    Notes: We have examined basal phosphoinositide turnover in synaptosomes obtained from the dentate gyrus of anaesthetized rats in which long-term potentiation was induced unilaterally in perforant path-granule cell synapses. Relative to the unpotentiated side. [3H]myo-inositol labelling of inositol phosphates was significantly enhanced 45 min and 3 h after induction of long-term potentiation, but reduced after 2.5 min. Similarly, [14C]arachidonic acid labelling of 1,2-diacylglycerol was increased 45 min and 3 h after induction of long-term potentiation, but reduced after 2.5 min. In a second series of experiments, induction of long-term potentiation was blocked by stimulation of the commissural projection to granule cells. In synaptosomes prepared from this tissue, there was no difference in phosphoinositide turnover between tetanized and control sides at any of the three post-tetanic intervals. We conclude that in the dentate gyrus, long-term potentiation is associated with an increase in phosphoinositide turnover which is established between 2.5 min and 45 min post-tetanus and which persists for at least 3 h.

    Notes: The release of amino acids and the hydrolysis of inositol phospholipids were examined in parallel in three hippocampal areas following classical conditioning. Paired or unpaired tone(CS) - shock(US) presentations were given to animals engaged in a previously acquired food-motivated lever-pressing task. Conditioned suppression of lever-pressing was the behavioural measure of conditioning. Twenty-four hours after the last conditioning session, the dentate gyrus and areas CA3 and CA1 of the hippocampus were removed bilaterally from conditioned and pseudoconditioned animals, and slices cut and stored in liquid nitrogen for subsequent analysis. Crude synaptosomal pellets were prepared to investigate: (i) potassium-stimulated release of preloaded [3H]glutamate and [14C]aspartate in the presence and absence of extracellular Ca2+; (ii) [3H]inositol labelling of phosphoinositides and inositol phosphates; and (iii) [14C]arachidonic acid labelling of 1,2-diacylglycerol (1,2-DG). Potassium-stimulated, Ca2+-dependent release of [3H]glutamate in synaptosomes prepared from the dentate gyrus and area CA3 was significantly greater in conditioned animals than in pseudoconditioned animals. In area CA1, K+-stimulated, Ca2+-dependent release of [14C]aspartate was significantly increased in conditioned animals. These results confirm in synaptosomes, and extend to a period of 24 h our previous report of an increased release of transmitter in the dentate gyrus and hippocampus associated with classical conditioning. In parallel with the increased release of amino acids, learning was associated with a significant increase in labelling of phosphoinositides and inositol phosphates by [3H]inositol and a significant increase in labelling of 1,2-DG by [14C]arachidonic acid in the three hippocampal areas examined. It is suggested that a long-lasting presynaptic activation of inositol lipid metabolism may contribute to the learning-dependent increase in the capacity of hippocampal terminals to release transmitter and hence to the maintenance of a neurochemical trace which may, at least in part, underlie lasting changes in synaptic function built up during associative learning.

    Notes: We examined the induction and maintenance of long-term potentiation (LTP) in vivo in the dentate gyrus of 4-month-old and 22-month-old urethane-anaesthetized rats. High-frequency stimulation of the perforant path induced an immediate increase in the slope of the population excitatory postsynaptic potential (EPSP), which was sustained in the 4-month-old animals for the duration of the experiment (45 min post-tetanus). In the 22-month-old group, the mean slope of the population EPSP decreased almost to baseline by the end of the experiment. Examination of the individual records indicated that LTP was sustained for the duration of the experiment in half of the 22-month-old animals, while in the others only post-tetanic potentiation was observed. Membrane arachidonic acid concentration was reduced in aged compared with young animals and was lowest in the subgroup of aged animals which failed to sustain LTP. Potassium-stimulated, calcium-dependent release of glutamate was also decreased in aged compared with young animals, but LTP was associated with an increase in glutamate release in the 4-month-old group and 22-month-old subgroup in which LTP was successfully sustained; no change was observed in the 22-month-old group in which LTP was not sustained. The results indicate a correlation between membrane arachidonic acid concentration, glutamate release and ability to sustain LTP in aged animals.

    Notes: Increasing restrictions on sewage sludge disposal routes and the tightening of treatment requirements have led to an escalation of sludge-disposal costs which are out of proportion to other sewage-treatment costs: consequently reducing sludge volume is becoming increasingly cost-effective. Consolidation of anaerobically digested sludge has traditionally been difficult and takes many weeks in secondary digesters or deep storage tanks. Pre-aeration of the sludge using the Aercon process can enable consolidation to take place in days instead of weeks.This paper reports the results of a pilot-scale plant designed to investigate further the mechanism by which aeration speeds up consolidation. It was concluded that the rate of consolidation depended on aeration intensity and cooling of the sludge. Inhibition of anaerobiosis was more important than gas stripping in enhancing consolidation.

    Notes: Abstract Background: A significant proportion of breast cancer patients receiving tamoxifen therapy relapse during treatment following acquisition of tamoxifen-resistant or oestrogen-independent phenotypes. The mechanism behind this rapid progression to oestrogen autonomy is at present unclear and further treatment modalities are limited. Suramin represents a novel potential second line therapy. The mechanism of the antineoplastic activity of suramin is not completely understood, although the drug binds to many growth factors including epidermal growth factor and insulin-like growth factors and can also dissociate growth factors from their receptors. In this study we have related suramin sensitivity to the expression of receptors for epidermal growth factor and insulin-like growth factor-I in a number of breast cancer cell lines including lines resistant to tamoxifen. Materials and methods: The anti-proliferative effects of suramin were investigated in two oestrogen dependent breast cancer lines (ZR-75-1 and MCF-7), oestrogen independent (ZR-PR-LT) and tamoxifen resistant (ZR-75-9a1) variants of ZR-75-1 and a tamoxifen resistant (LY2) variant of MCF-7. Full dose response curves were constructed and IC50values determined for each cell line. Sensitivity to suramin was correlated with the level of expressio n of receptors for epidermal growth factor (EGFR) and insulin-like growth factor-I (IGFR). On observing stimulation of cell proliferation by suramin in the tamoxifen resistant cell lines in the presence of tamoxifen we also investigated the possible role of suramin sequestration of transforming growth factor-β in mediating this effect. Results: All cell lines exhibited a dose- and time-dependent response to suramin treatment. Tamoxifen resistant ZR-75-9a1 cells (day 6 IC5085 µg ml−1) were more resistant to suramin than oestrogen independent ZR-PR-LT cells (day 6 IC5045 µg ml−1), and the parent ZR-75-1 line (day 6 IC5056 µg ml−1). Increased sensitivity to suramin was associated with increased expression of IGFR and decreased expression of EGFR. Tamoxifen resistant LY2 cells were significantly more sensitive to suramin (day 6 IC5070 µg ml−1) than MCF-7 cells (day 6 IC50350 µg ml−1). Both IGFR and EGFR expression by LY2 cells was lower than in the parent line. The antioestrogen-resistant ZR-75-9a1 and LY2 lines grown in the presence of 8 µM tamoxifen were growth stimulated by concentrations of the drug below 100 µg/ml. As growth stimulation observed in the presence of tamoxifen may have been due to suramin sequestration of tamoxifen induced TGF-β1 secretion we also investigated the response of the cells to this peptide in the presence and absence of suramin. All cell lines were growth inhibited by TGF-β1 except ZR-75-9a1 which was unresponsive. Responses to TGF-β1 were modified in the presence of 100 µg suramin ml−1 although TGF-β1 was unable to mimic the ability of tamoxifen to stimulate proliferation in the presence of suramin. Conclusions: These results suggest that for ZR-75-1 cells and variants, increased sensitivity to suramin is associated with an increase in expression of IGFR and a decrease in EGFR numbers. However, tamoxifen resistant LY2 cells, in which both IGFR and EGFR expression is reduced were considerably more sensitive than parental MCF-7 cells suggesting that there is no clear relationship between EGFR and IGFR expression and suramin sensitivity. The unexpected stimulation of cell proliferation of the tamoxifen resistant variants by suramin in the presence of tamoxifen could not be explained by suramin sequestration of transforming growth factor-β and the mechanism of this interaction remains unclear.

    Notes: Abstract Objective. The objective of this study was to evaluate the postoperative technetium-99m-labeled methylene diphosphonate (99mTc-MDP) scintigraphic appearance of limb-sparing procedures in patients treated for bone tumors.Materials and methods. We retrospectively reviewed the medical records and assessed planar bone scans, subjectively and semiquantitatively, of all patients treated with limb-sparing procedures at our institution who survived at least 1 year following resection of the primary lesion.Results. The operative sites of 20 of the evaluable 45 patients (44 %) demonstrated normal tracer avidity during follow-up (median 12 months). Nine patients (20 %) demonstrated normal avidity on their first follow-up bone scans (median 6 months). Coincident99mTc-MDP bone scans were obtained on 11 patients who developed 12 postoperative complications or injury during the study and accurately identified the lesion in eight (67 %).Conclusion. Although many patients have abnormal99mTc-MDP avidity in the operative site after limb-sparing surgery, almost half eventually have normalization of uptake. However, planar bone scans have limited use for assessing the primary tumor site postoperatively as persistent abnormal avidity may preclude detection of changes associated with development of postoperative complications.
