    Notes: The glass-former salol was studied using carbon and deuteron NMR. For the 13C measurements protonated samples were used. For the deuteron experiments salol was selectively deuterated at the phenyl ring. The measurement of the spin-lattice relaxation times provides clear evidence for an anisotropic reorientation of the molecules in the supercooled liquid. Close to the glass transition a bimodal longitudinal magnetization recovery is observed which indicates the presence of relatively slow intramolecular degrees of freedom. From deuteron solid echo spectra this process is identified as a uniaxial large amplitude motion of the phenyl group which exhibits a broad distribution of correlation times. © 2000 American Institute of Physics.

    Notes: Proton, deuteron, and carbon NMR methods were applied to study various isotopic species of supercooled and glassy propylene carbonate. The molecular diffusion was investigated via static field gradient diffusometry and yielded evidence for a particularly pronounced enhancement of the translational over the reorientational dynamics. The latter was monitored using measurements of spin-lattice relaxation times and of two-time stimulated echoes. The sin–sin correlation functions exhibited a pronounced dependence on the evolution times. From these results it is concluded that the geometry of molecular reorientation can be described as a superposition of moderately small jump angles (about 30°) and a quasi-diffusive component which is responsible for about one-third of the total angular displacement. Finally, four-time stimulated echoes confirmed that the nonexponential relaxation in propylene carbonate is dynamically heterogeneous in nature. © 2000 American Institute of Physics.

    Notes: A confocal laser-scanning microscope for ultrasensitive fluorescence lifetime imaging on surfaces is presented. The system employs a compact electronics for time-correlated single-photon counting (TCSPC), allowing for measuring fluorescence lifetime with 40 ps time resolution, and for continuously recording photon arrival times with 100 ns time resolution. Additionally developed driver electronics serve for synchronization of scanning and data acquisition, which is significant for achieving high spatial image resolution. The capabilities of the measurement system are demonstrated on imaging single molecules immobilized on glass substrates. Finally, it is shown how the TCSPC capabilities of the system can be used not only for lifetime imaging but also for multichannel measurements. © 2001 American Institute of Physics.

    Notes: Surface expression of the glial glutamate transporter EAAT1 is stimulated by insulin-like growth factor 1 through activation of phosphatidylinositol-3-kinase. Downstream targets include serum and glucocorticoid-sensitive kinase isoforms SGK1, SGK2 and SGK3, and protein kinase B. SGK1 regulates Nedd4-2, a ubiquitin ligase that prepares cell membrane proteins for degradation. To test whether Nedd4-2, SGK1, SGK3 and protein kinase B regulate EAAT1, cRNA encoding EAAT1 was injected into Xenopus oocytes with or without additional injection of wild-type Nedd4-2, constitutively active S422DSGK1, inactive K127NSGK1, wild-type SGK3 and/or constitutively active T308D,S473DPKB. Glutamate induces a current in Xenopus oocytes expressing EAAT1, but not in water-injected oocytes, which is decreased by co-expression of Nedd4-2, an effect reversed by additional co-expression of S422DSGK1, SGK3 and T308D,S473DPKB, but not K127NSGK1. Site-directed mutagenesis of the SGK1 phosphorylation sites in the Nedd4-2 protein (S382A,S468ANedd4-2) and in the EAAT1 protein (T482AEAAT1, T482DEAAT1) significantly blunts the effect of S422DSGK1. Moreover, the current is significantly larger in T482DEAAT1- than in T482AEAAT1-expressing oocytes, indicating that a negative charge mimicking phosphorylation at T482 increases transport. The experiments reveal a powerful novel mechanism that regulates the activity of EAAT1. This mechanism might participate in the regulation of neuronal excitability and glutamate transport in other tissues.

    Notes: The automatic ICD improves survival in patients with a history of sudden cardiac arrest. However, some patients do not meet the guidelines for ICD implantation or are unable to receive an implantable device. This study tested the hypothesis that these patients could benefit from a wearable cardioverter defibrillator. Patients with symptomatic heart failure and an ejection fraction of 〈0.30 (WEARIT Study) or patients having complications associated with high risk for sudden death after a myocardial infarction or bypass surgery not receiving an ICD for up to 4 months (BIROAD Study) were enrolled into two studies. After a total of 289 patients had been enrolled in the trial (177 in WEARIT and 112 in BIROAD), prespecified safety and effectiveness guidelines had been met. Six (75%) of eight defibrillation attempts were successful. Six inappropriate shock episodes occurred during 901 months of patient use (0.67% unnecessary shocks per month of use). Twelve deaths occurred during the study 6 sudden deaths: 5 not wearing and 1 incorrectly wearing the device). Most patients tolerated the device although 68 patients quit due to comfort issues or adverse reactions. The results of the present study suggest that a wearable defibrillator is beneficial in detecting and effectively treating ventricular tachyarrhythmias in patients at high risk for sudden death who are not clear candidates for an ICD and may be useful as a bridge to transplantation or ICD in some patients.     Notes: Sliding friction between railway wheels and rails results in elevated contact temperatures and gives rise to severe thermal stresses at the wheel and rail surfaces. The thermal stresses have to be superimposed on the mechanical contact stresses. Due to the distribution of stresses, the rail surface is generally subjected to higher stresses than the wheel surface. The elastic limit is reduced and yield begins at lower mechanical loads. During the first cycles of plastic deformation, the material hardens and residual stresses build up. The residual stresses provide the structure to shake down to pure elastic behaviour in subsequent load cycles up to a shakedown limit. The kind of hardening observed for rail steel has a considerable influence on the shakedown limit. The shakedown limit is dropped to lower mechanical loads due to the thermal stresses in the rail surface as well. This might cause structural changes in the rail material and rail damage.

    Notes: The contribution of Ca2+-activated K+ channels to hyperpolarizing after-potentials (HAP) of action potentials, to spike-frequency adaptation and thus to the shaping of discharge pattern, was examined in rat supraoptic magnocellular neurosecretory cells. In addition, the expression of BK channels and SK3 subunits of SK channels was studied using double immunofluorescence detection. The presence of BK channels and SK3 subunits was detected in many supraoptic neurones containing either vasopressin or oxytocin. Current-clamp recordings of current-induced spike trains revealed that HAPs comprise a fast and a slow HAP (fHAP and sHAP). Correlation analyses revealed that the increase of the fHAP in amplitude and spike broadening were correlated to a moderate gradual increase of the interspike interval and thus to weak spike-frequency adaptation. By contrast, marked prolongation of the interspike interval and strong spike-frequency adaptation depended on the appearance and on the amplitude of the sHAP. The sHAP and spike-frequency adaptation were blocked by cadmium, as well as by the SK channel antagonist apamin. The fHAP was attenuated by the BK channel antagonist iberiotoxin (IbTX), by the BK/IK channel antagonist charybdotoxin (ChTX) and by apamin. ChTX attenuated fHAPs throughout the entire spike train. By contrast, the IbTX-induced attenuation of the fHAP was restricted to the initial part of the spike train, while the apamin-induced attenuation slowly increased with the progression of the spike train. These results suggest that strong spike-frequency adaptation in supraoptic neurones essentially depends on the generation of the sHAP by activation of SK channels. Comparison of effects of IbTX, ChTX and apamin suggests a complementary contribution of SK-, BK- and IK-channels to fHAPs.

    Notes: Background The standard assay for the clinical detection of contact allergy against nickel is the patch test. For research purposes, the in vitro lymphocyte transformation test (LTT) is mostly applied. However, the in vivo application of allergens includes the potential of sensitization, whereas the LTT demands 6 days of cell culture and radioactive labelling procedures.Objective The purpose of this study was to validate an enzyme-linked immunospot (ELISpot) assay to specifically detect cellular response against nickel sulphate in correlation with patch test, anamnestic data, and LTT results.Methods In probands with positive (n=60) and negative patch test results (n=19), cytokine ELISpot and LTT were performed in parallel using various concentrations of nickel sulphate, various numbers of responding peripheral blood mononuclear cells (PBMC), and various durations of cell cultures.Results Concentrations of 50 and 75 μM nickel sulphate were found to be optimal to stimulate in the ELISpot 4×105 and in the LTT 2×105 PBMC, respectively. An increase in ELISpot sensitivity was reached by pre-incubation with nickel sulphate for 24 h prior to transfer to ELISpot plates. In nickel-sensitive probands, an average precursor cell frequency of 19×105, 1.7×105, and 0.7×105 could be defined for IFN-γ, IL-2, and IL-4 producing PBMC, respectively. In non-sensitive probands IFN-γ producing cells were detectable, but with significantly lower frequency (2×105; P=0.004). The prior performance of patch tests had no significant effect on cytokine production or lymphocyte proliferation. Overall, the parameters patch test, anamnestic nickel allergy, ELISpot, and LTT results were positively correlated (P〈0.05).Conclusions Based on these experiments, the quantitative in vitro detection of cell-mediated reactions towards nickel as well as other heavy metal ions should be possible utilizing the above-described ELISpot assay.
