    Notizen: Cells with procollagen α1(I) transcripts were demonstrated at the interface of natural coral mineral implanted in the femur of rats after 7, 14, 21 and 28 days by in situ hybridization with digoxigenin-labelled RNA probes. Procollagen α1(I) transcripts were detectable in osteoblasts between 7 and 28 days after implantation. As early as 7 days after implantation, procollagen α1(I) RNA expression was also demonstrated in fibroblasts; the transcript steady state levels decreased until the 28th day, when they subsided completely. The development of bone was significant during the 4 weeks of implantation. Inhibition of bone development by the coralline material could not be recorded.

    Beschreibung / Inhaltsverzeichnis: The aim of this study was to assess the prevalence of oral leukoplakia (OL) in HIV-positive patients and to evaluate the clinical and histopathological features and the presence of Candida hyphae and viral infections (HSV 1, 2, HPV, EBV and CMV) by immunocytochemical methods using mono- or polyclonal antibodies. A total of 269 HIV-seropositive patients, (222 men, median age 36.7 years and 47 women, median age 32 years) were registered. OL occurred in 20/269 (7.4%) patients (men, smokers, median age 38 years); the non-homogeneous form was observed in 11%. A lesional biopsy specimen was obtained from 8/20 patients. Specimens did not show signs of epithelial dysplasia. Candida hyphae were evident in 2/8 cases. Two patients tested positive for HPV and CMV; antibodies against EBV, HSV1 and 2 were negative. In conclusion, the prevalence of OL is comparatively high in this group of HIV-seropositive patients (7.4% vs. 0.9–2.2%). Moreover, the mean age of the study group at the time of diagnosis for oL was lower (38 years vs. 60 years).
    Notizen: Ziel der Studie war, die Prävalenz der oralen Leukoplakie (oL) bei Patienten mit HIV-Infektion zu ermitteln, die klinischen und histopathologischen Charakteristika und Kofaktoren wie Herpes-simplex-Virus 1 und 2, Humane Papillomviren, Epstein-Barr-Virus und Zytomegalievirus mittels immunhistochemischer Nachweismethoden (APAAP) sowie Candidahyphen (PAS) zu evaluieren. 269 HIV-seropositive Patienten, (222 Männer, Altersmedian: 36,7 Jahre und 47 Frauen, Altersmedian 32 Jahre) wurden anhand standardisierter Protokolle erfaßt. 20/269 (7,4%) Patienten (Männer, Raucher, Medianwert des Alters 38 Jahre) wiesen eine orale Leukoplakie auf (11% inhomogene Form). Bei 8/20 wurde eine Biopsie entnommen: Dysplasien wurden nicht nachgewiesen, Candidahyphen waren in 2/8 Fällen vorhanden, HPV und CMV wurde jeweils in einem, EBV und HSV1/ 2 in keinem Fall nachgewiesen. Die Prävalenz der oL mit 7,4% scheint im Vergleich zur Normalbevölkerung (0,9–2,2%) höher zu liegen, auch ist diese bei HIV-Infizierten schon 25 Jahre früher zu diagnostizieren.

    Beschreibung / Inhaltsverzeichnis: Summary The lymph nodes of the newborn mouse represent „Anlagen” that become fully developed during the 3rd week of life. At the time of birth the degree of development corresponds to about the 5th to 6th fetal month in the human. The postnatal development of the mouse lymph node is systematically traced, especially in regards to certain structures of this organ. The relationships between these structures during the differentiation of the lymph node are also presented. The manner by which the sinuses and the blood vessels develop is significant for the final structure of the lymph node. The primarily pure reticulum cell „Anlage” is capable of assuming various forms depending upon its origin and determination. Its growth and differentiation are discussed from the cytopoetical aspect. Plasma cells are not formed during the developmental stage. In the mature lymph node the postcapillary veins are characterized by peculiar endothelial structures which suggest an intravasal cytopoesis. In human lymphatic tissue reactive processes demonstrate the same phenomenon, as shown in 2 cases. The acting mechanism, however, remains unknown. These observations are discussed with regard to their significance for „general pathology”.
    Notizen: Zusammenfassung Die Lymphknoten bei der Maus stellen bei der Geburt primitive Anlagen dar und entwickeln sich erst in den ersten 3 Lebenswochen zur vollen Reife. Ihre Ausbildung zur Zeit der Geburt entspricht etwa dem 5.–6. Embryonalmonat beim Menschen. Die postnatale Entwicklung des Mäuselymphknotens wird aufgezeigt. Das Verhalten der Sinus und Blutgefäße ist für die endgültige Struktur des Lymphknotens maßgebend. Plasmazellen werden während der Entwicklungsphase nicht gebildet. Im ausgereiften Lymphknoten der Maus zeichnen sich die postcapillären Venen durch eigentümliche endotheliale Strukturen aus, die die Annahme einer intravasalen Cytopoese nahelegen. Ein gleichartiges Phänomen findet sich auch im lymphatischen Gewebe des Menschen, wie an zwei Beispielen gezeigt wird.

    Beschreibung / Inhaltsverzeichnis: Zusammenfassung Kritisch gefüllte Thermosyphons für den Wärmetransport unterscheiden sich von kritisch gefüllten Testzellen für Stoffwertuntersuchungen durch größere Ausdehnung und aufgeprägtes Temperaturgefälle. Die thermodynamischen Zustände im Fluid des Thermosyphons sind damit wesentlich heterogener. In einem gläsernen Thermosyphon ließen sich Strömungen und Phasenverteilungen beobachten. Das bekannte Zusammenbrechen des Wärmetransportmechanismus zeigte sich als Folge des Überflutens der Kühlzone durch Flüssigkeit. Der Einfluß von Heizleistung, Druck und Füllmenge auf die Verteilung der Phasen, das Auftreten von Pulsationen und die Auswirkung eines Heizimpulses nahe dem kritischen Zustand wurden beobachtet und mit Hilfe des Dichte/Druck-Diagramms erklärt.
    Notizen: Abstract Critically filled thermosyphons for heat transport differ in size and temperature distribution from critically filled testcells for property research. Consequently, the thermodynamic states within a thermosyphon are distributed more heterogeneously. In a glass thermosyphon, flows and phase distributions were observed. A break-down of the heat transport mechanism was found as a consequence of flooding of the cooling region by liquid. Observed and explained - by use of a density/pressure diagram - were the effects of heating, of pressure and of fluid quantity on the distribution of the phases, the appearance of pulsations and the propagation of a heat pulse near the critical state.

    Notizen: Abstract Natural coral consisting of calciumcarbonate (CaCO3) in the crystal form of aragonite was investigated after implantation into the cortex and marrow cavity of rat femur at 7, 14, 21, and 28 days by means of scanning and transmission electron microscopy (SEM and TEM) as well asin situ hybridization (ISH) in order to better understand its mechanism of bone bonding, which is somewhat different to that of other slowly degradable bioactive materials, e.g. hydroxyapatite or glass ceramics. Bone bonding was shown to be closely coupled with implant degradation. Degradation by dissolution started as early as the insertion of the implant leading to a pronounced surface rugosity. Additionally, at later stages, degradation by multi-nucleated osteoclast like cells was enhanced. Bone bonding was mediated by flat cells settling in groups on the implant surface. These cells produced mineralizing globules and collagen that anchored directly to the implant surface, i.e. on the tips of the surface rugosity and inside the pores. Through ISH the cells were shown to produce procollagen α1(l) transcripts. The calcification at the interface was enhanced by matrix vesicles similar to woven bone formation. Therefore, the calcification on the implant surface resembled woven bone formation and no distinct afibrillar intervening layer resembling a cement-line, as in other bioactive implants or as in bone-bone interfaces, e.g. in lamellar bone was observed in bone bonding areas. The mineralization in deeper micropores which did not include direct cell activity depended on other processes, e.g. dissolution and reprecipitation.

    Notizen: Abstract The prerequisite for the development of a biohybrid artificial kidney, is a substrate for confluent growth of renal cells forming an epithelial monolayer without any leaks. Conventional cell culture supports cannot be adapted for this purpose, because they lack adequate mechanical properties and thermal stability. From two suitable materials, polysulfone and polyacrylonitrile, two permeable polymeric membranes have been produced that were, according to ISO 10993-5, not cytotoxic. Cloned Madin Darby Canine Kidney (MDCK) cells (an established renal cell line) were cultured on the surface of the plastic materials, and on conventional cell culture supports. With all materials, assays of mitochondrial and lactate dyhydrogenases exhibited similar proliferation and the viability of the MDCK cells. Transmission electron microscopy showed the expression of a normal morphology of kidney tubular cells. Perfect barrier function, consequent on the formation of intercellular junctions in a confluent tight epithelium, was visualized in electron micrographs, and quantified by measurement of the transepithelial resistance. The uniformity of the cells grown was demonstrated in samples by electron microscopy and in the whole epithelium by intravital impedance analysis. It was concluded that polymeric membranes produced from polysulfone or polyacrylonitrile are appropriate substrates in the design of biohybrid kidney devices. © 1998 Kluwer Academic Publishers

    Notizen: Abstract A rat bone marrow stromal cell (RBM) culture was used to evaluate novel bioactive calcium phosphate ceramics. Three rapidly resorbable, glassy crystalline materials with the main crystalline phase Ca2KNa(PO4)2 were investigated (sample code GB 1a, GB 14, GB 9). These materials were designed to exhibit a higher degree of biodegradability than tricalcium phosphate. Additionally, a bioactive glass ceramic of low biodegradability was examined (sample code AP 40). RBM cells were cultured on the disc-shaped test substrata for 14 d. The culture medium was changed and calcium and phosphate concentrations of the medium were determined daily. Specimens were evaluated using light microscopy and morphometry of the cell-covered substrate surface, scanning electron microscopy and energy dispersive X-ray analysis. Except for GB 1a, the rat bone marrow cells attached and grew on all substrate surfaces. Of the different calcium phosphate ceramics tested, AP 40 facilitated osteoblast growth and the elaboration of the extracellular matrix to the highest degree followed by GB 9 and GB 14. The inhibition of cell growth encountered with GB 1a seemed to be related to its high phosphate ion release. © 1998 Chapman & Hall

    Notizen: Abstract There is evidence that not only the immune status, but also the genetic predisposition of certain hosts influence the clinical outcome of Toxoplasma gondii infection. By far the majority of our knowledge on genetic and immunological mechanisms involved in control of T. gondii infection has been obtained by studying mouse models, which in terms of clinical outcome of infection differ considerably from humans. Rats which show a rather similar course of infection in comparison to humans have not so far been investigated for effects of genetic differences on course of the infection. In this study we show that, like mice, different strains of rats exhibit a remarkable variation in the number of brain cysts arising from chronic infection. LEW rats seem to be highly resistant to cyst formation, in contrast to F344 rats that are susceptible. In addition, F344 rats express high numbers of γδ T cells during the acute phase of infection, whereas LEW rats express elevated but comparably low numbers of γδ T cells. The RT1 (rat MHC) haplotypes of both strains are identical in the RT1A and RT1B/D regions, which encode the restriction elements for conventional peptide antigens. Consequently, rat strain-specific differences may be useful to define MHC-independent mechanisms of resistance against T. gondii, which may also act in humans.
