    Notes: Microglia respond in a stereotypical pattern to a diverse array of pathological states. These changes are coupled to morphological and immunophenotypical alterations and the release of a variety of reactive species, trophic factors and cytokines that modify both microglia and their cellular environment. We examined whether a microglial-produced cytokine, tumour necrosis factor-α (TNF-α), was involved in the maintenance of microglial activation after spinal cord injury by selective inhibition using TNF-α antisense deoxyoligonucleotides (ASOs). Microglia and macrophages harvested from 3 d post-contused rat spinal cord were large and rounded (86.3 ± 9.6%). They were GSA-IB4-positive (GSA-IB4+) (Griffonia simplicifolia lectin, microglia specific; 94.8 ± 5.1%), strongly OX-42 positive (raised against a type 3 complement/integrin receptor, CD11b; 78.9 ± 9.1%), ED-1 positive (a lysosomal marker shown to correlate well with immune cell activation; 97.2 ± 2.6%) and IIA positive (antibody recognizes major histocompatibility complex II; 57.2 ± 5.6%), indicative of fully activated cells, for up to 48 h after plating. These cells also secreted significant amounts of TNF-α (up to 436 pg/µg total protein, 16 h). Fluoroscein isothiocyanate-labelled TNF-α ASOs (5, 50 and 200 nm) added to the culture medium were taken up very efficiently into the cells (〉 90% cells) and significantly reduced TNF-α production by up to 92% (26.5 pg/µg total protein, 16 h, 200 nm TNF-α ASOs). Furthermore, few of the treated cells at this time were round (5.4 ± 2.7%), having become predominantly spindle shaped (74.9 ± 6.3%) or stellate (21.4 ± 2.7%); immunophenotypically, although all of them remained GSA-IB4 positive (91.6 ± 6.2%), many were weakly OX-42 positive and few expressed either ED-1 (12.9 ± 2.5%) or IIA (19.8 ± 7.4%). Thus, the secretion of TNF-α early in spinal cord injury may be involved in autoactivating microglia/macrophages. However, at the peak of microglial activation after injury, the activation state of microglia/macrophages is not stable and this process may still be reversible by blocking TNF-α.

    Notes: Dermal absorption experiments form an important component in the assessment of risk from exposure to pesticides and other substances. Much dermal absorption data is gathered in rat experiments carried out using a certain standard protocol. Uncertainties in these data arise from many sources and can be quite large. For example, measurements of the systemic absorption of hexaconazole differed by more than an order of magnitude within a single experiment. Two diniconazole studies produced quite different results, due to minor differences in protocol and in chemical formulation. Limits of detection can also prevent accurate measurement when the amounts absorbed are small. These examples illustrate the need for measuring and reporting uncertainties in estimates that are based on these data. The most direct way to estimate uncertainty is to compute the sample standard deviations of replicate measurements. By pooling these estimates across dose and duration groups for which they are similar, the number of degrees of freedom is increased, and more precise confidence intervals can be obtained. In particular, the ratio of upper to lower 95% confidence limits was reduced by as much as ten-fold for hexaconazole, seven-fold for uniconazole, and nearly four-fold for propiconazole.

    Notes: An anaerobic mixed culture enriched over 16 transfers (1/10) from Saale river sediment reductively dehalogenated 1,2,4- and 1,2,3-trichlorodibenzo-p-dioxin (TrCDD) to di- and monochlorinated congeners in the presence of pyruvate (or lactate) and fumarate as cosubstrates. Besides TrCDD, tetrachloroethene and 1,2,3-trichlorobenzene were dechlorinated. Dioxin dehalogenation was sensitive to pasteurization, but was not remarkably influenced by inhibitors of methanogens, sulfate-reducing bacteria or Gram-positive bacteria. The rate of 1,3-dichlorodibenzo-p-dioxin formation increased with rising initial concentrations of 1,2,4-TrCDD (1–250 μM) from 0.05 to 5.4 μmol l−1 day−1. Two isolates, belonging to Sulfurospirillum and Trichococcus, did not show reductive dehalogenation. 16S rDNA-targeted methods further revealed the presence of Acetobacterium, Desulfitobacterium, Desulfuromonas and Dehalococcoides. Nested polymerase chain reaction (PCR) indicated the presence of Dehalococcoides in highest most probable number (MPN) dilutions that were positive for dioxin dechlorination.

    Notes: The instrumental transparency functions of five commercially available texture goniometers were measured experimentally with six monocrystalline samples cut in different orientations from a large highly perfect silicon crystal with a rocking curve of less than 0.01°. Transparency functions were measured in steps of 0.02 to 0.2° in the pole-figure angles α, β. The window size Δα depends on the Bragg angle θ in the form 1/sinθ; the window size Δω is constant for each goniometer. The dominant instrumental parameter determining the long axis Δα of the pole-figure window is the axial width of the detector entrance slit. This parameter is smallest for area detectors (smaller by more than an order of magnitude compared with conventional scintillation detectors as well as one-dimensional position-sensitive detectors). The main features of the pole-figure window and their dependence on the instrumental parameters can be deduced fairly well from a simple geometrical model. The particular shapes of the transparency functions of the studied goniometers are markedly different. Particularly, they are not very well represented by Gauss functions. The two-dimensional transparency function can be fairly well characterized by its α and β profiles. The normalized profiles are virtually independent of the goniometer angles 2θ and α. The increasing size of the pole-figure window with decreasing θ puts a lower limit on the Bragg angle below which pole-figure measurement ceases to be meaningful.

    Notes: Slices of ∼12 mm thickness cut out of an octahedritic iron–nickel meteorite of the Gibeon shower were investigated by conventional X-ray transmission radiography. When the direction of the incident X-ray beam is parallel to one or two of the four habit planes of the kamacite lamellae, black–white contrast lines are observed in the radiographs. These lines are attributed to Bragg reflection, at lattice planes parallel to the lamellae planes, of a narrow band of wavelengths filtered out of the incident white spectrum by absorption in the sample.
