Notes: Seven 3H-TdR-labelled early psoriatic lesions were quantitatively evaluated after serial sectioning and autoradiography. Epidermal thickness was measured planimetrically, whereas labelled cells as well as the number of nuclei present in the dermis were counted.Marked differences in the number of labelled cells as well as in their grain density were observed. These differences correlated with the density of the dermal infiltrate. It is concluded that psoriatic epidermal hyperplasia possibly develops as a response to antecedent and persisting events in the dermis.

Notes: SUMMARY.— Single psoriatic lesions were analysed planimetrically and auto-radiographically after 3H-thymidine labelling. The results indicate maximum alterations of epidermis and papillary body in the centre of a lesion. Epidermal volume values are elevated at the periphery while the papillary body is still normal. After stripping, the proliferative activity of the peripheral epidermis increases in proportion to epidermal volume, while no stimulatory effect can be exerted in the centre of a lesion. Comparison of several lesions reveals large differences for volume values and between single rete pegs for 3H-thymidine labelling. The role of an epidermal defect in evoking the changes is discussed.

Notes: SUMMARY. The removal of the horny layer of the guinea-pig ear by tape stripping causes rapid development of a parakeratotic scale during the first 12 hr. Some epidermal cells, which were labelled with 3H-Thymidine when stripping was performed, could be detected autoradiographically in the parakeratotic stratum corneum a few hours later. Since the proliferative response of the epidermis is retarded after vigorous stripping, these cells have become parakeratotic due to exposure and subsequent cell death. Topical application of Vitamin A-acid produces morphological alterations of the keratogenous zone together with increased DNA-synthesis in the lower portion of the epidermis, leading to the persistence of nuclei in the stratum corneum. Prolonged treatment with Vitamin A-acid and Hexadecane increases the epidermal thickness to values such as are found in psoriasis. The turnover time of this hyperplastic epidermis is reduced to about 3 days, yet no parakeratosis can be found. These observations lead to the conclusion that maximally stimulated epidermopoiesis does not result in parakeratosis unless there is a defect in cell differentiation.

Notes: Summary Carbonic anhydrase activity, as demonstrated histochemically, is present in human skin. Especially high enzyme activity was found in eccrine sweat glands. Probably there are relations between acide nature of sweat and enzyme activity. Full-length report see in Arch. Dermat.
