    Notes: The relationship is investigated between superoxide levels in single cultured rat cerebellar granule neurons exposed continuously to glutamate in low KCl medium and the deregulation of cytoplasmic Ca2+. Cells that maintain a regulated cytoplasmic-free Ca2+ and mitochondrial polarization in the presence of glutamate show no increase in superoxide levels until the onset of cytoplasmic Ca2+ deregulation. Oxidative stress of mitochondrial origin is readily detectable, as the inhibitors rotenone and antimycin A markedly increase superoxide levels with no effect on cytoplasmic-free Ca2+. The potent cell-permeant superoxide dismutase/catalase mimetic manganese tetrakis (N-ethylpyridinium-2yl) porphyrin, MnTE-PyP, abolishes the deregulation-related increase in superoxide but has no effect on deregulation itself. A combination of catalase with the free radical scavenger 4-hydroxy-TEMPO also fails to reduce deregulation. Following the loss of Ca2+ homeostasis nuclei undergo condensation; this morphological change is not inhibited by MnTE-PyP and cannot account for the increased ethidium fluorescence. Phospholipase A2 inhibitors decrease the deregulation-related increase in superoxide without protecting against deregulation. In conclusion, our study indicates that deregulation is not caused by NMDA receptor-mediated oxidative stress as NMDA receptor activation does not increase superoxide levels until the onset of deregulation. Furthermore, the majority of superoxide is produced in the cytoplasm rather than in mitochondria.

    Notes: Effects of nicotine and caffeine, separately and in combination, were assessed in 12 male habitual smokers in a repeated-measures design. Caffeine (0-mg vs. two 150-mg doses administered in a decaffeinated/sugar-free cola drink post-baseline and 90 min later) was crossed with nicotine (ad libitum own dosing vs. 1.0-mg machine-delivered dose vs. 0.05-mg machine-delivered dose). Participants smoked a total of five cigarettes at 30-min intervals over a 2-hr period. Caffeine and nicotine had large effect sizes on electroencephalogram (EEG) power; however, these effects were modulated by the eyes open versus closed condition, the other drug, and electrode site. EEG effects of open versus closed eyes tended to be of the same size and direction as those of nicotine and caffeine. However, whereas nicotine increased EEG power in some higher frequency bands in some conditions, caffeine decreased EEG power across almost all conditions. Serum cortisol concentration, vigor, and pleasantness were increased by nicotine, but not by caffeine. Level of depressive mood depended on an interaction of caffeine and nicotine. Vigilance performance was enhanced significantly by caffeine and was increased almost significantly by nicotine. The findings were interpreted in terms of common and differential mechanisms of the two drugs.

    Notes: Atomic force microscopy is used to characterize the evolution of film morphology produced by heavy-ion bombardment. Pt films, 3 and 5 nm thick, are deposited on SiO2 substrates and subsequently bombarded by 800 keV Kr+. Ion doses of 〉2×1014 initiate pattern formation and the dewetting of Pt films from the substrate. The film morphology becomes increasingly disconnected with increasing dose; at the highest doses, (∼2×1016 cm−2), isolated nanoparticles are formed with a uniform spacing. The results are explained by the nucleation of bare substrate patches and subsequent coarsening of the morphology by the molten zones created by individual Kr+ impacts. © 2000 American Institute of Physics.

    Notes: This article investigates onion epidermal tissue (Allium cepa) using a combination of mechanical testing, microscopy and modeling and relates tissue mechanical properties to the known structure of the cell walls. Onion epidermal tissue has a simple, regular structure of elongated cells, which have been used to enable the contributions to mechanical properties of cell walls and of higher order structures to be separated and analyzed. Two models of wall behavior were used to explore how Poisson's ratio of cell walls parallel to the plane of the epidermal surface may vary with applied strain. In the first model, cellulose microfibrils can be reorientated in an unrestricted way with the result that the cell wall volume decreases. In the second model the volume of the cell wall remains constant, which controls the reorientation of microfibrils, hence the Poisson's ratio. Measurements made from uniaxially stretched cells show that the data most closely fits model I, therefore, it is concluded that the bulk of the matrix has little influence on the observed mechanical properties (at a test rate of 1 mm/min), allowing cellulose microfibrils to reorient through the matrix in an unrestricted way during uniaxial tests. In its mechanical attributes the primary cell wall resembles more a knitted cloth than a semisolid composite material. When biaxial stretching is applied to tissue, so that there is no re-orientation of microfibrils, the cell wall material is still able to reach surprisingly large elastic strains of up to 12.5% and no plastic deformation was recorded. Current theory suggests that cellulose microfibrils can stretch elastically by a maximum of 7%, therefore further work is required to identify mechanisms that could account for the extra elastic strain.

    Notes: Abstract –Objective: Clinical patterns of early childhood caries (ECC) encompassing specific teeth or surfaces have been previously proposed on an a priori basis and have been used as case definitions. The underlying assumption is that the patterns result from different host response and environmental conditions. Identifying caries patterns has utility in refining case definitions of ECC. Well-defined caries patterns should enhance the ability of an analysis to identify meaningful associations between suspected risk factors and ECC. The purpose of this project was to identify patterns of caries in the dentition of preschool children using multidimensional scaling without a priori pattern delineation.Methods: Between February 1994 and September 1995, five examiners visually examined 5171 Arizona preschool children aged 5–59 months old. Multidimensional scaling (MDS) was used as a classification/taxonomy technique to identify any underlying structure of the caries data. MDS provided a classification scheme for individual tooth surfaces based on the dissimilarity measures of squared Euclidean distance and of variance using an alscal analysis. Both two- and three-dimensional solutions were pursued; s-stress, stress, R-square and residual patterns were assessed in determining the best dimensional model, with the resulting quadrant positions of the tooth surfaces suggesting potential caries patterns.Results:  All models demonstrated excellent fit. Two- and three-dimensional solutions suggested four caries patterns: (i) any maxillary incisor surfaces, (ii) first molar occlusal surfaces, (iii) second molar pit and fissure surfaces, and (iv) any smooth surfaces, excluding the maxillary incisor surfaces.Conclusions: This is the first delineation of primary dentition caries patterns produced by a classification analysis without a priori pattern definitions. The identified caries patterns may arise from specific risk factors and/or be a function of the timing of various risk factor exposures. Use of these patterns as case definitions should enhance the ability to identify associations between suspected risk factors and ECC.

    Notes: The rocks of western Ny-Friesland, northern Svalbard, are part of a tectonostratigraphy including four thrust sheets, each composed mainly of orthogneisses overlain by younger metasedimentary rocks. Previous geochronological studies have shown that the orthogneisses are dominated by ca. 1750 Mya granitoids. This study of a quartz-monzonite in one of the thrust sheets, the Nordbreen Nappe, yields a single-zircon U-Pb ion-microprobe age of 2709 Ø 28 My. This is the oldest rock unit so far reported in the Svalbard Caledonides. However, age-determinations on detrital zircons in the metasediments of western Ny-Friesland have shown that Late Archean rocks were prominent sources. The new ages presented here provide the first evidence of a local source for these sedimentary rocks.
