 1-β-D arabinofuranosylcytosine and hydroxyurea, increases from mitosis to interphase. Analysis of the dose dependence of DNA break accumulation indicates that both the affinity of the endonuclease for dimer sites and the maximum enzyme activity at saturating levels of dimers are significantly lower in mitosis than in interphase.The killing of CHO cells by UV is enhanced if repair is temporarily inhibited by ara C. The DNA gyrase inhibitor novobiocin prevents UV-induced incision.
